AB  - Background: This study was carried out for an understanding of the antioxidant mechanisms of field pea varieties under osmotic stress conditions caused by a lack of water. The activities of antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APx) and glutathione reductase (GR) were examined and analyzed. The gene expression levels of Cu/Zn SOD, cAPx and GR genes were also examined.
Methods: Osmotic stress was stimulated using PEG 6000 with the osmotic potential of -0.1 MPa in 10 days old plant seedlings. The activity of the antioxidant enzymes was measured in the shoots and roots of pea seedlings. The gene expression levels of genes coding antioxidative enzymes were examined by the reverse transcription polymerase chain reaction (RT-PCR) technique. Arabidopsis 18S rRNA was used as endogenous control.
Result: Osmotic stress changed the activities of antioxidant enzymes in the shoots and roots of pea seedlings. Varieties more tolerant to osmotic stress showed a significant increase in antioxidant activities in shoots and roots, while sensitive varieties showed a significant decrease. The results of the analysis of the expression of genes, coding for antioxidant enzymes, showed that the reaction of the tested cultivars to ROS was the result of increased expression of the tested genes in tolerant cultivars, i.e. decreased expression in sensitive cultivars.

AB  - The effects of physical invigoration seed treatment are manifested in many ways, including morphological and structural aspects, changes in the gene expression and the accumulation of proteins and metabolites. To invigorate seed and to investigate the effects of different invigoration techniques on germination and growth seedling characteristics of sunflower, an experiment was conducted with sunflower hybrid Fantazija at the Laboratory for seed testing, Institute of Field and Vegetable Crops, Novi Sad, Serbia. The process of seed invigoration was performed after seeds were exposed to double stressful condition of high temperature and high humidity (72h at 41°C) (the accelerate aging test) by H2O treatment (the seeds were soaked in water for 24 hours) and KH2PO4  treatment (the seeds were soaked in KH2PO4  solution for 24 hours), followed by washing and drying near to initial weight. Treatments with H2O and KH2PO4  affected the decrease of germination, but they were effective in reducing mean germination time (MGT). After the accelerate aging test, the process of invigoration (both H2O and KH2PO4) influenced a minor increase germination percentage, but MGT was prolonged.

AB  - Seed deterioration is an unavoidable process to which seeds of oilseed plant species such as sunflower are especially sensitive. This study aimed to determine whether different invigoration techniques can improve the attributes of germination in sunflower and alleviate the effects of seed ageing. Both aged and non-aged seeds were subjected to invigoration by pre-soaking with distilled water and with 3% of KH2PO4 for 24 hours at 25°C. Germination performance, seedling growth and SDS-PAGE protein profile were determined. The outcome of invigoration depended on the condition of the seeds: invigoration of seeds with both water and KH2PO4 reduced the percentage of germination in non-aged seeds, while in aged seeds there was an increase in germination compared to the control. The SDS-PAGE seed protein profiles revealed that the low molecular weight proteins produced high-intensity bands and the high molecular weight proteins were in low concentrations. After accelerated ageing followed by H2O treatment, some bands of proteins appeared in the region of 2S albumins and were associated with a higher percentage of germination. Our results point out that invigoration treatments were more effective in low vigour seeds which can provide wide practical benefits.

AB  - Grass pea (Lathyrus sativus L.) is a legume with high protein content and good quantities of essential amino acids. It has high tolerance to drought, high adaptability to extreme conditions, disease resistance and low input requirement for cultivation, which makes it very useful for cultivation by resource-poor farmers. Potential usage of grass pea is limited due to the presence of anti-nutritional factors including protease inhibitors, especially trypsin inhibitors. They have a major impact on nutritional value, by inhibiting pancreatic serine proteases (thus impairing protein digestion), but are also likely to protect legume seed against attack by predators. Although once extensively grown throughout Balkan Peninsula, grass pea was nearly forgotten. In recent years there was an effort to reintroduce and increase the interest of researchers and farmers in this crop. The aim of this study was to establish whether genotype and environment have significant influences on trypsin inhibitor activity (TIA) in grass pea and to determine whether TIA is correlated with seed protein content.

