AB  - Isoflavones are a group of phytoestrogens, naturally-occurring substances important for their role in human health. Legumes, particularly soybeans (Glycine max (L.) Merr.), are the richest source of isoflavones in human diet. Since there is not much current data on genetics of isoflavones in soybean, particularly in the aglycone form, elucidation of the mode of inheritance is necessary in order to design an efficient breeding strategy for the development of high-isoflavone soybean genotypes. Based on the isoflavone content in 23 samples of soybeans from four different maturity groups (00, 0, I and II), three crosses were made in order to determine the inheritance pattern and increase the content of total isoflavones and their aglycone form. Genotype with the lowest total isoflavone content (NS-L-146) was crossed with the low-(NS Zenit), medium (NS Maximus), and high-(NS Virtus) isoflavone genotypes. There were no significant differences in the total isoflavone content (TIF) between F-2 populations, and there was no transgression among genotypes within the populations. Each genotype within all three populations had a higher TIF value than the lower parent (NS-L-146), while genotypes with a higher TIF value than the better parent were found only in the NS-L-146 x NS Zenit cross. However, significant differences in the aglycone ratio (ratio of aglycone to glycone form of isoflavones) were found between the populations. The highest aglycone ratio was found in the NS-L-146 x NS Maximus cross. The results indicate that the genetic improvement for the trait is possible.

AB  - The influence of soybean seed coat color and genotype on antioxidant capacity, phenolic content and isoflavone profile was investigated. Isoflavone content and composition of 21 seed samples - yellow, green, brown, black and rarely mentioned ocher and reddish, were determined by high-performance liquid chromatographic analysis. Antioxidant capacity and total phenolic content (TPC) were evaluated spectrophotometrically. Significant correlation between total isoflavone content and total genisteins was established in all colored groups. Total isoflavone content was in the range from 2.11 mg/g in a green wrinkled sample to 5.24 mg/g in yellow seed. It was found that black genotypes had the highest TPC and antioxidant capacity, which were significantly different (p lt 0.05) from other colored soybeans. The obtained interconnections among analyzed isoflavones can be used as a model for estimation of their specific content.

Isoflavone content and antioxidant activity of soybean inoculated with plant-growth promoting rhizobacteria

AB  - Plant-growth promoting rhizobacteria (PGPR) elicit activation of the phenylpropanoid pathway in plants, which leads to phenolics production and enhanced antioxidant capacity. The purpose of this work was to assess the antioxidant activity of soybean plants, Glycine max L., inoculated with PGPR (isolates of Azotobacter chroococcum, Streptomyces sp. and mixture of these) during plant development, as well as the yield of inoculated soybean plants. PGPR applied in the experiment stimulated flavonoids and isoflavone synthesis, which enhanced the non-enzymatic antioxidant ability of the soybean plants. Moreover, PGPRs stimulated the accumulation of daidzein and genistin in soybean seedlings (5-fold and 2-fold compared to the control values, respectively). The mixture of PGPRs had a positive impact on the antioxidant activity (10-20 % higher activity) and yield components of soybean, which proposed this inoculum as possibly a potent bio-fertilizer in soybean production.

Antioxidant potential and total phenolic content of Serbian red clover cultivars

AB  - Red clover (Trifolium pratense) is rich in phytoestrogens as well as other phenolic substances which contribute to its antioxidative properties. Still, its antioxidative potential has not been thoroughly investigated. The aim of this study was to determine radical scavenging capacity and total phenolic content of red clover cultivars from Serbia observing different plant parts. Leaves, stems and flowers from four red clover cultivars (Kolubara, Una, Avala, K17) were grounded and extraction with water, 3M HCl and ethanol was performed. Radical-scavenging capacity was measured by reaction with the stable DPPH (2,2-diphenyl-1-picrylhydrazyl) radical and calculation of the amount necessary to decrease initial DPPH concentration by 50% (IC50). Total phenolic content (TPC) was determined according to the Folin-Ciocalteu method and expressed as gallic acid equivalents (GAE). Antioxidant potential and TPC of leaves and flowers were not statistically different, but on average, flower extracts had the highest antioxidant activity (IC500.087 mg/ml) and leaf extracts the highest TPC (30.30 mg/g GAE).

