Analysis of storage proteins in a worldwide collection of pea seeds

AB  - Peas (Pisum sativum L.) are an important source of vegetable protein for both humans and animals. However, pea seed proteins are limited in sulfur-containing amino acids (SCAA) and thus considered to be an insufficient source. The main seed proteins in pea are 7S (vicilins, convicilins) and 11S (legumins) globulins. Because 11S globulins contain more SCAA than 7S globulins, variations in the 7S/11S ratio affects the nutritional quality of pea seeds. Sodium Dodecyl Sulphate Polyacrylamide Gel Electrophoresis (SDS-PAGE) was used to separate seed proteins from 260 pea genotypes from around the world (the EUCLEG collection). Based on the electropherogram, 18 protein bands were densitometrically measured. Protein compositional data are intrinsically multivariate. The protein concentration of each genotype is presented in closed form i.e., sum up to 100%. Therefore, for multivariate data analysis specific statistical methodology was used. All methods used in the study were carried out in classical and robust manner that was resistant to outlier observations. The storage protein composition of the genotypes studied varied greatly; in some cases, only traces of the main components were found. The results suggest that some of the genotypes tested could be useful in various breeding programs aimed at producing agronomically viable plants, yielding high-protein seed with specific composition of storage proteins for specific food applications.

AB  - Rubbery taproot disease (RTD) of sugar beet was observed in Serbia for the first time in the 1960s. The disease was already described in neighboring Bulgaria and Romania at the time but it was associated with abiotic factors. In this study on RTD of sugar beet in its main growing area of Serbia, we provide evidence of the association between 'Candidatus Phytoplasma solani' (stolbur phytoplasma) infection and the occurrence of typical RTD symptomatology. 'Ca. P. solani' was identified by PCR and the sequence analyses of 16S ribosomal RNA, tuf, secY, and stamp genes. In contrast, the causative agent of the syndrome “basses richesses” of sugar beet-namely, 'Ca. Arsenophonus phytopathogenicus'-was not detected. Sequence analysis of the stolbur strain's tuf gene confirmed a previously reported and a new, distinct tuf stolbur genotype (named 'tuf d') that is prevalent in sugar beet. The sequence signatures of the tuf gene as well as the one of stamp both correlate with the epidemiological cycle and reservoir plant host. This study provides knowledge that, for the first time, enables the differentiation of stolbur strains associated with RTD of sugar beet from closely related strains, thereby providing necessary information for further epidemiological work seeking to identify insect vectors and reservoir plant hosts. The results of this study indicate that there are differences in hybrid susceptibility. Clarifying the etiology of RTD as a long-known and economically important disease is certainly the first step toward disease management in Serbia and neighboring countries.

Effect of different sowing dates on Cercospora beticola infection level

AB  - Cercospora leaf spot caused by the fungus Cercospora beticola is the most significant foliar disease of sugar beet in Serbia. Measures to reduce damage of this disease are based on the use of tolerant varieties, crop rotation and fungicide application. Climate changes and the resistance of Cercospora beticolastrains to fungicides from the strobilurin group further complicate the fight against this disease. In extremely favourable years for the development of disease with high temperatures and precipitation during the summer months, growers in Serbia perform 4-8 fungicide treatments.

AB  - There are three main yellowing viruses that damage the beet crop in Europe: Beet yellows virus (BYV), Beet mild yellowing virus (BMYV) and Beet chlorosis virus (BChV). Although not a true yellowing virus, Beet mosaic virus (BtMV) is found in sugar beet in Europe and can cause damage. The yellowing viruses are transmitted by aphids. The recent ban of neonicotinoids in the EU endangered sugar beet production in terms of crop protection against aphids. Since, Republic of Serbia agenda is to become member state of the EU, in order to do that it is necessary to completely synchronise legislation with the EU in all domains, including agriculture and crop protection.