AB  - Legumes represent a valuable source of proteins in human and animal nutrition, as well as of different nutrients. Also, they enable diversification of agroecosystems and can be considered climate smart crops. Therefore, they are gaining importance in both developed and developing countries. The most important, and often limiting factor in legume production is the presence of the bean weevil (Acanthoscelides obtectus). This work tested the suitability of three different legume species, i. e. common bean, grass pea and faba bean (two accessions of each species) with growing interest in human nutrition, for the development and population growth of this pest, with special emphasis on the levels of anti-nutritive compounds – trypsin inhibitors. The suitability was assessed based on the progeny production after each month. The development and progeny production were significantly affected by the legume species, but also by the accessions. The highest number of emerged adults was on common bean, while the lowest regardless on observation period, was on faba bean, indicating its low suitability for weevil development. Correlation analysis detected significant influence of the level of trypsin inhibitors on progeny production.

AB  - Germination and seedling growth of seven genotypes of field pea (Pisum sativum L.) were studied in PEG-6000 solution having osmotic potentials -0.1 and -0.2 MPa. A study was undertaken to evaluate the influence of different osmotic potentials (MPa) on seed germination percentage (GP) and mean germination time (MGT). Results show that the percentage of germination decreased with a decrease in osmotic potential, while mean germination time increased. Variety Javor is much more sensitive than the other varieties at all levels of osmotic stress. By contrast, the lowest sensitivity of germination and MGT was found in varieties Mraz and Trezor. Seed germination tests at -0.1 to -0.2 MPa have the potential to be used as tests in field pea. Osmotic stress exposure and its duration significantly affected the growth of seedlings (shoot and root) and the accumulation of biomass, while its effect was more prominent on the growth of the shoot than on root growth, which was also confirmed by the root tolerance index.
AB  - U radu je proučavano klijanje i porast ponika sedam genotipova stočnog graška (Pisum sativum L.) u rastvoru PEG-6000 sa vrednostima osmotskih potencijala od -0,1 i -0,2 MPa. Ispitivanje je izvršeno kako bi se utvrdio uticaj različitih osmotskih potencijala (MPa) na procenat klijavosti semena (GP) i prosečno vreme klijanja (MGT). Rezultati pokazuju da se procenat klijavosti smanjivao uporedo sa smanjenjem osmotskog potencijala, dok se prosečno vreme klijanja povećavalo. Sorta „Javor” mnogo je osetljivija od ostalih sorti pri svim nivoima osmotskog stresa. Suprotno tome, najmanji uticaj na klijanje i MGT utvrđen je kod sorti „Mraz” i „Trezor”. Određivanje klijavosti semena u rastvorima PEG od -0,1 do -0,2 MPa može se koristiti za testiranje stočnog graška. Izloženost osmotskom stresu i njegovo trajanje značajno su uticali na rast ponika (izdanak i koren) i akumulaciju biomase, pri čemu je njegov uticaj bio izraženiji na rast izdanka nego na rast korena, što je potvrdio i indeks tolerantnosti korena.

laboratory seed testing conditions are often in conflict with field conditions. Validated tests for vigour evaluation in legumes are
the conductivity test, the accelerated ageing test and the tetrazolium test. Also, other types of vigour tests have been used to solve
different problems in the seed sector. The modern approach, the computerised image analysis of legume seeds and sprouts, based
on interactive and traditional methods, is a promising alternative for vigour determination.
AB  - Vitalnost semena i vigor su dva najčešća parametra u vezi sa kvalitetom semena. Veoma je važno imati pouzdane metode i testove za ispitivanje kvaliteta i vigora semena. Standardni test klijavosti se može koristiti za predviđanje nicanja u polju, ali uslovi u kojima se nalazi seme tokom ispitivanja često su u suprotnosti sa uslovima na polju. Validovani testovi za procenu vigora leguminoza su test provodljivosti, test ubrzanog starenja i tetrazolijum test. U proučavanju životne sposobnosti semena leguminoza, zavisno od problema koji bi trebalo rešiti, koriste se i drugi vigor testovi. Savremeni pristup, zasnovan na interaktivnim i tradicionalnim metodama mašinskog učenja, omogućava pouzdanu ocenu semena i ponika leguminoza i predstavlja dobru alternativu za objektivno određivanje vigora.

