AB  - ‘Candidatus Phytoplasma solani’ (stolbur phytoplasma) is associated with rubbery taproot disease (RTD) of sugar beet (Beta vulgaris L.), while Macrophomina phaseolina is considered the most important root rot pathogen of this plant in Serbia. The high prevalence of M. phaseolina root rot reported on sugar beet in Serbia, unmatched elsewhere in the world, coupled with the notorious tendency of RTD-affected sugar beet to rot, has prompted research into the relationship between the two diseases. This study investigates the correlation between the occurrence of sugar beet RTD and the presence of root rot fungal pathogens in a semi-field ‘Ca. P. solani’ transmission experiment with the cixiid vector Reptalus quinquecostatus (Dufour), in addition to naturally infected sugar beet in the open field. Our results showed that: (i) Reptalus quinquecostatus transmitted ‘Ca. P. solani’ to sugar beet which induced typical RTD root symptoms; (ii) Macrophomina phaseolina root rot was exclusively present in ‘Ca. P. solani’-infected sugar beet in both the semi-field experiment and naturally infected sugar beet; and that (iii) even under environmental conditions favorable to the pathogen, M. phaseolina did not infect sugar beet, unless the plants had been previously infected with phytoplasma.

Plants under Siege: Investigating the Relevance of ‘Ca. P. solani’ Cixiid Vectors through a Multi-Test Study

AB  - Crop losses caused by the plant pathogenic bacterium ‘Candidatus Phytoplasma solani’ (CaPsol) underscore the need to better understand its perplexing epidemiological pathways. Hyalesthes obsoletus (Hemiptera, Cixiidae) is a prominent CaPsol vector with three plant associations in Serbia (ex Urtica dioica/HobsUd; ex Convolvulus arvensis/HobsCa; ex Crepis foetida/HobsCf). Another cixiid planthopper, Reptalus quinquecostatus (Dufour), has been recently confirmed as a noteworthy CaPsol vector. A multi-test study assessed the relevance of H. obsoletus associations and R. quinquecostatus populations from Crataegus monogyna and Prunus spinosa in CaPsol occurrence in sugar beet, maize, and tobacco. Molecular typing of the CaPsol strains transmitted to test plants in experimental trials provided the first evidence of HobsUd transmitting CaPsol tuf-a type to sugar beet, HobsCa infecting maize and tobacco with tuf-b type, and HobsCf transmitting CaPsol tuf-b to maize. Affiliation of R. quinquecostatus with the specific CaPsol genotype, dSTOLg, was reaffirmed in this study. The possible involvement of R. quinquecostatus in maize redness disease and tobacco stolbur was suggested, given that this cixiid was identified as a vector of CaPsol to these crops. The obtained results indicate that the tested vectors pose a threat to cultivated plants in Serbia, underscoring the need to recognize their relevance in CaPsol disease occurrences.

AB  - Two phloem-limited pathogens, ‘Candidatus Arsenophonus phytopathogenicus’ and ‘Candidatus Phytoplasma solani’, threaten sugar beet production in France, Switzerland, and Germany. Previous studies of these pathogens in Germany had focused on its western and southern regions, leaving a knowledge gap about eastern Germany. Despite their importance, this study is the first to investigate phytoplasmas in sugar beet in Saxony-Anhalt, Germany. A phytoplasma strain related to ‘Ca. P. solani’ is found predominant in Saxony-Anhalt, unlike in France, where ‘Ca. P. solani’ has a minor role compared with ‘Ca. A. phytopathogenicus’. The phytoplasma strain infecting sugar beet in Saxony-Anhalt was classified into a new subgroup designated as 16SrXII-P. The multilocus sequence analysis (MLSA) of nonribosomal genes of the novel phytoplasma strain showed that it is significantly different from the reference and all previously reported ‘Ca. P. solani’ strains including the strain from western Germany. Analyses of sugar beet samples from previous years confirmed the presence of the 16SrXII-P strain in sugar beet as early as 2020 and also in Bavaria in southern Germany. Based on 16S rDNA analysis, ‘Ca. A. phytopathogenicus’ in Saxony-Anhalt is identical to strains in sugar beet in other parts of Germany and France, as well as to a strain in potato from Germany. The presence and prevalence of two phytoplasmas in sugar beet in Germany suggest that more attention should be directed toward understanding phytoplasma infection in sugar beet in this country.