AB  - Trulež korena poslednjih nekoliko godina nanosi najveće štete u proizvodnji šećerne repe. Na području severne Srbije u 2018. godini zabeleženi su ogromni gubici zbog velikog procenta trule repe koji se kretao i do 50%. Na oglednoj parceli Instituta za ratarstvo i povrtarstvo ispitivan je procenat truleži u zavisnosti od roka setve i gajenih hibrida šećerne repe. Rezultati analize varijanse dvofaktorijalnog ogleda su pokazali da su uticaj hibrida, roka setve i interakcije između hibrida i roka setve imali značajan efekat na pojavu truleži korena šećerne repe. Testom najmanje značajne razlike utvrđene su razlike u pogledu truleži
između pojedinačnih hibrida i rokova setve. Najmanje trule repe je zabeleženo kod hibrida Nansen i Tajfun, dok je najveći prosek trulog korena utvrđen kod hibrida Vandana. Među ispitivanim rokovima setve najveći procenat trule repe zabeležen je u drugom roku. Ostaje pitanje koji su primarni uzročnici pojave truleži, jer dobijeni rezultati ukazuju da je bolest truleži korena posledica, dok nam pravi uzrok slabosti biljaka šećerne repe i prilagođavanja uslovima sredine i dalje ostaje nepoznat.
AB  - During the last few years sugar beet root rot has caused the greatest losses in sugar beet cultivation. A northern part of Serbia, main region of sugar beet production was especially hit by large percentage of infected roots in growing season of 2018. Field plots of IFVCNS were used for determining root rot percentage in sugar beet plants at different hybrids and planting dates. ANOVA results showed that hybrids, planting dates and their interaction had significant effect on the root rot. The least significant differences from the aspect of root rot infection were found between individual hybrids and planting dates. The lowest level of root rot had hybrids Nansen and Tajfun while the highest score of rot was recorded at hybrid Vandana. Second planting date had the highest level root rot. However questions which are the primary reasons for this disease is still open, since the results suggest that fungal infection is only the consequence of the disease, while the real reasons of weak plant response and their adaptation to environmental factors remains unknown.

AB  - Semihibridna strategija oplemenjivanja lucerke se bazira na ukrštanju populacija sa ciljem izbegavanja potrebe za inbred linijama u cilju delimičnog zahvatanja efekata heterozisa. Cilj istraživanja je bio: a) da se proveri agronomski potencijal novostvorenih semihibrida lucerke, b) da se reafirmiše hibridno oplemenjivanje, i c) ukaže na posledice i razlike u metodici primenjene semihibridne strategije. Izbor heterotične grupe zavisi od: genetičke uniformnosti roditelja, primenjenog sistema kosdibe, rejonizacije sorte i načina umnožavanja semena semihibrida. Roditeljske populacije trebaju biti iste grupe dormantnosti, slično tolerantno na abiotski i biotski 
stres u cilju smanjenja genetičkog opterećenja semihibrida. Kako je lucerka autotetraploidna vrsta, oplemenjivanje se radi na populacionom nivou/populacionim hibridima, te U sledećim koracima treba 
eksperimentalno potvrditi da li će se heterotični sfekti pojavljivati u narednim generacijama (4-6) umnožavanja semena semihibrida.

AB  - Drought is the prime abiotic factor that limits sugar beet (Beta vulgaris L.) production in Serbia and other regions where the crop is not usually irrigated. As increased irrigation is not an economically viable solution, the most effective one is development of varieties adapted for successful growth in drought-prone environments. Within the framework of ongoing projects, in Institute of Field and Vegetable Crops Novi Sad (IFVCNS) research was performed with the aim to select drought tolerant sugar beet genotypes, improve production under water deficit conditions, and clarify the physiological processes of drought tolerance in sugar beet. Genotypic diversity for drought-related tolerance indices were assessed in the field trials and the strength of association between them and crop performance was measured. As the drought tolerance is a complex trait, very difficult to evaluate in the field, the study was also conducted through the greenhouse experiments and in vitro screening. The plant material was studied for morphological and physiological parameters of water regime and the expression of genes that are known to respond to osmotic stress.