Occurrence and molecular characterization of alfalfa mosaic virus in eggplant in Serbia

AB  - In the 2018–19 growing season, a total of 51 leaves of eggplant plants grown under field conditions were collected randomly from nine private gardens at four different localities in the Province of Vojvodina. Eggplants with nearly 40% of plants showing bright yellow to white mosaic or mottling of leaves were found throughout the inspected fields (gardens). The collected samples were analyzed for the presence of alfalfa mosaic virus (AMV), cucumber mosaic virus (CMV) and potato virus Y (PVY) using commercial double-antibody sandwich (DAS)-ELISA kits. Serological analysis of eggplant samples revealed the presence of AMV in 80.39% collected samples. None of the analyzed samples was positive for CMV and PVY. The virus was successfully mechanically transmitted to test plants including Nicotiana benthamiana, Chenopodium quinoa, C. amaranticolor, as well as eggplant seedlings, confirming the infectious nature of the disease. The presence of AMV in eggplants was further verified by reverse transcription–polymerase chain reaction (RT-PCR) and sequencing, using the primers CP AMV1 and CP AMV2 that amplify part of the coat protein (CP) gene. The phylogenetic analysis showed that Serbian AMV isolates grouped into a separate well-supported group together with AMV isolates from Italy, Croatia and previously characterized isolates from Serbia. To our knowledge, this is the first report of AMV infection of eggplant in Serbia.
AB  - Tokom 2018. i 2019. godine, prikupljen je 51 uzorak biljaka plavog patlidžana iz devet useva sa četiri različita lokaliteta gajenja ove biljne vrste u Vojvodini. Tokom pregleda, na blizu 40% biljaka plavog patlidžana mogli su se uočiti simptomi u vidu žućkastobeličastih pega raspoređenih po čitavoj površini liske. Prikupljeni uzorci analizirani su na prisustvo virusa mozaika lucerke (alfalfa mosaic virus, AMV), virusa mozaika krastavca (cucumber mosaic virus, CMV) i Y virusa krompira (potato virus Y, PVY) korišćenjem komercijalno dostupnih kitova za DAS-ELISA test. Serološkim analizama utvrđeno je prisustvo AMV u 80,39% prikupljenih uzoraka. Prisustvo CMV i PVY nije dokazano ni u jednom od testiranih uzoraka. Virus je uspešno prenet mehaničkim inokulacijama test biljaka Nicotiana benthamiana, Chenopodium quinoa, C. amaranticolor, kao i na klijance plavog patlidžana, čime je potvrđena infe ktivna priroda oboljenja. Efficiency of hydro- and osmopriming in improving garden pea seed quality and initial plant development under saline stress

AB  - Worldwide, agricultural productivity is exposed to increasing constraints in the form of abiotic
stresses such as salinity which affects plant growth and development causing yields loss.
Previous studies suggest that seed priming improves stand establishment under optimal and
stressful environment, given that seed priming might improve seed germination and initial
development by altering seed vigour. This research aimed to define whether various seed priming
treatments could be a feasible strategy to improve garden pea (Pisum sativum L.) resistance to
abiotic stress, namely to salinity. Four garden pea cultivars obtained from the Department for
Vegetable and Alternative Crops, Institute of Field and Vegetable Crops, Novi Sad, Serbia were
included. Seeds of the tested pea cultivars were primed in distilled water (hydropriming), -0.5
MPa polyethylene glycol PEG 6000 solution, and 0.5% KNO3 solution for 24 hours at 25°C in
dark. Non-primed seeds were taken as control. Seeds were submitted to germination test, using
120 mM NaCl to simulate salt stress. The energy of germination, germination, abnormal
seedlings, shoot and root length, fresh and dry shoot and root biomass, and seedling vigour index

stresses such as salinity which affects plant growth and development causing yields loss.
Previous studies suggest that seed priming improves stand establishment under optimal and
stressful environment, given that seed priming might improve seed germination and initial
development by altering seed vigour. This research aimed to define whether various seed priming
treatments could be a feasible strategy to improve garden pea (Pisum sativum L.) resistance to
abiotic stress, namely to salinity. Four garden pea cultivars obtained from the Department for
Vegetable and Alternative Crops, Institute of Field and Vegetable Crops, Novi Sad, Serbia were
included. Seeds of the tested pea cultivars were primed in distilled water (hydropriming), -0.5
MPa polyethylene glycol PEG 6000 solution, and 0.5% KNO3 solution for 24 hours at 25°C in
dark. Non-primed seeds were taken as control. Seeds were submitted to germination test, using
120 mM NaCl to simulate salt stress. The energy of germination, germination, abnormal
seedlings, shoot and root length, fresh and dry shoot and root biomass, and seedling vigour index
were estimated. The results showed significant differences among garden pea cultivars and seed
priming treatments for the assessed traits.",

