AB  - Bacterial diseases present serious problem in agricultural production which is
reflected in reduction of quality and yield of cultivated plants. Long term use of
copper compounds in bacterial diseases control can induce resistance in bacterial
population. Considering the economic importance of bacterial diseases on
cultivated plants the aim of this research was to tested different copper compounds
(copper-hydroxide, copper-oxychloride, copper (II) sulphate, copper (III) sulphate)
in different concentrations (100, 200, 300, 350 ppm) against one of the most
distributed plant pathogenic bacteria (37 strains) in vitro conditions. Obtained
results suggested that bacterial strains showed different sensitivity to tested copper
compounds and concentrations. The lowest sensitivity was observed in strains of
Pseudomonas syringae pv. morsprunorum race 1 (KBNS75; M25) and Erwinia
amylovora (JČ5, DHM1) in copper-hydroxide and copper-oxychloride. Strains of
Xanthomonas axonopodis pv. phaseoli (Xap1-5) and X. campestris pv. campestris
(UR-1) showed the highest sensitivity to copper compounds compared to other
tested strains.

Foliar pathogens of sweet and sour cherry in Serbia

AB  - In recent years, sweet and sour cherry production in Serbia has increased. Under Serbian agroecological conditions, pathogens causing leaf diseases threaten the success of sweet and sour cherry production. In the period 2012-2019, the health status of cherries was monitored in more than 30 locations. Depending on the production system, the following leaf pathogens were identified: Blumeriella jaapii, Wilsonomyces carpophilus, Mycosphaerella cerasella, Phoma prunicola, Podosphaera clandestina and Pseudomonas syringae pv. syringae and morsprunorum race 1. Leaf pathogens caused premature defoliation, which adversely affected bud formation for the next growing season and increased susceptibility to freezing. Therefore, attention should be focused on correct leaf pathogen identification, and proper selection, application and rotation of fungicides.
AB  - Poslednjih godina proizvodnja trešnje i višnje u Srbiji je povećana. U našim agroekološkim uslovima, patogeni lista ugrožavaju proizvodnju ovih perspektivnih voćnih vrsta. U periodu 2012-2019. godine praćeno je zdravstveno stanje trešnje i višnje, na više od 30 lokaliteta. avisno od sistema proizvodnje identifikovani su sledeći patogeni lista: Blumeriella jaapii, Wilsonomyces carpophilus, Mycosphaerella cerasella, Phoma prunicola, Podosphaera clandestina, Pseudomonas siringae pv. syringae i morsprunorum rasa 1. Patogeni lista izazivaju prevremenu defolijaciju, što negativno utiče na formiranje pupoljaka za narednu vegetaciju i povećanu osetljivost na izmrzavanje. Stoga pažnju treba usmeriti na ispravnu identifikaciju patogena, pravilan izbor i rotaciju fungicida različitog mehanizma delovanja, kao i optimalno vreme za njihovu primenu.

Methods for diagnostic of prune dwarf virus

AB  - Plant diseases caused by phytopathogenic viruses represent a group of diseases (virosis) that can cause high economic losses and become a limiting factor in achieving full fertility and a quality yield. The transmitting ways of the viruses are multiple, and the main way to combat these diseases is the production, or use of healthy, virus-free seed and planting material. Since viruses cause the most significant damage on woody plants, fruits and vines, it is important to carry out a plant virus screening test in order to avoid multiple harmful effects in both aspects, the material and in the aspect of the time it takes for these plant species to grow and achieve a full fertility. One of the most common viruses infecting stone fruit species and causing significant economic damages is the Prune dwarf virus (PDV). Given the general presence in the world and high destructive potential of PDV, its diagnostic and identification methods are described in detail in this study.
AB  - Bolesti gajenih biljaka čiji su prouzrokovači fitopatogeni virusi, predstavljaju grupu bolesti (viroze), koje mogu prouzrokovati značajne ekonomske gubitke i postati ograničavajući faktor u postizanju pune rodnosti i dobijanja kvalitetnog prinosa. Načini prenošenja virusa su višestruki, a glavni način borbe protiv ovih oboljenja je proizvodnja, odnosno korišćenje zdravog semena i bezvirusnog sadnog materijala. Budući da virusi najveće štete ispoljavaju na drvenastim biljkama, voću i vinovoj lozi, važno je sprovesti proveru sadnog materijala na prisustvo virusa, kako ne bi došlo do višestrukih štetnih efekata, kako u materijalnom aspektu tako i u aspektu vremena koje je potrebno da ove biljne vrste stasaju i postignu punu rodnost. Jedan od najčešćih virusa koji inficira koštičave voćne vrste i na njima pričinjava značajne ekonomske štete je virus kržljavosti šljive (PDV). Imajući u vidu opšte prisustvo u svetu i visok destruktivni potencijal PDV virusa, metode njegove dijagnostike i identifikacije detaljno su opisane u ovom radu.