AB  - Constant progress in plant breeding, the development of new hybrids and varieties, accompanied by climate change requires continuous review of growing technology. The most important part of plant production is fertilization, particularly with nitrogen. Nitrogen is required in great quantities for growing plants and it affects the yield of many crops. However this is very dynamic element and large nitrogen reserves cannot be stored in the soil. Considering the specificity of sugar beet production, where nitrogen, in addition to its impact on root yield, significantly affects the quality of the root, it is necessary to constantly review fertilization protocols. Effects of four levels of nitrogen fertilizers (40, 80, 120 and 160 kg N ha-1) on root yield and quality of commercial sugar beet hybrids were examined in the field trials during the growing season 2017. Nitrogen fertilizers were applied before the sowing. Field trial was organised by Randomised Complete Block Design (RCBD) with four replications.

abstract = "Constant progress in plant breeding, the development of new hybrids and varieties, accompanied by climate change requires continuous review of growing technology. The most important part of plant production is fertilization, particularly with nitrogen. Nitrogen is required in great quantities for growing plants and it affects the yield of many crops. However this is very dynamic element and large nitrogen reserves cannot be stored in the soil. Considering the specificity of sugar beet production, where nitrogen, in addition to its impact on root yield, significantly affects the quality of the root, it is necessary to constantly review fertilization protocols. Effects of four levels of nitrogen fertilizers (40, 80, 120 and 160 kg N ha-1) on root yield and quality of commercial sugar beet hybrids were examined in the field trials during the growing season 2017. Nitrogen fertilizers were applied before the sowing. Field trial was organised by Randomised Complete Block Design (RCBD) with four replications.",

AB  - Šećerna repa je najvažniji usev za proizvodnju šećera u umerenim klimatskim regionima sveta. Oplemenjivanje i selekcija su veoma složeni jer je neophodno ostvariti ravnotežu između brojnih osobina korena. Cilj ovog istraživanja je da se analiziraju korelacije između osobina korena šećerne repe gajenih u različitim uslovima sredine. Tokom dve uzastopne godine (2014. i 2015.) odnosi između osobina (prinos korena, sadržaj šećera, štetni kalijum, štetni natrijum, alfa amino azot, iskorišćenje šećera i prinos kristalnog šećera) su ispitivani u različitim tretmanima mineralne ishrane. Biljni materijal se sastojao iz osam hibrida šećerne repe dok su sredine predstavljene rastućim dozama mineralnih đubriva (N, P, K).
AB  - Sugar beet is the most important sugar crop in the temperate regions of the world. Breeding and selection are very complex since it is necessary to achieve the balance between the numerous sugar beet root characteristics. The aim of this study was to analyse correlations between root traits of sugar beet in different environmental conditions. The relations between traits (root yield, sugar content, potassium, sodium, amino nitrogen, recoverable sugar and white sugar yield) were examined during the two consecutive years (2014 and 2015) in different mineral nutrition treatments. Plant material consisted of eight sugar beet hybrids, while environments were represented as treatments with growing levels of mineral fertilizers (N, P, K).

AB  - Sugar beet is one of the most important industrial crops. In many areas of sugar beet production, the water deficit during the summer significantly limits the root yield and the sugar content. Since breeding for drought tolerance is economically the most viable solution for overcoming this problem, it is necessary to understand the reaction of sugar beet genotypes to the conditions of water deficit. As part of an extensive study of sugar beet reaction to drought stress, this research was conducted with the aim to detect changes in growth parameters, proline content and the expression of candidate genes during the in vitro induced water deficit.

AB  - Sugar beet is one of the most important industrial crops. Beet sugar now accounts for almost all sugar production in the Europe and for about 20% of total world production. Crop improvement is important in order to enhance productivity, sugar content or other desirable traits, and molecular techniques can help to accelerate such efforts. One of our research interests is focused on to the development of new sugar beet lines and hybrids combining marker assisted selection with conventional breeding methods. DNA markers were used to assess the genetic diversity and distance among breeding lines and correlate them with hybrid performance, in order to define the heterotic groups and predict heterosis effect. Our activities also involve the investigation of different reaction of sugar beet genotypes to reduced water supply, as the major limiting environmental factor in sugar beet growing.