AB  - Field peas are mostly used for animal nutrition in the Balkan region and in the countries of the former USSR, while in the countries of Western Europe, Canada, the USA and the Middle East they are mostly used for human nutrition. Over three years (2010-2012), the yield and grain yield components of the most widespread field pea varieties were examined. The lowest number of plants was determined in 'NS Junior' intended for combined use (65 plants/m2), while 'Dukat' and 'Partner' had 88 and 85 plants/m2 on average, respectively. The average stem height was 158 cm for 'NS Junior', and 65 and 70 cm for 'Dukat' and 'Partner', respectively. 'Dukat' (4846 kg/ha) and 'Partner' (4521 kg/ha) had significantly higher grain yields compared with 'NS Junior' (2717 kg/ha). The content of crude protein in the grain was highest in 'NS Junior' (27.6%), while 'Dukat' and 'Partner' had lower contents (23-25%).
AB  - Proteinski grašak se uglavnom koristi za ishranu životinja u našem regionu i u zemljama bivšeg SSSR-a, dok se u zemljama zapadne Evrope, Kanade, SAD-a i Bliskog istoka najviše koristi za ishranu ljudi. Tokom tri godine (2010-2012) ispitivane su komponente prinosa i prinos zrna najrasprostranjenijih sorti proteinskog graška. Najmanji broj biljaka utvrđen je kod sorte NS Junior, namenjene za kombinovanu upotrebu (65 biljaka / m2), dok su sorte Dukat i Partner imale u proseku 88 i 85 biljaka / m2. Pectobacterium and Dickeya species causing potato blackleg and tuber soft rot

AB  - World potato production is affected by plant pathogenic bacteria, which significantly limit crop yield and quality in recent years. Bacteria belonging to Pectobacterium and Dickeya genera, causal agents of blackleg and tuber soft rot of potato, are considered as one of the most important pathogens that affect plant production worldwide. Main source of infection are latently infected seed tubers, which transmitted over long trade distances significantly increase disease incidence. Main identification tools for Pectobacterium and Dickeya species until recently were morphological and biochemical characterization methods, followed by pathogenicity tests. Due to phylogenetic heterogeneity of the soft rot and blackleg causing bacteria, identification and differentiation to species and subspecies level became increasingly difficult. Therefore, development of efficient and reliable identification methods is of high importance. Molecular and serological methods offer accurate, reliable and cost-effective differentiation of high specificity and reproducibility. This work aimed to review Pectobacterium i Dickeya species occurring on potato, with special highlight on available molecular identification techniques.
AB  - Proizvodnja krompira u svetu značajno je ugrožena delovanjem fitopatogenih bakterija, te godišnji gubici prinosa poprimaju sve veće razmere. Među deset ekonomski najznačajnijih fitopatogenih bakterija koje smanjuju prinos i kvalitet poljoprivrednih kultura ističu se vrste rodova Pectobacterium i Dickeya - prouzrokovači crne truleži prizemnog dela stabla "crna noga" i vlažne truleži krtola krompira. Najznačajniji izvor infekcije su zaražene semenske krtole, te bakterije ovim putem lako dospevaju u nova područja. Identifikacija bakterija rodova Pectobacterium i Dickeya se do nedavno zasnivala na proučavanju morfoloških odlika bakterijskih kolonija uz upotpunjavanje testovima patogenosti i biohemijskim analizama. Zbog heterogenosti populacije patogena precizna identifikacija prouzrokovača vlažne truleži je dodatno otežana. S obzirom na potrebe za brzom i pouzdanom identifikacijom, u primenu su uvedene serološke i molekularne metode. Molekularne tehnike identifikacije, zbog svoje visoke specifičnosti, brzine izvođenja, pouzdanosti i ponovljivosti danas zauzimaju vodeće mesto u identifikaciji patogena. U radu su prikazane vrste rodova Pectobacterium i Dickeya koje se javljaju na krompiru, sa posebnim osvrtom na molekularne tehnike identifikacije.

AB  - The seed vigour can be explained and described as the theoretical capacity of the seed which allows it to accomplish its substantial function under different environmental conditions. The key purpose of testing seed vigour as an underlying physiological seed characteristic is to establish more susceptible and precise seed quality parameters than a germination test. In seeds of oilseed species such as hemp seed (Cannabis sativa), the vigour can be a limiting factor due to the low chemical stability of the lipids compared to starch. The aim of this study was to evaluate the vigour of hemp seed and to make the difference between the tested hemp varieties in terms of vigour capacity. In addition to that, the aim was to identify and standardize the methods for assessing seed vigour and storability of hemp seed. The experimental materials include five varieties of hemp (Helena, Bacalmas, Hu×Uso, Monoica×Uso, Monoica) obtained from the Institute of Field and Vegetable Crops.