Postharvest changes in primary and secondary metabolites of sweet cherry cultivars induced by Monilinia laxa

AB  - The aim of the study was to evaluate in which way nine cultivars of sweet cherry fruit cope with postharvest, artificial inoculation with Monilinia laxa pathogenic fungus through changes in sugars, organic acids and polyphenolic compounds, as well as which phenolics compounds are crucial participants in coping with the disease. Contents of sugars in this study were (g kg(-1) dry weight): glucose 205-439 and 268-443, fructose 175-398.9 and 208.6-365.8 and sucrose 20-47.6 and 19.2-38.6 in healthy and infected fruit, respectively. Organic acids detected in sweet cherry cultivars were citric, malic, quinic, shikimic and fumaric acid and their content varied depending on cultivar and treatment. Only 'Burlat' accumulated tartaric acid after the infection. Of all phenolic acids, which content decreased in infected fruit, it is clear that only the more tolerant genotypes 'Merchant', 'Lionska', and 'Sue' enhanced content of 3-feruloylquinic acid (in healthy up to 93 and in infected fruit 105.3-139.1 mg kg(-1)). The most abundant anthocyanins were cyanidin derivatives (80-266.1 and 10-3700 mg kg(-1) in healthy and infected fruit, respectively). 'Burlat', 'Junska rana', 'Merchant' and 'Summit' dominantly had cyanidin-glucoside, while only infected fruit of 'Priusadebnaja' and 'Asenova rana' accumulated cyanidin-glucoside. 'Priusadebnaja', 'Asenova rana' and 'Lionska' had cyanidin-rutinoside. 'Lionska', 'Sue' and 'Asenova rana' had higher cyanidin and peonidin derivative contents in infected fruit (1.2 to 3-fold higher). Peonidine derivatives (peonidin glucoside and rutinoside), 5-carboxypyrano-cyanidin-rutinoside content mostly decreased after the infection (14.9-97.7 %). Bearing in mind obtained results, 3-feruloylquinic acid, flavonols and anthocyanins could be proposed as crucial participants in coping with the disease.

The objective of this paper was to determine different responses to brown rot among nine sweet cherry genotypes grown under the field conditions of infection in open field. In terms of enzyme and antioxidant activity occurring under infection, the studied genotypes showed significant differences in comparison with healthy fruits. However, at the biochemical level, soluble proteins (SP), superoxid dismutase activity (SOD), guaiacol peroxidase activity (GPX), pyrogallol peroxidase activity (PPX), lipid peroxidation (LP) and antioxidant activity (ABTS and TRC assays) in sweet cherry fruits was significantly affected by genotype, plant disease and interaction of these two factors. Significant differences between studied parameters of healthy and infected fruits were recorded in genotypes Junska rana, Merchant, Sue and III/VAL. The parameters studied can be used as a method of assessment of resistance to brown rot among sweet cherry cultivars.
AB  - Mrka trulež (uzročnik: Monilinia laxa Aderh. i Ruhl.) je jedna od najčešćih i najdestruktivnijih bolesti koštičavih voćaka, naročito trešnje. Cilj ovog rada je bio da se utvrde različiti odgovori na mrku trulež ploda između devet genotipova trešnje, uzgajanih u uslovima prirodne infekcije na otvorenom polju. U pogledu enzimatske i antioksidativne aktivnosti koja se javila u uslovima infekcije, ispitivani genotipovi su pokazali značajne razlike u odnosu na zdrave plodove. Na razlike u biohemijskom pogledu uticali su: genotip, prisustvo biljne bolesti kao i interakcija ova dva faktora. Biohemijski parametri koji su pokazali razlike u uslovima infekcije su: sadržaj rastvorljivih proteina (SP), aktivnost supereksid dismutaze (SOD), aktivnost gvajakol peroksidaze (GPX), aktivnost pirogalol peroksidaze (PPX), lipidna peroksidacija (LP) i antioksidativna aktivnost (ABTS i TRC testovi). Značajne razlike između ispitivanih parametara zdravih i zaraženih plodova zabeležene su u genotipovima Junska rana, Merchant, Sue i III/VAL. Ispitani parametri se mogu koristiti kao metoda procene otpornosti na mrku trulež izmedju genotipova trešnje.