AB  - Ascochyta pisi, Dydimella pinodes and Dydimella pinodella are three worldwide-distributed fungal pathogens of pea that occur singly or in combination and are sometimes referred to as the Ascochyta complex. They are among the most prevalent and damaging pathogens of legumes worldwide and their identification is currently based on symptoms and morphology. Since the identification based on these criteria remains difficult and uncertain, the aims of this study were to identify the pea-associated Ascochyta species and estimate their molecular phylogenies, through two main approaches: (i) using sequence data from the ribosomal internal transcribed spacer regions (ITS) and elongation factor 1-alpha (EF); and (ii) using specific PCR-based marker (IGS1). Eighty nine isolates assumed to be A. pisi, D. pinodes and D. pinodella of diverse geographical origins were used.

AB  - Sensitivity of alfalfa to acidity and aluminium (Al) toxicity in soil is the major limiting factor in broadening of its growing area. Due to lack of Al tolerance in primary alfalfa germplasm, there is a need for transfer of genes for Al tolerance from other Medicago germplasm. One of the identified sources of Al tolerance is M. sativa ssp. caerulea accession PI 464724, which was used as a female parent in our study. The objectives of this study were: (i) obtaining the tetraploid offspring from 2x-4x M. sativa ssp. caerulea - M. sativa ssp. sativa spontaneous crosses, and (ii) development of a breeding strategy for Al/acid tolerance in alfalfa, using M. sativa ssp. caerulea as a source of Al tolerance. Out of eleven fully developed plants, five were morphologically similar to M. sativa ssp. caerulae, while six plants were similar to M. sativa ssp. sativa. All tested plants were fertile, with the pollen viability ranging from 21.45 to 97.09% and the average number of ovules per plant from 8.80 to 12.29. Eleven SSR primer pairs confirmed the hybrid nature of M. sativa ssp. caerulae x M. sativa ssp. sativa offspring. Both the Cluster Analysis and the Principal Coordinates Analysis separated plants in the caerulae type from plants in the sativa type, with one exception. Strategies based on conventional and molecular marker breeding efforts could lead towards development of tolerant alfalfa cultivars and successful crop production on acidic, Al-contaminated soils.

abstract = "Sensitivity of alfalfa to acidity and aluminium (Al) toxicity in soil is the major limiting factor in broadening of its growing area. Due to lack of Al tolerance in primary alfalfa germplasm, there is a need for transfer of genes for Al tolerance from other Medicago germplasm. One of the identified sources of Al tolerance is M. sativa ssp. caerulea accession PI 464724, which was used as a female parent in our study. The objectives of this study were: (i) obtaining the tetraploid offspring from 2x-4x M. sativa ssp. caerulea - M. sativa ssp. sativa spontaneous crosses, and (ii) development of a breeding strategy for Al/acid tolerance in alfalfa, using M. sativa ssp. caerulea as a source of Al tolerance. Out of eleven fully developed plants, five were morphologically similar to M. sativa ssp. caerulae, while six plants were similar to M. sativa ssp. sativa. All tested plants were fertile, with the pollen viability ranging from 21.45 to 97.09% and the average number of ovules per plant from 8.80 to 12.29. Eleven SSR primer pairs confirmed the hybrid nature of M. sativa ssp. caerulae x M. sativa ssp. sativa offspring. Both the Cluster Analysis and the Principal Coordinates Analysis separated plants in the caerulae type from plants in the sativa type, with one exception. Strategies based on conventional and molecular marker breeding efforts could lead towards development of tolerant alfalfa cultivars and successful crop production on acidic, Al-contaminated soils.",

Modifying the sowing and harvesting dates are one of the most often used adaptations in sugar beet cultivation. The aim of this study was to assess the interactions between planting date and sugar beet genotypes for different harvest dates with recommendation for duration of vegetation period for specific hybrids in order to achieve the best performance and to evaluate influence of climatic factors on sugar yield. Three-way analysis of variance and AMMI (Additive main effect and multiple interactions) analysis were performed to investigate interaction between main factors. Analysis of variance revealed that genotypes (G), planting date (PD), harvest date (HD) and interaction G x PD significantly affected sugar yield in 2016. In 2017 genotypes, planting date, harvest date and G x PD interaction significantly affected sugar yield on probability level of 1%, while PD x HD interaction had significant effect on probability level of 5%. Results of AMMI analysis enabled discrimination of genotypes with the highest level of stability in certain planting dates. Hybrids with combined yield and sugar content (NZ type) should have the advantage in earlier planting dates compared to of sugar beet hybrids with higher sugar content (Z type). However, in shortened vegetation period Z type hybrids are more stable and with better sugar yield results. Results of our study suggest that delaying the harvest date decreases differences between sugar yields obtained from hybrids sown in different planting dates. Major factors in the study affecting sugar yield were growing degree days, insolation and number of days from planting to harvest.

