Germination and vigor of soybean seed produced under different agro-meteorogical conditions

AB  - International seed trade considers seed germination as the main indicator of the seed quality. Different results of seed germination under laboratory conditions and emergence in the field encouraged the development of the concept of the seed vigor and the term of vigor. Seed viability or vigor presents a set of those traits that affect seed germination and the establishment of strong and healthy seedlings under different environmental conditions. The aim of this work was to determine the effect of locality and production year on germination and vigor of soybean seed. The testing was performed on 6 soybean varieties (Valjevka, Bojana, Balkan, Novosađanka, Vojvođanka i Morava) produced in the towns of Inđija and Vrbas in 2009 and 2010. The seed germination of all samples was determined by using standard laboratory and vigor tests (Cold test and Accelerated aging test). When the standard laboratory test was applied the seed germination of all tested varieties produced in 2009 in Inđija ranged from 78% to 87%, and from 94% to 99% in Vrbas. The germination of seeds produced in 2010 in both localities was higher in all tested varieties in relation to 2009. When the accelerated aging test was applied the germination of seeds produced in Inđija in 2009 was statistically significantly lower (67-85%) in all tested varieties in comparison to seed germination obtained by the standard laboratory test (78-87%). The seed germination obtained by the application of the cold test was statistically significantly lower (25-90%) for the seed produced in Inđija in comparison to the value obtained for the seeds produced in Vrbas (80-97) for both years of production.
AB  - Međunarodna trgovina semena, kao osnovni pokazatelj kvaliteta uzima klijavost semena. Različiti rezultati ispitivanja klijavosti semena u laboratorijskim uslovima i nicanja u polju podstakli su razvoj koncepta životne sposobnosti semena i pojma vigora. Životna sposobnost semena ili vigor predstavlja skup onih osobina semena koje utiču na klijavost semena i formiranje snažnog i zdravog ponika u različitim uslovima spoljašnje sredine. Cilj rada je bio da se utvrdi uticaj lokaliteta i godine proizvodnje na klijavost i vigor semena soje. Ispitivanja su izvršena na 6 sorata soje (Valjevka, Bojana, Balkan, Novosađanka, Vojvođanka i Morava) proizvedenih u Inđiji i Vrbasu u 2009 i 2010. godini. Kod svih uzoraka utvrđena je klijavost semena primenom standardnog laboratorijskog metoda i vigor testova (hladni test i test ubrzanog starenja). Primenom standardnog laboratorijskog metoda klijavost semena, kod svih ispitivanih sorata na lokalitetu Inđija, proizvedenih u 2009. godini, se kretala od 78% do 87%, a na lokalitetu Vrbas od 94% do 99%. Klijavost semena proizvedenog u 2010 godini, na oba lokaliteta je bila veća, kod svih ispitivanih sorti, u odnosu na 2009. godinu. Primenom testa ubrzanog starenja klijavost semena proizvedenog na lokalitetu Inđija u 2009. godini, kod svih ispitivanih sorata je bila statistički značajno niža (67-85%) u odnosu na klijavost semena dobijenu standardnim laboratorijskim metodom (78-87%). Klijavost semena dobijena primenom hladnog testa, u obe godine proizvodnje, na lokalitetu Inđija (25-90%) je bila statistički značajno niža u odnosu na vrednosti dobijen za seme proizvedeno na lokalitetu Vrbas (80-97).