AB  - Increased use of pesticides has resulted in contamination of the environment causing
also many associated long-term effects on human health. Therefore, validated analytical
methods that produce reliable results for the assessment of pesticide residues in fruits
and vegetables are highly needed. The main objective of this study was validation of
the method for the analysis of acetamiprid in tomato and determination of its residues
after the application at recommended rates under controlled conditions. Obtained
results of acetamiprid half-life in tomato are compared with DTso in sweet cherry. For
sample pre-treatment QUEChERS procedure was used. Insecticide determination and
quantification were performed by HPLC with diode-array detection (Agilent 1100 Series)
and Zorbax Eclipse C18 column (50 mm x 4.6 mm internal diameter, 1.8 pm particle size)
This method fulfilled validation criteria described in the European Union guidelines (SANCO
12571/2013), by evaluating the accuracy, precision, linearity, limit of detection (LOD) and
limit of quantification (LOQ), as well as matrix-effect (ME), The accuracy and precision were
satisfactory, showing mean recovery values higher than 80% and precision below 20%,
in all cases. The validated method was applied for the analysis of acetamiprid residues in
real samples. Half-life of acetamiprid in tomato was 4.33 day and it is quite similar to DT,
obtained in the experiment with sweet cherries. On the sixth day after the acetamiprid
application residues in tornato were at MRL level, as well as in sweet cherries (according
to Serbian MRL, 2010), while the PHI was 14 days

Mitigating lipid peroxidation in leaves of Prunus sp. rootstocks using exogenously applied auxins

AB  - The effect of exogenously applied auxins on mechanical injury induced oxidative stress was studied in 6 promising rootstock selections of Prunus cerasus L., P. mahaleb L., and P. fruticosa Pall. Investigated selections were included in low vigorous rootstock breeding programme for sweet and sour cherries. The standard rootstock PHL-A was used as a control, due to successful rooting. Leaves of investigated rootstocks were collected on: 0, 1st, 3rd and 7th day of inserting softwood cuttings into the rooting substrate under a fogging system with 95-99% average relative humidity. Exogenously applied auxin mixture consisted of 0.8% α-naphthylacetic acid (NAA) and 0.5% indolebutyric acid (IBA). Mechanical injuries during rooting period represent one of the factors that induce stress in softwood cuttings. Intensity of LP is used as a valuable biomarker of plant response to various abiotic factors. Differences in intensity of LP between auxin-treated and -untreated cuttings were examined. Almost all of investigated selections had lower LP intensity after auxin application (11.4-47.2%) between 1st and 3rd day. However, the most prominent change was in leaves of PHL-A (49.2%). The best LP-lowering effect were recorded in leaves of P. fruticosa, SV4 selection (56.9%) on 7th day and in P. mahaleb, M4 selection, on 1st and 3rd day (5.9, 5.8%), in comparison to untreated softwood cuttings. As for OV21 selection, LP intensity significantly increased in both treated and untreated cuttings on the 1st, 3rd and 7th day, but auxin-treated cuttings showed lower LP values, except on 7th day, reaching 221.3 nmol MDA equivalents g-1 fresh weight. Intensity of LP during vegetative propagation of selected genotypes could be used as one of the biochemical parameters in further rootstock selection for sweet and sour cherries.