AB  - Waitea circinata var. zeae (anamorf Rhizoctonia zeae) je značajan patogen uglavnom onokotiledonih biljaka, koji je prisutan u mnogim delovima sveta, a održava se u zemljištu duži niz godina. Poslednjih godina krug domaćina W. circinata var. zeae širi se i na neke dikotiledone biljke. U okviru nedavnih istraživanja u Srbiji W. circinata var. zeae je utvrđen kao prouzrokovač truleži korena i korenovog vrata, i propadanja biljaka kupusa i uljane repice na lokalitetima Futog i Rimski Šančevi. U radu su sistematizovani publikovani podaci o rasprostranjenosti, krugu domaćina, načinu održavanja i širenja, kao i suzbijanju W. circinata var. zeae, kao i rezultati istraživanja koja su obavljena u Srbiji.

Pathogenic potential of Waitea circinata var. zeae

AB  - Waitea circinata Warcup & P.H.B. Talbot (multinuclear Rhizoctonia spp.) is a widespread, soil-borne plant pathogen causing root rot mainly of monocotyledonous plants. Among five described varieties, the most common and widespread is W. circinata var. zeae Toda, Mushika, T. Hayak., Tanaka, Tani & Hyakum. Besides monocotyledonous hosts, a natural infection of a limited number of dicotyledonous plants, including soybeans, beans, peas, sugar beets and carrots were recorded recently. During 2017, cabbage and oilseed rape plants showing symptoms of root and root neck rot, sampled at the localities of Futog and Rimski Šančevi (Serbia), were proved to be infected with W. circinata var. zeae. This was the first record of natural infection of plants from Brassicaceae family worldwide. Two selected isolates from cabbage (299-17) and oilseed rape (300-17) formed fast-growing orange colonies with red sclerotia and multinucleate hyphae with a characteristic branching pattern. The identification was further confirmed by BLAST analysis of sequences of ITS rDNA, RPB2, LSU and β-tubulin genomic regions. Koch’s postulates were fulfilled after inoculation of cabbage and oilseed rape seedlings, while an experimental host range was determined by inoculations of 14 different plant species from 8 families.

During 2017, cabbage and oilseed rape plants showing symptoms of root and root neck rot, sampled at the localities of Futog and Rimski Šančevi (Serbia), were proved to be infected with W. circinata var. zeae. This was the first record of natural infection of plants from Brassicaceae family worldwide. Two selected isolates from cabbage (299-17) and oilseed rape (300-17) formed fast-growing orange colonies with red sclerotia and multinucleate hyphae with a characteristic branching pattern. The identification was further confirmed by BLAST analysis of sequences of ITS rDNA, RPB2, LSU and β-tubulin genomic regions. Koch’s postulates were fulfilled after inoculation of cabbage and oilseed rape seedlings, while an experimental host range was determined by inoculations of 14 different plant species from 8 families.",

AB  - Rubbery taproot disease (RTD) of sugar beet was observed in Serbia for the first time in the 1960s. The disease was already described in neighboring Bulgaria and Romania at the time but it was associated with abiotic factors. In this study on RTD of sugar beet in its main growing area of Serbia, we provide evidence of the association between 'Candidatus Phytoplasma solani' (stolbur phytoplasma) infection and the occurrence of typical RTD symptomatology. 'Ca. P. solani' was identified by PCR and the sequence analyses of 16S ribosomal RNA, tuf, secY, and stamp genes. In contrast, the causative agent of the syndrome “basses richesses” of sugar beet-namely, 'Ca. Arsenophonus phytopathogenicus'-was not detected. Sequence analysis of the stolbur strain's tuf gene confirmed a previously reported and a new, distinct tuf stolbur genotype (named 'tuf d') that is prevalent in sugar beet. The sequence signatures of the tuf gene as well as the one of stamp both correlate with the epidemiological cycle and reservoir plant host. This study provides knowledge that, for the first time, enables the differentiation of stolbur strains associated with RTD of sugar beet from closely related strains, thereby providing necessary information for further epidemiological work seeking to identify insect vectors and reservoir plant hosts. The results of this study indicate that there are differences in hybrid susceptibility. Clarifying the etiology of RTD as a long-known and economically important disease is certainly the first step toward disease management in Serbia and neighboring countries.