AB  - Global resources are being invested heavily for research and development involving new genetic modification technologies. Growing genetically modified (GM) crops has opened up a debate on the safety of GM crops and the potential long-term harmful effects on the environment, human health and sustainability of new technologies. Food that contains some of the GM ingredients is progressively present on the world market. National legislations differ in countries around the world, from the complete ban on GM foods, to propaganda and acceptance without marking. In our country, according to current regulations, import raw materials for the food industry are controlled for the presence of genetic modification. In order to protect consumers and food safety, it is necessary to monitor the presence of genetic modification at key points in production, from primary production, through processing to market placement.
AB  - Za istraživanja i razvoj novih tehnologija proizvodnje genetičkih modifikacija se na globalnom nivou ulažu ogromna sredstva. Gajenje genetički modifikovanih (GM) biljaka otvorilo je veliku debatu o bezbednosti GM useva i mogućih dugoročnih štetnih uticaja na životnu sredinu, zdravlje ljudi i održivost novih tehnologija. Hrana koja sadrži neki od sastojaka koji je GM je sve više prisutna na svetskom tržištu. Nacionalna zakonodavstva se razlikuju u zemljama u svetu, od potpune zabrane GM hrane, do propagiranja i prihvatanja bez obeležavanja. U našoj zemlji se, prema važećim propisima, uvozne sirovine za prehrambenu industriju kontrolišu na prisustvo genetičke modifikacije. AB  - The most critical aspect of plant survival and reproduction is seed germination. Considering the specificity of the Cannabis sativa production, obtaining high-quality hemp seed is not an easy task. Germination testing is often the most reliable way of assessing viability, but the selection of appropriate methods is complex. This study aimed to evaluate the effect of different conditions (i.e. various methods of germination testing) on the successful process of hemp seed germination, achieving maximum germination potential. Our results showed that C. sativa seeds germinated well in different methods, such as between filter papers, top of filter papers and sand, but there were significant differences between the applied methods. This means that the hemp seeds may have different requirements for germination than conditions covered by the Rules recommended methods. Further, the results show that hemp seed may have certain residual dormancy that needs to be investigated in detail.
AB  - Najkritičnija faza preživljavanja i razmnožavanja biljaka je klijanje semena. S obzirom na specifičnost proizvodnje industrijske konoplje (Cannabis sativa), dobijanje visokokvalitetnog semena konoplje nije jednostavno. Ispitivanje klijavosti je često najpouzdaniji način procene vitalnosti, ali je odabir odgovarajućih metoda složen. Ovo istraživanje je sprovedeno kako bi se procenio efekat različitih uslova tj. različitih metoda testiranja klijavosti, na proces klijanja semena konoplje uz postizanje maksimalnog potencijala za klijanje. Dobijeni rezultati pokazuju da seme C. sativa dobro klija primenom različitih metoda (između filter papira, na filter papiru, pesak), ali postoje značajne razlike između primenjenih metoda. Ovo praktično znači da seme konoplje može imati zahteve za klijavost različite od onih koje su obuhvaćene preporučenim metodama. Takođe, rezultati pokazuju da kod semena konoplje možda postoji neki vid zaostale dormantnosti što je potrebno dodatno istražiti.

AB  - Salinity affects almost every aspect of the physiology and biochemistry of plants, including the
whole plant and at the cellular level. There is a significant variation in salt tolerance within vetch
species, providing opportunities for improving salt-stress tolerance using genetic resources.
Considering the significance of successful seed germination and seedling emergence for further
plant establishment, plant development and achieving high crop yields, the study of the interaction
between salt stress and germination is of great importance. The aim of this experiment was to
evaluate the seed vigour of vetch species (Vicia sativa, V. pannonica), through vigour tests under
saline conditions. Germination and vigour were assessed through the standard test, accelerated
ageing test and cold test, estimating the germination percentage, root and shoot length, and total fresh and dry matter accumulation, under three levels of salt stress (100 mM, 150 mM,200 mM NaCl). The obtained results of the experiment showed significant differences between the two species in response to salinity stress. Furthermore, a species that has a higher vigour has shown noticeably higher tolerance to stress. After the accelerated ageing test, V pannonica showed significantly lower values in all the tested parameters, at all concentrations of NaCl.