Dissipation rate of acetamiprid in sweet cherries

AB  - Degradation of acetamiprid in sweet cherry samples was evaluated at several intervals from the product application until the end of the pre-harvest interval. An orchard of sweet cherries located at Stepanovićevo village near Novi Sad was used in this study. Acetamiprid was applied according to the manufacturer's recommendation for protecting sweet cherries from their most important pests. Sweet cherry fruit samples were collected at eight intervals: immediately after acetamiprid application and 2, 4, 6, 8, 10, 12 and 14 days after application. The extraction of acetamiprid from sweet cherry samples was performed using a QuEChERS-based method. Determination was carried out using an HPLC-UV diode array detection system (Agilent 1100, United States) with an Agilent Zorbax Eclipse C18 column (50 mm × 4.6 mm internal diameter, 1.8 μm particle size). The method was subjected to a thorough validation procedure. The recovery data were obtained by spiking blank sweet cherry samples at three concentration levels (0.1-0.3 mg/ kg), yielding 85.4% average recovery. Precision values expressed as relative standard deviation (RSD) were below 1.61% for the intraday precision. Acetamiprid showed linear calibrations from 0.05 to 2.5 μg/ml with correlation coefficient (R2) of 0.995%. The limit of detection and limit of quantification were found to be 5 μg/kg and 14 μg/kg, respectively. The validated method was applied in the analysis of acetamiprid in sweet cherry samples. During the study period, the concentration of acetamiprid decreased from 0.529 mg/kg to 0.111 mg/kg. The content of acetamiprid in sweet cherry samples at the end of the pre-harvest interval was below the maximum permissible level specified by the Serbian and EU MRLs.
AB  - U cilju praćenja razgradnje acetamiprida u plodovima trešanja u periodu od primene preparata do isteka karence, izvršen je tretman preparatom na bazi ove aktivne materije u preporučenoj dozi. Ogled je postavljen u zasadu srednje kasne sorte trešnje na lokalitetu Stepanovićevo u okolini Novog Sada. Plodovi su uzorkovani osam puta - odmah nakon primene preparata, 2, 4, 6, 8, 10, 12 i 14 dana. Ekstrakcija acetamiprida iz trešanja izvedena je QuEChERS metodom. Za određivanje acetamiprida korišćena je tečna hromatografija sa DAD detektorom (Agilent 1100, United States) i Agilent Zorbax Eclipse C18 kolonom (unutrašnji prečnik 50 mm x 4.6 mm, veličina čestica 1.8 μm). Kao mobilna faza upotrebljeni su acetonitril i 1.5% rastvor CH3COOH (30/70), sa protokom 1 ml/min, temperaturom kolone 25 oC i injektovanom zapreminom 2,5 μl, dok je kao odgovarajuća talasna dužina usvojena vrednost od 254 nm. Validacija metode je u potpunosti sprovedena u skladu sa zahtevima standarda SANCO/12495/2011 (EU Commission Health and Consumer Protection Directorate- General, 2011). Prosečna vrednost prinosa ekstrakcije acetamiprida iz trešanja proverena na tri nivoa obogaćenja (0.1-0.3 mg/kg) iznosila je 85.4%. Preciznost merenja razmotrena proverom ponovljivosti određivanja acetamiprida izražena je relativnom standardnom devijacijom (RSD) sa vrednošću manjom od 1.61%. U opsegu masenih koncentracija acetamiprida od 0,05 do 2,5 μg/ml postignuta je dobra linearnost odziva detektora sa koeficijentom varijacije od 0,995%. Limit detekcije i kvantifikacije za određivanje acetamiprida u trešnjama prikazanom metodom iznose 5 μg/kg i 14 μg/kg. An orchard of sweet cherries located at Stepanovićevo village near Novi Sad was used in this study. Acetamiprid was applied according to the manufacturer's recommendation for protecting sweet cherries from their most important pests. Sweet cherry fruit samples were collected at eight intervals: immediately after acetamiprid application and 2, 4, 6, 8, 10, 12 and 14 days after application. The extraction of acetamiprid from sweet cherry samples was performed using a QuEChERS-based method. Determination was carried out using an HPLC-UV diode array detection system (Agilent 1100, United States) with an Agilent Zorbax Eclipse C18 column (50 mm × 4.6 mm internal diameter, 1.8 μm particle size). The method was subjected to a thorough validation procedure. The recovery data were obtained by spiking blank sweet cherry samples at three concentration levels (0.1-0.3 mg/ kg), yielding 85.4% average recovery. Precision values expressed as relative standard deviation (RSD) were below 1.61% for the intraday precision. Acetamiprid showed linear calibrations from 0.05 to 2.5 μg/ml with correlation coefficient (R2) of 0.995%. The limit of detection and limit of quantification were found to be 5 μg/kg and 14 μg/kg, respectively. The validated method was applied in the analysis of acetamiprid in sweet cherry samples. During the study period, the concentration of acetamiprid decreased from 0.529 mg/kg to 0.111 mg/kg. The content of acetamiprid in sweet cherry samples at the end of the pre-harvest interval was below the maximum permissible level specified by the Serbian and EU MRLs., U cilju praćenja razgradnje acetamiprida u plodovima trešanja u periodu od primene preparata do isteka karence, izvršen je tretman preparatom na bazi ove aktivne materije u preporučenoj dozi. Ogled je postavljen u zasadu srednje kasne sorte trešnje na lokalitetu Stepanovićevo u okolini Novog Sada. Plodovi su uzorkovani osam puta - odmah nakon primene preparata, 2, 4, 6, 8, 10, 12 i 14 dana. Ekstrakcija acetamiprida iz trešanja izvedena je QuEChERS metodom. Za određivanje acetamiprida korišćena je tečna hromatografija sa DAD detektorom (Agilent 1100, United States) i Agilent Zorbax Eclipse C18 kolonom (unutrašnji prečnik 50 mm x 4.6 mm, veličina čestica 1.8 μm). Kao mobilna faza upotrebljeni su acetonitril i 1.5% rastvor CH3COOH (30/70), sa protokom 1 ml/min, temperaturom kolone 25 oC i injektovanom zapreminom 2,5 μl, dok je kao odgovarajuća talasna dužina usvojena vrednost od 254 nm. Validacija metode je u potpunosti sprovedena u skladu sa zahtevima standarda SANCO/12495/2011 (EU Commission Health and Consumer Protection Directorate- General, 2011). Prosečna vrednost prinosa ekstrakcije acetamiprida iz trešanja proverena na tri nivoa obogaćenja (0.1-0.3 mg/kg) iznosila je 85.4%. Preciznost merenja razmotrena proverom ponovljivosti određivanja acetamiprida izražena je relativnom standardnom devijacijom (RSD) sa vrednošću manjom od 1.61%. U opsegu masenih koncentracija acetamiprida od 0,05 do 2,5 μg/ml postignuta je dobra linearnost odziva detektora sa koeficijentom varijacije od 0,995%. Limit detekcije i kvantifikacije za određivanje acetamiprida u trešnjama prikazanom metodom iznose 5 μg/kg i 14 μg/kg. AB  - Pojava ostataka pesticida u voću i povrću moguća je i pored poštovanja principa dobre poljoprivredne prakse. Identifikacija prisutnih pesticida i određivanje njihovih količina u ovim matriksima zahteva primenu metoda visoke osetljivosti i selektivnosti. Istraživanje je sprovedeno u cilju iznalaženja optimalnih eksperimentalnih uslova za razdvajanje i određivanje acetamiprida i njegovog metabolita, 6-hlornikotinske kiseline (6-HNK) u uzorcima trešanja, korišćenjem tečne hromatografije sa nizom fotoosetljivin dioda (DAD). Uzorci trešanja su korišćeni da bi se odredila selektivnost, linearnost, tačnost i limit kvantifikacije (LOQ). Tačnost metode je kvantifikovana kroz procenu merne neizvesnosti zasnovanu na podacima validacije metode.