AB  - Proučavanja fitopatogenih gljiva i pseudogljiva u našoj zemlji imaju dugu tradiciju, a početak se uglavnom povezuje sa osnivanjem Poljoprivredno-šumarskog fakulteta davne 1919. godine. Fitopatogene gljive su danas u svetu široko poznati patogeni koji pričinjavaju oko 70% šteta od svih biljnih bolesti. U Srbiji, fitopatogene gljive i pseudogljive takođe redovno nanose velike štete u biljnoj proizvodnji pa su među proizvođačima prepoznate kao važni uzročnici gubitaka koji se mogu sprečiti ili ublažiti samo uz podršku nauke. Po svemu, istraživanja fitopatogenih gljiva i pseudogljiva danas predstavljaju jednu dobro profilisanu i značajnu oblast primenjenih istaživanja.

Višejedarne anamorfne Rhizoctonia spp. sa teleomorfom opisanim kao Waitea circinata su biljni patogeni koji se dugo održavaju u zemljištu i rasprostranjeni su i vrlo štetni širom sveta. Od pet opisanih varijeteta W. circinata var. zeae je najrasprostranjeniji. Za sada poznat krug domaćina W. circinata var. zeae obuhvata monokotiledone i mali broj dikotiledonih biljaka, među kojima soju, pasulj, grašak, šećernu repu i mrkvu.

AB  - Cabbage, a widely used and popular vegetable, and oilseed rape, the second most valuable oilseed crop in the world, are two important species from the Brassicaceae family. Two geographically separated outbreaks of cabbage and oilseed rape root rot with estimated incidence of 15 and 20%, respectively, were recorded during 2017 in the Vojvodina region, Serbia. Twelve hyphal-tip isolates were obtained from symptomatic cabbage and oilseed rape plants and identified as Waitea circinata var. zeae based on morphological and molecular features. This indicates that W. circinata var. zeae has expanded its host range to the Brassicaceae family. Sequence analyses of internal transcribed spacer (ITS) and large subunit of the ribosomal DNA, RPB2, and B-tubulin genes revealed the highest similarity with multiple W. circinata var. zeae. Neighbor-joining analyses of ITS sequences resulted in a phylogenetic tree with one welldefined branch of W. circinata var. zeae, with two separate groups. All Serbian isolates and the majority of isolates originating from natural infection of dicotyledonous plants grouped together in group I. Following artificial inoculation, W. circinata var. zeae isolates caused mild to medium root necrosis of seedlings of 2 monocotyledonous and 12 dicotyledonous plant species, implying a wider host range than was known for W. circinata var. zeae. Additionally, this is the first occurrence of W. circinata var. zeae on dicotyledonous host plants in Europe. Because cabbage and oilseed rape are important crops grown worldwide, the occurrence of this new soilborne pathogen with a broad host range imposes the necessity for changes in routine disease control practices, particularly crop rotation.