AB  - Genetic markers have been used at the Institute of Field and Vegetable Crops Novi Sad (tFVCNS)
for many years, both for seed quality control and for research purposes. The first laboratory in
Serbia that started applying the electrophoresis of isoenzymes was at the IFVCNS, in 1986. The
distinct advantage of laboratory seed purity control is that the seed can be inspected until the
next sowing at different stages, before harvesting or ripening, during or after seed processing. In
addition to conventional laboratory methods in identifying pathogens, serological and molecular
methods are used nowadays. The main feature of these methods is that they are fast, reliable and
suitable for analysing a large number of samples. Determination of the presence and distribution
of viral and bacterial diseases of paprika (Capsicum annuum L.) were performed using serological
and molecular detection. ln addition to determining the presence and distribution of pepper virus,
identification of the most important and most frequent virus isolates was performed using the
reverse transcription method followed by a polymerase chain reaction (RT-PCR). Differentiation
of Xanthomonas spp., the causative agent of the bacterial pungency. of pepper, was carried
out using restriction fragment length polymorphism (RFLP) analysis of the hrp gene (HrpB
region) and the PCR method, thus confirming the similarity of studied strains with the species
X. euvesicatona. According to the valid legislation in Serbia, commercial cultivation of genetically
modified organism (GMO) crops is not allowed. The laboratories involved in GMO analyses have
been accredited according to ISO I IEC 17025:2006 standard and ISTA standards, and authorised
by the Ministry of Agriculture, Forestry and Water Management for analysis of seeds and product
consignments of plant origin from import. All laboratories use internationally recognised methods
and participate in testing the expertise for GMO analysis in inter-laboratory comparisons. Our
laboratory for seed testing has conducted several studies in order to demonstrate the presence
and quantity of Roundup Ready soybean in crude oil, assess DNA degradation, DNA amplification,
and GMO quantification during tofu production, to report on the validity of labels on imported food
samples from different countries and uptake of GM food derived from soybean into the food chain
in Serbia.

AB  - Izlaganje stresnim uslovima životne sredine, kao što je suša, ima negativan efekat na rast biljaka. Abcisinska kiselina (ABA) se akumulira u uslovima osmotskog stresa i igra važnu ulogu u odgovoru na stres i u toleranciji biljaka. Cilj ovog istraživanja je bio da se odredi koncentracija ABA i ekspresija aldehid oksidaznog enzima (AO) kod 7 sorti stočnog graška u početnim fazama klijanja semena i u stadijumu trećeg para listova. Osmotski stres je stimulisan primenom PEG 6000. Stres suše je izazvan prekidanjem zalivanja. Koncentracija ABA je merena Elisa metodom. U nadzemnom delu ponika utvrđeno je značajno povećanje sadržaja ABA kod jedne grupe sorti (Javor, Partner, Junior) i smanjenje koncentracije ABA kod drugih sorti (Mraz, Dukat, Pionir, Trezor).
AB  - Exposure to to environmental stresses, such as drought, causes adverse effects on the growth of plants. Abscisic acid (ABA) accumulates under osmotic stress conditions and plays an important role in the stress response and tolerance of plants. The aim of this research was to determine the rates of ABA and the expression of Aldehyde oxidase enzyme (AO) among 7 cultivars of field pea at early seedling stage and at stage of third pair of leaves. Osmotic stress was stimulated using PEG 6000. The drought stress was induced by a termination of irrigation. ABA concentration was measured by the Elisa method. In the shoot of seedlings a significant increase in ABA content in one group of cultivars (Javor, Partner, Junior), and a decrease in ABA concentration in other cultivars (Mraz, Dukat, Pionir, Trezor) was confirmed.

Uncertainty of Trypsin Inhibitor Activity Measurement of Legume Crops Using Microtiter Plate Method

AB  - Trypsin inhibitors could limit utilization of legumes in human nutrition, but they could also have beneficial health effects. The objective of this study was to measure trypsin inhibitor activity (TIA) of different legumes using microtiter plate method and to identify factors that contribute to uncertainty of TIA measurement. TIA measurements were performed on seeds of faba bean, pea, common vetch, soybean, and common bean cultivars. The significant effect of legume crop on TIA measurement uncertainty was confirmed with P = 0.045. Certain sources of measurement uncertainty were related with the content of trypsin inhibitors (Tis) in legume seeds. In respect to that, significant effect of level of sample dilution (P E, 0.001) was confirmed. Significant influence of the repeated absorbance measurement of sample reaction mixture on uncertainty of TIA measurement was identified (P E, 0.001), and it took 60% of overall TIA measurement uncertainty for soybean cultivars. TIA of soybean cultivars exceeded 90 TUI/mg. Repeated absorbance measurement of positive control reaction mixture took 70% of TIA measurement uncertainty of cultivars with TIA lesser than 4.5 TUI/mg. Graduated cylinder used for preparation of the final sample solutions took the range from 45 to 90% of overall TIA measurement uncertainty of the cultivars whose TIA were in the middle of previously mentioned. The uncertainty of TIA measurement of legume crops was not studied before; thus, this study pointed out that acquiring insight into factors contributing to uncertainty of TIA measurement could give directions for improvement of TIA testing if microtiter plate method is used.