AB  - Acetamiprid is a broad-spectrum neonicotinoids insecticide used on a wide range crops, especially fruits. Besides its positive effects, acetamiprid also has been posing various health risks to consumers. Due to the growing use of insecticides from the family of neonicotinoids, their increased presence in the environment is evident. For this reason, the concentration of acetamiprid residues, including its metabolite 6-chloronicotinic acid, in agricultural products should be monitored.
A rapid and simple method for the confirmation, simultaneous analysis and quantification of acetamiprid and 6-chloronicotinic acid as intermediate of acetamiprid decomposition in sweet cherry samples has been developed. This residue analysis method is based on the reversed phase separation on C18 column with gradient elution. Analytes’ determination and quantification were performed by high performance _ liquid chromatography (HPLC) with photodiodes detection and chromatograms were extracted at 230nm. Extraction efficiency experiments demonstrated the ability of this method to extract neonicotinoids from sweet cherry samples. These insecticides were extracted with mixture acetonitril/0.1N ammonium-chloride (8/2, v/v). The extract was filtered through layer of celit and evaporated, the residue dissolved with acetone and then analyzed by liquid chromatography. The standard addition method was used for acetamiprid and 6-chloronicotinic acid determination in order to eliminate the matrix effect. Sweet cherry samples spiked with the concentration levels of 0.5mg/kg and 1mg/kg was used to ensure method accuracy (recovery) and data precision. The repeatability of the retention times and peak areas were checked by injecting the standard mixture of acetamiprid and 6-chloronicotinic acid solution five times. Average recoveries of acetamiprid and 6-chlornicotinic acid from sweet cherry samples were in the range of 95-101% and 73-83%, respectively with the associated relative standard deviations (RSDs) < 5%. Limit of detection (LOD) for the analyzed acetamiprid and 6-chloronicotinic acid was estimated from fortified samples. The limit of quantification (LOQ) was 10 and 30u9/kg for acetamiprid and 6-chloronicotinic acid, respectively. Thus, it can be concluded that the developed HPLC-DAD method represents a useful tool for a sensitive and rapid determination of acetamiprid and 6-chloronicotinic acid. Hence, the method may find further application in the analysis of real sweet cherry samples contaminated with these insecticides at a ppb level.