AB  - Changes in antioxidant systems in soybean [Glycine max (L) Merr., Fabaceae] genotypes infected with Sclerotinia sclerotiorum were studied 12, 24, 48 and 72 h after inoculation. Generation of superoxide and hydroxyl radicals was evaluated together with the production of malonyldialdehyde, main end product of lipid peroxidation. Several enzymatic and non-enzymatic parameters were monitored as well, such as the activity of antioxidant enzymes superoxide dismutase and pyrogallol and guaiacol peroxidases, reduced glutathione, soluble proteins and total carotenoids content. Results showed that genotypes expressed oxidative burst as well as different antioxidant systems in response to biotic stress caused by pathogen invasion. It has been confirmed that, although hypersensitive cell death is efficient against biotrophic pathogens, it does not protect soybean plants against infection by the necrotrophic pathogen such as S. sclerotiorum. Still, some genotypes showed distinctive and combined activity of several biochemical parameters which may point to further directions in exploring host-pathogen relations and lead to selection and production of new genotypes with higher levels of tolerance

Advantages of the Lab-on-a-Chip Method in the Determination of the Kunitz Trypsin Inhibitor in Soybean Varieties

AB  - Qualitative and quantitative determination of the Kunitz trypsin inhibitor (KTI) in soybean experimental lines is very important in processes of selecting and breeding of new varieties. The total enzyme activity assay, sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE), and Lab-on-a-Chip (LoaC) method were used for the determination of the presence and quantity of KTI in 15 soybean experimental lines and varieties. From the total trypsin inhibitor enzyme assay, inhibitor activities were registered in all samples, even in a Kunitz variety that was a negative control. The SDS-PAGE method did not detect the presence of the KTI protein band in seven soybean experimental lines and Kunitz variety, while the LoaC method showed the absence of KTI only in the Kunitz variety sample. Results confirmed the superiority of the LoaC method over other two methods in selectivity and sensitivity when KTI determination is concerned. Relationships between the KTI content obtained by the LoaC method and total trypsin inhibitor enzyme activity were established and statistically confirmed.

Isoflavone Composition, Total Polyphenolic Content, and Antioxidant Activity in Soybeans of Different Origin

AB  - Twenty soybean cultivars, originating from the United States, Russia, Serbia, and China, were analyzed for their isoflavone composition, total polyphenolic content, and antioxidant activity. Isoflavones were extracted by aqueous methanol (80%) and analyzed by high-performance liquid chromatography/diode array detection. Precision and linearity of the applied method were within the standard limits of validation. The highest and the lowest total isoflavone contents were 4.59 and 1.45 mg/g of dried soybean weight, respectively. A significant difference (P lt .05) was found in isoflavone concentration among the different cultivars, but it was observed that origin is not a significant factor that could influence isoflavone content in soybeans. Total polyphenolic content varied between 2.13 and 3.45 mg of gallic acid equivalents/g of dried soybean weight. The free radical scavenging activity of soybean extracts assayed by 2,2-diphenyl-1-picrylhydrazyl in terms of 50% inhibitory concentration (IC50) ranged from 1.40 to 3.35 mg/mL. Negative correlation between total polyphenolic content and IC50 was observed, but there was no correlation between total isoflavone content and IC50. On the basis of this study, soybean cultivars with larger potency for biological activity could be recognized.

AB  - In our processing centre there are several storage types for soybean seed storage and it is very important to know which storage type exerts the most significant influence on seed quality, i.e. which storage type is appropriate for seed storage. This matter is most important for soybean because soybean seed is very difficult for storage. Six types of storage of soybean seed prior to processing were investigated (ground storage, storage with concrete cell, storage with metal cell, storage with 'Kongskilde' cell, seed storage in large transport, 'jambo' bags, and control, soybean seed processed directly after harvesting). Obtained results revealed that there were differences in quality of processed soybean seed depending on the storage type. Also, the influence of moisture content of harvested seed on quality of processed seed was observed, and higher influence was noticed in seed with low moisture. .
AB  - S obzirom da u našim doradnim centrima postoji nekoliko tipova skladišta u kojima se čuva seme soje pre dorade, neophodno je poznavati koji tipovi skladišta utiču više ili manje na kvalitet semena, odnosno koja su skladišta povoljna za čuvanje semena. Ovo pitanje je od izuzetnog značaja kod soje jer je poznato da ona spada u grupu biljaka čije se seme veoma teško čuva. Istraživanje je obuhvatilo šest tipova skladištenja semena soje pre dorade (podni magacin, skladište sa betonskim ćelijama, skladište sa metalnim ćelijama, skladište sa 'Kongskilde' ćelijama, seme skladišteno u velike transportne, 'džambo', vreće), kao i kontrolu, odnosno seme soje koje je odmah posle žetve dorađivano. Dobijeni rezultati ukazuju na postojanje razlike u kvalitetu dorađenog semena soje, u zavisnosti od tipa skladištenja pre dorade. Takođe je ispitivan i uticaj sadržaja vlage u semenu soje prilikom žetve na kvalitet dorađenog semena, pri čemu je utvrđen veći uticaj kod semena sa nižom vlažnošću. .