AB  - The molecular diversity studies of alfalfa (Medicago sativa L.) germplasm could contribute to a more precise selection of parental populations in many breeding programs. Sequence-related amplified polymorphism (SRAP) markers were used to assess the genetic diversity of 110 individual plants from 13 selected alfalfa cultivars, landraces, and natural populations from Tunisia, Australia, Serbia, and Kazakhstan. Ten polymorphic SRAP primer combinations generated 137 alleles with 0.90 polymorphism information content. The percentage of polymorphic bands per genotype ranged from 57.66% to 70.07% with a mean of 64.29% and overall value of 100%. The genotype Sardi 10 had the highest value for the effective number of alleles; Nei's gene diversity and Shannon information index, exhibited the highest variability level (Ne = 1.453, He = 0.259, I = 0.381, respectively), whereas the genotype Nera exhibited the lowest variability level (Ne = 1.359, He = 0.211, I = 0.317, respectively). The AMOVA analysis showed that 68% of the variance was within the genotypes; this was in line with the coefficient of genetic differentiation (Gst = 0.370). The genetic relatedness of alfalfa individuals analyzed by the neighbor-joining dendrogram was consistent with the Bayesian model-based clustering approach. The exceptions were individuals from genotypes Slavija and Nera, which were grouped separately by STRUCTURE analyses. These results provide useful information for the management of alfalfa genetic resources and the rational use of local and foreign alfalfa populations in breeding programs focused on the development of new, high-yielding cultivars more adapted to drought conditions in North Africa.

AB  - Multi-environment trials are performed every year with the aim of evaluating sugar beet varieties in different environmental conditions of Serbia. The objective of this research was to interpret the effects of genotype, environment and genotype x environment interaction on root yield, sugar content and white sugar yield. GGE biplot method was used to estimate stability of seven sugar beet hybrids developed at the Institute of Field and Vegetable Crops, Novi Sad, Serbia. Mixed model of analysis of variance (ANOVA) was used for the analysis of trial results. Factor genotype effect was assumed fixed while effects of year and environment were assumed random. ANOVA showed that the environment had the greatest effect on root and white sugar yield, while year had the strongest effect on sugar content. Variations of all traits were under significant impact of genotype, while the GEI influence was not a significant. Results of GGE biplot analysis suggested that hybrid Sara was the most stable variety for root yield, while Lara had the greatest stability for sugar content and white sugar yield among the tested genotypes. Also, optimal locations were identified for each genotype, which can be useful when recommending sugar beet varieties for certain growing areas of this field crop.
AB  - Multilokacijski ogledi izvode se svake godine sa ciljem procene hibrida šećerne repe u različitim agroekološkim uslovima Srbije. Cilj ovog istraživanja je bio da se utvrdi uticaj genotipa, spoljašnje sredine, kao i interakcije genotipa i spoljašnje sredine na prinos korena, sadržaj šećera i prinos kristalnog šećera. GGE biplot metod je korišćen za procenu stabilnosti sedam hibrida šećerne repe stvorenih u Institutu za ratarstvo i povrtarstvo. Mešoviti model analize varijanse (ANOVA) je korišćen za analizu rezultata ogleda. U pretpostavci analize varijanse efekat genotipa je bio fiksan, dok su efekti godine i sredine bili slučajne promenljive. ANOVA je pokazala da je najveći efekat na prinos korena i prinos kristalnog šećerna imala spoljašnja sredina, dok je godina imala najveći uticaj na sadržaj šećera. Genotip je imao značajan uticaj na varijabilnost svih ispitivanih osobina, dok uticaj interakcije genotip/sredina nije bio značajan. Rezultati analize GGE biplota ukazuju da je Sara bila najstabilniji hibrid za prinos korena, dok su sadržaj šećera i prinos kristalnog šećera bili najstabilniji kod hibrida Lara. Identifikovane su optimalne lokacije za svaki genotip, što može biti od koristi pri davanju preporuka za gajenje hibrida šećerne repe u određenim regionima proizvodnje ovog useva.