Basal part of the cuttings and leaves of five rootstocks (Mahaleb 1 and 2, Gisela 5, European ground cherry (EGC) and “Oblačinska” sour cherry) were sampled 0, 2, 4 and 6 days after cutting. Cuttings were treated with 0.5% solutions of three auxins: α-naphthylacetic acid (NAA), indolebutyric acid (IBA) and combination of these two (INCIT K). Results obtained for the LP intensity varied depending on the rootstock, plant organ and auxin applied. The best LP-lowering effect of auxins occurred in all genotypes when treated with NAA. In the leaves, the LP peaked on the 2nd day, while the best LP-lowering effect was recorded in European ground cherry rootstocks on the 4th day, independently of auxin applied. The highest MDA production in leaves was recorded in Mahaleb 2 and European ground cherry rootstocks treated with IBA (60-90% higher than control) two days after the cuttings were made. It has been established that in the most of the rootstocks examined auxins showed lowering effect on LP which points to their positive effect, not only on rooting of softwood cuttings, but on their antioxidant protection system, as well.
AB  - U radu je ispitan nivo oksidativnog stresa izazvanog mehaničkom povredom biljke prilikom proizvodnje reznica, u bazalnom delu i listovima pet podloga za višnju i trešnju (Magriva 1 i 2, Gizela 5, Stepska (EGC) i “Oblačinska” višnja). Istovremeno, reznice su tretirane 0.5% rastvorima tri auksina (α-naftilsirćetna kiselina, NAA, indol- buterna kiselina, IBA i kombinacija ovih hormona, INCIT K), u periodu od 60 min, kako bi se utvrdio efekat auksina na antioksidantni status reznica višnje. Uzorci su uzimani nakon 0, 2, 4 i 6 dana. Kontrolu su činile reznice držane u vodi bez hormona. Intenzitet lipidne peroksidacije (LP) je meren kao produkcija malondialdehida (MDA), spektrofotometrijski na 532 nm, i izražen je u nmol MDA g-1 sveže mase.
Dobijeni rezultati su varirali u zavisnosti od podloge, organa biljke i primenjenih auksina. Utvrđeno je da je intenzitet LP u bazalnom delu bio najveći prvog dana, na- kon ozleđivanja, a da su svi ispitivani auksini snižavali produkciju MDA u narednim danima. Najbolji efekat pokazala je Magriva 1 tretirana sa NAA. U listovima, LP je bila najveća 2. dana, a najbolji efekat na smanjenje pokazali su primenjeni auksini u Stepskoj višnji (4. dana). Najveća produkcija MDA u listovima zabeležena je u Magrivoj 2 i Stepskoj višnji tretiranih sa IBA, 2. dana nakon odsecanja reznica (60-90% više od kontrole). Utvrđeno je da u većini podloga pod dejstvom ispitivanih hormona dolazi do smanjenja intenziteta LP što ukazuje na pozitivan efekat primenjenih auksina, ne samo na ožiljavanje reznica nego i na njihov antioksidantni sistem.