Alisa and Sava increased their total isoflavone content (33.9% and 6.2% higher than control, respectively) as well as 1511/99, although 48 h after the inoculation its content decreased significantly. Meli exhibited the highest rate of coumestrol biosynthesis (72 h after the inoculation) and PAL activity (48 h after the inoculation). All investigated cultivars are invariably susceptible to this pathogen. Recorded changes could point to possible differences in mechanisms of tolerance among them.

Meli exhibited the highest rate of coumestrol biosynthesis (72 h after the inoculation) and PAL activity (48 h after the inoculation). All investigated cultivars are invariably susceptible to this pathogen. Recorded changes could point to possible differences in mechanisms of tolerance among them.",

AB  - Introduction: Red clover (Trifolium pratense L., Fabaceae) is one of the main sources of phytoestrogens, among which are the most present isoflavones: daidzein, genistein, formononetin and biochanin A. These substances are used for reduction of menopausal symptoms, and they are considered to have certain role in prevention of osteoporosis, cancer and cardiovascular diseases. Aim: The aim of this study was to determine content of isoflavones in different red clover cultivars, cultivated in Vojvodina, and also in their plant parts. Material and methods: Five cultivars were analyzed (stem, leaf and flower). After extraction, isoflavones were identified and quantified in samples by high-performance liquid chromatography (HPLC). Results: Total isoflavone content varied between 0,41 do 3,81 mg/g, and it was on average the highest in leaves (2,73±0,97 mg/g), and the lowest in stems of red clover cultivars (0,47±0,06 mg/g). In leaves formononetin was dominant (on average 1,62±0,97 mg/g), while the lowest average content had biochanin A in stems (0,04±0,06 mg/g). Cultivar m21 had the highest average total isoflavone content in stem, leaf and flower - 1,78±1,77 mg/g. Conclusion: The highest concentration of isoflavones was determined in leaves. Cultivar m21 was the richest in isoflavones, and on average in all analyzed samples formononetin was the most present isoflavone. .
AB  - Uvod: Crvena detelina (Trifolium pratense L., Fabaceae) je jedan od značajnijih izvora fitoestrogena, od kojih su najzastupljeniji izoflavoni: daidzein, genistein, formononetin i biohanin A. Ove supstance se koriste za ublažavanje tegoba u menopauzi, a smatra se da mogu da imaju ulogu u prevenciji osteoporoze, kancera i kardioloških poremećaja. Cilj: Cilj rad je utvrđivanje sadržaja pojedinačnih izoflavona različitih sorti crvene deteline gajenih na području Vojvodine, kao i u pojedinim delovima biljke. Materijal i metode: Analizirano je pet sorti crvene deteline (stabljika, list i cvet). Nakon ekstrakcije, u uzorcima su identifikovani i kvantifikovani izoflavoni visoko-efikasnom tečnom hromatogafijom (HPLC). Rezultati: Sadržaj ukupnih izoflavona varira od 0,41 do 3,81 mg/g, a u proseku najzastupljeniji su u listovima (2,73±0,97 mg/g), dok stabljike crvene deteline sadrže najmanju koncentraciju tih jedinjenja (0,47±0,06 mg/g). AB  - Twenty F1 soybean progenies were analyzed for isoflavone content in order to investigate whether this attribute can be translated from parents to the corresponding hybrids. Content of total isoflavones in analyzed samples was ranged from 1.56 to 3.66 mg/g of dried soybean. Ten analyzed F1 progenies in one group had a mutual parent while the rest in the second group had randomly chosen parents. Contents of total isoflavones. total daidzeins and total genisteins were significantly different (p lt 0.05) in these two sample groups. A significantly strong correlation (p lt 0.05) between total isoflavone content in parents and corresponding F1 progenies was established (R-2 = 0.915). Similarity of the F1 progenies with a mutual parent was confirmed by Principal Component Analysis. Obtained results suggest that isoflavone content is the trait that can be derived from parent genotype to its F1 progenies and therefore the breeding of genotypes with better health promoting characteristics can be directed.