Anti-nutritional factors in some grain legumes

AB  - Grain legumes, such as soybean (Glycine max (L.) Merr.), pea (Pisum sativum L.), faba bean (Vicia faba L.), lupins (Lupinus spp.), common vetch (Vicia sativa L.) and grass pea (Lathyrus sativus L.), represent one of the most quality and least expensive solutions for a long-term demand for plant protein in animal husbandry. One of the limitations to an increased use of grain legumes as feed is the presence of diverse compounds in their grain, commonly referred to as anti-nutritional factors, that both decrease nutritive value of grain legumes and, if taken in larger amounts, cause health problems that may be fatal for both human and the animals. By this reason, breeding programmes of all grain legumes is aimed at decreasing the content of anti-nutritional factors to a safe extent. Breeding of soybean cultivars for reduced amount of antinutritive factors resulted in the development of Kunitz-free soybean cultivars, which are suitable for thermal processing at lower temperatures and during a shorter period of time. This is the way of saving energy and preserving valuable nutritional composition of soybean grain. As for other species, the most significant progress has been made in protein pea, where all modern cultivars have either low or very low content of various antinutritional factors. Among the improvements are also 'zero-tannin' cultivars in faba bean, with a wide utilization in both animal feeding and bread industry, lowtoxin common vetch cultivars, 'sweet' cultivars in lupins and low-ODAP cultivars in grass pea.
AB  - Zrnene mahunarke, poput soje (Glycine max (L.) Merr.), graška (Pisum sativum L.), boba (Vicia faba L.), lupina (Lupinus spp.), obične grahorice (Vicia sativa L.) i sastrice (Lathyrus sativus L.) predstavljaju jedno od najkvalitetnijih i najjevtinijih rešenja za dugoročni nedostatak biljnih proteina u stočarstvu. Jedno od ograničenja povećanju korišćenja zrnenih mahunarki u ishrani domaćih životinja jeste prisustvo različitih sastojaka u zrnu, tzv. antinutritivnih činilaca, koji umanjuju njihovu hranljivu vrednost i, u slučaju da se unesu u većoj količini, mogu da dovedu do teških posledica po ljude i životinje. Iz tog razloga, oplemenjivanje svih zrnenih mahunarki usmereno je i ka snižavanju sadržaja antinutritivnih činilaca na bezopasan nivo. Oplemenjivanje soje na smanjeni sadržaj antinutritivnih činilaca dovelo je do stvaranja Kunitz-free sorti soje, pogodnih za termičku obradu na nižim temperaturama i kraćeg trajanja, što je način uštede energije i očuvanja vrednih hranljivih sastojaka sojinog zrna. Što se tiče drugih zrnenih mahunarki, najveći napredak ostvaren je kod proteinskog graška, kod kojeg sve savremene sorte imaju nizak ili vrlo nizak sadržaj antinutritivnih činilaca. Među dostignućima su i zero-tannin sorte boba, sa širokom upotrebom u stočarstvu i industriji hleba, sorte obične grahorice sa niskim sadržajem toksina, tzv. slatke sorte lupina i sorte sastrice sa niskim sadržajem ODAP.