AB  - A rapid and simple method for simultaneous determination of acetamiprid and its metabolite 6-chloronicotinic acid in sweet cherry samples has been developed. This residue analysis method is based on the reversed phase separation on C18 column with gradient elution. Analytes' determination and quantification were performed by high performance liquid chromatography (HPLC) with diode-array detector and chromatograms were extracted at 230 nm. Extraction efficiency experiments demonstrated the ability of this method to extract neonicotinoids from sweet cherry samples. These insecticides were extracted with a mixture of acetonitril/0.1N ammonium-chloride (8/2, v/v). The average recoveries of acetamiprid and 6-chlornicotinic acid from sweet cherry samples were in the range of 95-101% and 73-83%, respectively, with the associated relative standard deviations (RSDs)  lt 5%. Expanded measurement uncertainties for the analyzed compounds were 2.7 and 3.01%. The limit of quantification (LOQ) was 10 μg/kg and 30 μg/kg for acetamiprid and 6-chloronicotinic acid, respectively. Thus, the developed HPLC/DAD method can be considered a useful tool for sensitive and rapid determination of acetamiprid and 6-chloronicotinic acid. Hence, the method may find further application in the analysis of real sweet cherry samples contaminated with these insecticides at a ppb level.
AB  - U radu je predstavljena jednostavna metoda za određivanje acetamiprida i njegovog metabolita, 6-hlornikotinske kiseline, u uzorcima trešanja. Metoda je bazirana na primeni reverzno-faznog razdvajanja na C18 koloni primenom gradijentnog eluiranja. Određivanje i kvantifikacija analita je vršena tečnom hromatografijom (HPLC) sa DAD detektorom, pri čemu je korišćena talasna dužina od 230 nm. Tačnost metode je ocenjena procenom merne nesigurnosti. Ekstrakcija acetamiprida i 6-hlornikotinske kiseline iz uzoraka trešanja je vršena smešom acetonitril/amonijum-hlorid (0,1N) u odnosu 80:20 (v/v). Sva merenja su vršena u tri ponavljanja, pri čemu su dobijeni prinosi određivanja acetamiprida i 6-hlornikotinske kiseline u rasponima 95-101% i 73-83%, respektivno. Relativne standardne devijacije (RSD) merenja su u svim slučajevima bile ispod 5%. Limiti kvantifikacije za acetamiprid i 6-HNK iznosili su 10 i 30 μg/kg, respektivno. Kombinovana merna nesigurnost rezultata analize acetamiprida i njegovog metabolita procenjena je na 1,35, odnosno 1,50%, a proširena na 2,7 i 3,01%, upotrebom faktora pokrivanja (k=2) koji odgovara nivou poverenja od 95%, za normalnu raspodelu. Nakon validacije i procene merne neizvesnosti dobijeni rezultati pokazuju da se razvijena HPLC/DAD metoda može primeniti za određivanje sadržaja acetamiprida i 6-hlornikotinske kiseline u uzorcima trešanja i relevantnim matriksima kontaminiranim ovim jedinjenjima.

AB  - The programmes for pesticide residues monitoring in food which are continually carried out in the world ensure that the consumers are not exposed to pesticide residues as well as that only the registered compounds are used in the production. All the member countries of the EU are obliged to observe the official control of pesticide residues in food, in accordance with the Regulations EC 396/2005. At present, in the Republic of Serbia, the programme for monitoring the pesticide residues in food is being prepared as the results of numerous analyses of random samples of fruit, in the market over the past years justify the introduction of such a programme.
AB  - Programi praćenja ostataka pesticida u hrani koji se kontinuirano sprovode u svetu osiguravaju potrošače da ne budu izloženi nepoželjnom nivou ostataka pesticida, kao i da se u proizvodnji koriste samo registrovana jedinjenja. Sve zemlje članice EU su dužne da sprovode zvaničnu kontrolu prisustva ostataka pesticida u hrani, u saglasnosti sa Regulativom EC 396/2005. U Republici Srbiji je u pripremi monitoring program za ostatke pesticida u hrani, a rezultati analiza većeg broja slučajno izabranih uzoraka voća, na našem tržištu, tokom proteklih godina ukazuju na opravdanost uvođenja ovakvog programa.