The disease was already described in neighboring Bulgaria and Romania at the time but it was associated with abiotic factors. In this study on RTD of sugar beet in its main growing area of Serbia, we provide evidence of the association between 'Candidatus Phytoplasma solani' (stolbur phytoplasma) infection and the occurrence of typical RTD symptomatology. 'Ca. P. solani' was identified by PCR and the sequence analyses of 16S ribosomal RNA, tuf, secY, and stamp genes. In contrast, the causative agent of the syndrome “basses richesses” of sugar beet-namely, 'Ca. Arsenophonus phytopathogenicus'-was not detected. Sequence analysis of the stolbur strain's tuf gene confirmed a previously reported and a new, distinct tuf stolbur genotype (named 'tuf d') that is prevalent in sugar beet. The sequence signatures of the tuf gene as well as the one of stamp both correlate with the epidemiological cycle and reservoir plant host. This study provides knowledge that, for the first time, enables the differentiation of stolbur strains associated with RTD of sugar beet from closely related strains, thereby providing necessary information for further epidemiological work seeking to identify insect vectors and reservoir plant hosts. The results of this study indicate that there are differences in hybrid susceptibility. Clarifying the etiology of RTD as a long-known and economically important disease is certainly the first step toward disease management in Serbia and neighboring countries.",

Molecular characterization of 'Candidatus Phytoplasma solani' in celery: Case study in Futog

Celery plants with prominent leaf yellowing and chlorosis, sporadically with tissue necrosis, were observed during august 2020 in Futog, situated in Novi Sad suburbia in Vojvodina. Total of 12 sampled celery plants, 8 symptomatic and 4 asymptomatic ones, were analysed for ‘Ca. P. solani’ presence. All symptomatic celery plants were infected with stolbur phytoplasma according to the stol11 gene detection and therefore were further subjected to multigene molecular characterization on three genes: tuf, stamp and vmp1. Combining molecular tools PCR/RFLP and sequencing reviled two ‘Ca. P. solani’ multilocus genotypes in celery: tuf-b/Rqg31/V14 and tuf-b/Rpm35/V14, present in 4 samples each. Obtained results of the strain genotyping are in concordance with previous data on the ‘Ca. P. solani’ diversity on celery, but supplemented with genotyping of the vmp1 gene. Outbreak of stolbur phytoplasma in the assessed locality in Futog is linked to tuf-b epidemiological cycle correlated in Serbia mainly with weed Convolvulus arvensis which was present in the subjected celery plot, and could have been the phytoplasma inoculum source. Visual evaluation of the symptom occurrence suggests on 10-15% of ‘Ca. P. solani’ affected celery plants scattered throughout the plot corresponding to the pathogen dispersal in crop by cixiid planthoper Hyalesthes obsoletus (Hemiptera: Auchenorrhyncha) associated with C. arvensis, main vector of stolbur phytoplasma in Serbia.
AB  - ‘Candidatus Phytoplasma solani’, poznata po trivijalnom nazivu stolbur fitoplazma, biljni je patogen koji inficira brojne kulture u Srbiji. Biljke celera sa izraženim žutilom i hlorozom listova, sporadično i sa nekrozom tkiva, uočene su u avgustu 2020. u Futogu, predgrađu Novog Sada u Vojvodini. Ukupno 12 uzoraka celera, 8 simptomatičnih i 4 asimptomatične biljke, analizirane su na prisustvo ‘Ca. P. solani’. Svi simptomatični uzorci su se pokazali inficiranim stolbur fitoplazmom na osnovu analize stol11 gena i dalje su molekularno okarakterisani na 3 gena: tuf, stamp i vmp1. Kombinacijom molekularnih tehnika PCR/RFLP i sekvenciranja utvrđeno je prisustvo dva ‘Ca. P. solani’ multigenska genotipa, tuf-b/Rqg31/V14 i tuf-b/Rpm35/V14, prisutnih u po četiri uzorka. Dobijeni rezultati genotipizacije uzoraka stolbur fitoplazme odgovaraju prethodnim podacima o diverzitetu ovog patogena na celeru u Srbiji i dopunjeni su sa genotipizacijom vmp1 gena. Pojava stolbur fitoplazme u Futogu je vezana za tuf-b epidemiologiju, koja uglavnom izvire iz korovske vrste Convolvulus arvensis (poponac), koja je bila prisutna u istraživanom zasadu celera i mogla je biti izvor inokuluma. Vizuelna procena pojave simptoma pokazala je 10-15% prisustva simptomatičnih biljaka celera, nepravilno raspoređenih unutar zasada što ukazuje da je odgovorni vektor najverovatnije cikada Hyalesthes obsoletus (Hemiptera: Auchenorrhyncha) asocirana sa poponcem, najznačajniji vektor stolbur fitoplazme u Srbiji.