AB  - The aim of this work was to estimate the trypsin inhibitor activity (TIA) in seeds of cultivated pea varieties, wild varieties and selected crosses between varieties from the first group and wild pea varieties and to study the variation in genes coding trypsin inhibitors. Mean TIA in field pea varieties ranged from 3.12 TIU/mg of sample in field pea variety from Czech Republic to 12.90 TIU/mg of sample in field pea variety FP S4 of Serbian origin. Wild field pea varieties showed TIA between 0.98 TIU/mg of sample in Pisum elatius and 9.79 TIU/mg of sample in Pisum abyssinicum. Selected crosses between cultivated field pea varieties and Pisum elatius showed a decrease in TIA in comparison with a parent line that has higher TIA content. The PCR amplification resulted in variety-specific amplification. Varieties with low TIA activity showed amplification with At13/At5 primer pair, while varieties with higher TIA activity showed amplification with primer pairs At12/At5, At14/At5 and At14/At8. Thus, At13/At5 primer pair could be sufficient to distinguish most varieties. These matters can be applied during an early screening of the valuable materials for future breeding programs of pea cultivars with the low level of tripsin inhibitor.

Antioxidant capacity of oilseed rape (Brassica napus) in different soil types

AB  - This work considers the results obtained by testing the parameters of antioxidant status in leaves and roots of oilseed rape at different growth stages (phase I: formation of leaf rosettes and preparing for winter hibernation; phase II: after the winter hibernation; phase III: flowering) grown in semicontrolled field conditions on different soil types. The experiment was conducted on four oilseed rape genotypes (Banacanka, Jasna, Kata, and Zlatna) in semicontrolled conditions in containers with three soil types: chernozem, solonetz, and solonchak. The germination failed on solonchak due to the excessive amount of salt, but results were obtained for plant material on chernozem and solonetz. The following parameters were examined: superoxide dismutase (SOD) enzyme activity, glutathione peroxidase (GPx) activity, lipid peroxidation (LP) intensity, glutathione (GSH) content, and total antioxidant capacity (by ferric reducing antioxidant power, FRAP method). The activity of all the tested parameters of antioxidant status in all genotypes, both in leaves and roots, was higher in plants grown on solonetz soil compared to plants grown on chernozem. Furthermore, the activity of all the tested parameters on both soil types was highest in the second phase of testing, the stage after the winter hibernation, because the plants were exposed to low temperatures in addition to salt stress. The vital thing to note is that GPx activity is important for the defense at the initial stages of growth, while in the later growth stages SOD assumes this role. The genotype Banacanka has been distinguished as the most tolerant, while the genotype Zlatna was the most sensitive.

AB  - This work considers the results obtained by testing the parameters of antioxidant status in leaves and roots of oilseed rape at different growth stages (phase I: formation of leaf rosettes and preparing for winter hibernation; phase II: after the winter hibernation; phase III: flowering) grown in semicontrolled field conditions on different soil types. The experiment was conducted on four oilseed rape genotypes (Banacanka, Jasna, Kata, and Zlatna) in semicontrolled conditions in containers with three soil types: chernozem, solonetz, and solonchak. The germination failed on solonchak due to the excessive amount of salt, but results were obtained for plant material on chernozem and solonetz. The following parameters were examined: superoxide dismutase (SOD) enzyme activity, glutathione peroxidase (GPx) activity, lipid peroxidation (LP) intensity, glutathione (GSH) content, and total antioxidant capacity (by ferric reducing antioxidant power, FRAP method). The activity of all the tested parameters of antioxidant status in all genotypes, both in leaves and roots, was higher in plants grown on solonetz soil compared to plants grown on chernozem. Furthermore, the activity of all the tested parameters on both soil types was highest in the second phase of testing, the stage after the winter hibernation, because the plants were exposed to low temperatures in addition to salt stress. The vital thing to note is that GPx activity is important for the defense at the initial stages of growth, while in the later growth stages SOD assumes this role. The genotype Banacanka has been distinguished as the most tolerant, while the genotype Zlatna was the most sensitive.