Results of GGE biplot analysis suggested that hybrid Sara was the most stable variety for root yield, while Lara had the greatest stability for sugar content and white sugar yield among the tested genotypes. Also, optimal locations were identified for each genotype, which can be useful when recommending sugar beet varieties for certain growing areas of this field crop., Multilokacijski ogledi izvode se svake godine sa ciljem procene hibrida šećerne repe u različitim agroekološkim uslovima Srbije. Cilj ovog istraživanja je bio da se utvrdi uticaj genotipa, spoljašnje sredine, kao i interakcije genotipa i spoljašnje sredine na prinos korena, sadržaj šećera i prinos kristalnog šećera. GGE biplot metod je korišćen za procenu stabilnosti sedam hibrida šećerne repe stvorenih u Institutu za ratarstvo i povrtarstvo. Mešoviti model analize varijanse (ANOVA) je korišćen za analizu rezultata ogleda. U pretpostavci analize varijanse efekat genotipa je bio fiksan, dok su efekti godine i sredine bili slučajne promenljive. ANOVA je pokazala da je najveći efekat na prinos korena i prinos kristalnog šećerna imala spoljašnja sredina, dok je godina imala najveći uticaj na sadržaj šećera. Genotip je imao značajan uticaj na varijabilnost svih ispitivanih osobina, dok uticaj interakcije genotip/sredina nije bio značajan. Rezultati analize GGE biplota ukazuju da je Sara bila najstabilniji hibrid za prinos korena, dok su sadržaj šećera i prinos kristalnog šećera bili najstabilniji kod hibrida Lara. Identifikovane su optimalne lokacije za svaki genotip, što može biti od koristi pri davanju preporuka za gajenje hibrida šećerne repe u određenim regionima proizvodnje ovog useva.",

Eight pollinators originated from three research centers in the United States Department of Agriculture, while four pollinators and cms lines were from the Institute of Field and Vegetable Crops, Novi Sad, Serbia. Results: In total, 129 alleleswere obtained, with a mean of 3.2 alleles per SSR marker. The observed heterozygosity ranged from 0.00 to 0.87 (mean = 0.30). Expected heterozygosity and Shannon's information index were the lowest for marker BQ590934 and the highest for markers SB15s and FDSB502s; the same markers were the most informative, with PIC values of 0.70 and 0.69, respectively. Three private alleles were found in pollinator EL0204; two in pollinator C51; and one in pollinators NS1, FC221, and C93035. Molecular variance showed that 77.34% of the total genetic variation was attributed to intrapopulation variability. Cluster and correspondence analysis grouped sugar beet pollinators according to the breeding centers, with few exceptions, which indicate that certain amount of germplasm was shared, although centers had their own breeding programs. Conclusions: The results indicate that this approach can improve the selection of pollinators as suitable parental components and could further be applied in sugar beet breeding programs.

Relationship between hybrid performance and genetic variation in self-fertile and self-sterile sugar beet pollinators as estimated by SSR markers

AB  - Sugar beet hybrid varieties are produced through the crosses between male sterile lines and the multigerm pollinators. The uniformity of pollinators used for hybrid crosses depends on the presence of self-sterility (S-s) and self-fertility (S-f) genes. The aim of the study was to analyze correlation between hybrid performance and genetic distance or heterozygosity of the sugar beet pollinators. Twelve diploid pollinators classified as self-sterile (S-s) or self-fertile (S-f) and two cytoplasmic male sterile (CMS) lines were crossed in line 9 tester scheme, producing 24 F-1 hybrids. The parents and the hybrids were evaluated for root yield and quality traits, from which F-1 performance, combining abilities, mid-parent and high-parent heterosis were calculated. Parental genetic distance and diversity of the pollinators were estimated by SSR markers and, together with GCA and F-1 performance, correlated with the heterosis effects. The S-f hybrids had better GCA and higher values of root yield, root weight, and root circumference than the S-s hybrids. Heterosis was recorded in more combinations with the S-f than with the S-s pollinators. Parameters of genetic diversity were higher in the S-s (Na = 3.125; Ne = 2.341; He = 0.555) than in the S-f pollinators (Na = 3.000; Ne = 2.188; He = 0.510). Genetic distance between the tested pollinators and the CMS lines was low (0.072-0.224) indicating that the genetic base of the investigated germplasm was narrow. Correlation of the heterosis effects with GD and heterozygosity was detected only for the root yield traits.