Vilina kosica: stari problem traži nova rešenja

AB  - Vilina kosica je najrasprostranjenija parazitna cvetnica u Srbiji. Za sada se može tvrditi da su ekonomski najvažnije vrste, prisutne kod nas, Cuscuta campestris Yunck. i Cuscuta epythimum L. Ove dve vrste se po načinu vezivanja za biljku domaćina razlikuju i to utiče na izbor mera njihovog suzbijanja, pa je pre svega, potrebno determinisati koja vrsta je prisutna i tome prilagoditi način suzbijanja. Imajući u vidu značaj viline kosice i štete koje može da nanese, u mnogim zemljama, takođe i kod nas, ona je svrstana u kategoriju karantinskih biljnih parazita (A2 grupa), čije je sprečavanje širenja i suzbijanje obavezno u celoj zemlji. Uprkos tome, zakonska regulativa, kojom se sprečava širenje viline kosice se nedovoljno poštuje, što ovoj parazitnoj cvetnici omogućava veću ekspanziju. Najveću pažnju treba obratiti na useve u polju. Suzbijanje viline kosice treba početi čim se uoče početna žarišta zaraze i završiti ga pre nego što vilina kosica cveta i obrazuje seme. Na jednoj biljci može se obrazovati i do 15.000 semena koje može da sačuva vitalnost u zemljištu i preko 10 godina, što predstavlja glavni mehanizam održavanja ove parazitne cvetnice u prirodi. Prisustvo viline kosice na antropogenim staništima (različiti tipovi useva i ruderalna staništa urbanih i ruralnih područja) pre više od jedne decenije u Srbiji je zabeleženo na 25% ocenjenih površina, a danas su te površine značajno uvećane. Najveće štete kod nas vilina kosica pravi kada se u velikim infestacijama javi na tek zasnovanim višegodišnjim leguminozama (lucerištima, deteliništima), koji ujedno spadaju u najčešće parazitirane useve od strane ove parazitne cvetnice. Vilina kosica je jedan od glavnih uzroka proređivanja lucerišta i može izazvati smanjenje prinosa i do 80%.

AB  - Rezerve semena korovskih biljaka u zemljištu sa jedne strane predstavljaju stalni izvor zakorovljenosti, dok sa druge imaju ulogu u održavanju stabilnosti ekosistema i biodiverziteta. Malobrojna istraživanja na ovu temu su pokazala veliku varijabilnost u rezultatima, u sličnim eksperimentalnim uslovima, ali pri korišćenju različitih metoda. Stoga ciljevi u ovom istraživanja su bili odabir adekvatne i precizne metode za procenu rezerve semena korovskih biljaka i pravljenje pouzdanog modela za prognoziranje dinamike pojave korovskih populacija. Istraživanja su izvedena na oglednom polju Instituta za ratarstvo i povrtarstvo u Novom Sadu (N 45°19', E 19°50') u okviru dugogodišnjeg stacioniranog ogleda „Plodoredi”, koji se smatra jednim od najstarijih eksperimenata ovog tipa na prostoru jugoistočne Evrope. U ogledu se ispitivao uticaj sistema gajenja useva na sastav rezervi semena korovskih biljaka, a potom se upoređivao trоpoljni plodored (ozima pšenica-soja-kukuruz) sa monokulturom ozime pšenice. U obe varijante primenjen je isti sistem đubrenja i to 100 kg/ha N (50 kg/ha u jesen pred osnovnu obradu i 50 kg/ha u proleće u prihrani). Za uzorkovanje zemljišta primenjen je „metod kvadrata”, kao pouzdaniji u odnosu na šahovski tip i uzorkovanje po dijagonali koji se u praksi češće koriste. Procena rezerve semena korovskih biljaka urađena je primenom dva metodološka postupka: fizička ekstrakcija semena i naklijavanje zemljišnih uzoraka.