AB  - This work was aimed to investigate the effect of salt (NaCl) stress on three oilseed rape (Brassica napus L.) cultivars: 'Banacanka', 'Jasna' and 'Kata'. The following parameters were determined: antioxidant enzymes superoxide dismutase (SOD) and guaiacol peroxidase (GPX), reduced glutathione (GSH) quantity, lipid peroxidation (LP) intensity, radical-scavenging capacity (DPPH-RSC), hydroxyl radical scavenging ((OH)-O-center dot radicals) and ferric reducing antioxidant power (FRAP). Seed viability was determined using the standard laboratory test for germination as well as the seed accelerated aging test measuring the germination, shoot and root length. The plants were subjected to three salt treatments: 0, 100, 150 and 200 mmol dm(-3) NaCl for 7 days. All three cultivars responded with an increase of lipid peroxidation in seedlings and with a decrease of GPX activity. 'Banacanka' was found to be the most vulnerable cultivar, with an increase of 237% of LP intensity in comparison to the control at the highest NaCl concentration. The induction of the SOD activity was observed for all three cultivars by different NaCl concentrations. The antioxidant capacity measured by FRAP, DPPH-RSC and (OH)-O-center dot radicals was also induced, especially FRAP at the highest concentration in the cultivar 'Jasna' (by 282%). Germination, shoot and root length decreased for all investigated cultivars. Our results showed that cultivar 'Jasna' is the most adapted to salt stress because of the highest SOD activity and accumulation of antioxidants. Furthermore, this research suggests that some of the antioxidant parameters can be useful biochemical markers for the selection of oilseed rape tolerant cultivars under the conditions of salinity.

Threshold Level and Traceability of Roundup Ready (R) Soybeans in Tofu Production

AB  - The aim of this study is to assess DNA degradation, DNA amplification, and GMO quantity during tofu production. Soybean seeds were spiked with Roundup Ready (R) soybeans (RRS) at 0.9, 2, 3 and 5 % (by mass), to assess the level of RSS that would be of practical interest for threshold labelling. Real-time polymerase chain reaction (PCR) was more effective than conventional PCR in the analysis of raw soymilk, okara, boiled soymilk and tofu. The negative effect of grinding and mechanical manipulation was obvious in the okara sample prepared with 3 and 5 % RRS, where GMO content was reduced to (2.28 +/- 0.23) and (2.74 +/- 0.26) %, respectively. However, heating at 100 degrees C for 10 min did not cause significant degradation of DNA in all samples. The content of RRS in the final product, tofu, was reduced tenfold during processing, ranging from 0.07 to 0.46 %, which was below the labelling threshold level. The results are discussed in terms of global harmonization of GMO standards, which could have the positive effect on the trade of lightly processed foodstuffs such as tofu, especially regarding the labelling policies.

Identification and phylogenetic analysis of Fusarium sp. FIESC3 the causal agent of seed rot in onion (Allium cepa L.)

AB  - Onion (Allium cepa L.) is one of the most important vegetable crops in Serbia, where it is grown on an approximate surface of 20,000 ha. During the routine quality control analysis of onion seed in 2014, fungal infection was observed in an average of 28% of the seed. The objective of this paper was to isolate, determine, and identify Fusarium sp. based on the pathogen's morphological and molecular characteristics. Onion seed samples were collected from different localities in the region of Vojvodina. To obtain a DNA sequence-based identification, a total DNA of the 25 isolates was extracted directly from the mycelium (100 mg wet weight), with a DNeasy Plant Mini Kit (Qiagen, Hilden, Germany). Following DNA extraction, the translation elongation factor 1-alpha region was amplified by PCR with the primer pair EF1 and EF2. An amplicon of 700 bp was amplified in 25 tested isolates. Identification of one isolate was performed by sequencing the translation elongation factor EF-1α gene, which was deposited in the NCBI GenBank database under accession number KP658211 (Fusarium sp. FIESC3).
AB  - Crni luk (Allium cepa L.) spada u najznačajnije povrtarske biljne vrste u Srbiji. Gaji se na površini od oko 20.000 hektara. Tokom rutinske kontrole kvaliteta semena crnog luka u 2014. godini, primećena je pojava Fusarium sp. u visokom procentu - od 28%. Cilj rada bio je izolacija i identifikacija Fusarium sp. na osnovu morfoloških i molekularnih karakteristika patogena. Uzorci semena luka sakupljeni su s različitih lokaliteta i iz velikog broja skladišta u Vojvodini. Nakon izolacije patogena odabrano je i morfološki okarakterisano 25 izolata Fusarium sp. Izolacija DNK izvršena je direktno iz micelija gljive (100 mg), korišćenjem Dneasy Plant Mini Kit (Qiagen, Hilden, Germany).