AB  - Sugar beet is the most important crop for sugar production in Europe. Wide genetic variability is essential in sugar beet breeding programs. The aim of this study is to evaluate variability for the main root traits and differences between monogerm and multigerm sugar beet genotypes from the breeding collection at the Institute of Field and Vegetable Crops. The following traits were analyzed: root weight (g), dry matter content (%), root head weight (g), root/head ratio (%), number of cambial rings, root length (cm) and root diameter (cm). Mean values for two years per genotype were standardized and used for analysis. Principal Component Analysis (PCA) and Cluster Analysis (CA) were used to examine the level of diversity for 20 genotypes and to rank the contributions of the variables. According to CA genotypes could be placed into five main groups, where a large number of multigerm genotypes were put in one group. On average multigerm genotypes were characterized by higher mean values for root weight, length, diameter and lower root head ratio. Multigerm genotypes had higher coefficients of variation for nearly all measured root traits.

AB  - Alfalfa (Medicago sativa L.) is the most important forage crop in the world, regarding growing area and economic value, because of its high yield level, good persistence, high protein content and energy value. In alfalfa, concept of intra-population breeding means direct selection for yield, through recurrent phenotypic selection using some form of progeny testing. Significant differences between genotypes were found for green forage and dry matter yields. The observed variation for both yield traits was much higher at population level than for the selected plants that will form new improved population. In the next steps of breeding process selected plants will be intercrossed in isolation to develop a new, improved population. Alfalfa breeding programs should focus on yield per se, with the use of old and new knowledge on the genetic control of yield and with adaptation of existing or developing new breeding procedure.
AB  - Lucerka (Medicago sativa L.) je najvažnija krmna biljka u svetu, kako sa stanovišta površina koje zauzima tako i njene ekonomske vrednosti predstavljene kroz visok prinos, trajnost (dužinu života), odličan kvalitet i energetsku vrednost. Koncept intrapopulacionog oplemenjivanja kod lucerke podrazumeva direktnu selekciju na prinos primenom rekurentne fenotipske selekcije uz korišćenje testova potomstava. Značajne razlike između genotipova su zabeležene za prinos zelene krme i suve materije. Variranje osobina prinosa je bilo značajno veće na nivou cele populacije nego kod odabranih biljaka. Budući koraci u oplemenjivanju lucerke su sledeći: odabrane biljke će se umnožavati u izolaciji sa ciljem stvaranja nove, popravljene populacije. Novostvorena populacija će se testirati sa nekim od progenih testova i porediti sa početnom populacijom (sorta Nijagara) i komercijalnim sortama lucerke. Nova populacija biće testirana DNK markerima u cilju provere homogenosti, tj. u cilju provere genetičke ujednačenosti. Programi oplemenjivanja lucerke trebalo bi da se fokusiraju na prinos per se, uz korišćenje starih i novih znanja vezanih za genetičku kontrolu agronomski važnih osobina, uz adaptaciju postojećih i razvoju novih oplemenjivačkih procedura.

AB  - The genus vetch (Vicia L.) includes over 200 herbaceous annual and perennial species, containing food and forage legumes among them, with a prevailing Euro-Asiatic distribution. There are only few studies published considering the assessment of genetic diversity in Vicia using DNA markers, except for faba bean, where these were used extensively. The development of novel markers requires high costs and is time consuming. Instead of developing new markers, their transfer within genus is an alternative in less studied species. Having that in mind, the goal of This research was to investigate the possibility of transfer of SSR and ISSR markers within Vicia genus. Cross-species amplifications of the 6 SSR and 4 ISSR primers were carried out, using genomic DNA isolated from three accessions of eight Vicia species. Average transferability for SSR markers was 56.25%. Primers VfG14 and GAI1 displayed the highest, while Vig24 had the lowest rate of transfer.
