Genetic diversity and combining abilities for root traits of sugar beet pollinators

AB  - Information about genetic diversity and combining abilities of sugar beet parental components are of a great importance for hybrid creation. The aim of this research was to evaluate genetic diversity among sugar beet pollinators from different breeding programs and their combining abilities for main root traits of sugar beet, root weight, sugar content and sugar yield. As plant material were used eight pollinators originating from three different USDA-ARS breeding programs and four from Institute of field and vegetable crops Novi Sad. The analysis of variance and Duncan's multiple range test revealed significant differences (p = 0.05) among pollinators for all investigated traits. Pollinator CR10 differs from all others in terms of quantitative traits and in terms of combining ability. Despite small root weight CR10 had significantly positive GCA for that trait and showed that genotypes with small root should not be automatically discarded. Pollinators from Institute of field and vegetable crops used in this research had negative GCA for root weight and should be used only as hosts for introduction of new germplasm in future breeding program.
AB  - Informacije o genetskoj divergentnosti i kombinacionim sposobnostima roditeljskih komponenti šećerne repe su od izuzetnog značaja za proizvodnju hibrida. Cilj ovog istraživanja je bio ocena genetske divergentnosti oprašivaca šećerne repe iz različitih oplemenjivačkih programa kao i kombinacionih sposobnosti za glavne osobine korena: masu korena, sadržaj šećera i prinos šećera. Osam oprašivača vodi poreklo iz tri različita oplemenjivačka programa Američkog ministarstva poljoprivrede (USDA-ARS), a četiri iz Instituta za ratarstvo i povrtarstvo, Novi Sad. Analiza varijanse i Dankanov višestruki test intervala su pokazali značajne razlike (p = 0.05) između oprašivača za sva ispitivana svojstva. Oprašivač CR10 se izdvaja od svih ostalih kako u pogledu kvantitativnih vrednosti ispitivanih svojstava tako i u pogledu kombinacionih sposobnosti. Uprkos maloj masi korena CR10 je imao značajno pozitivnu vrednost opštih kombinacionih sposobnosti za ispitivano svojstvo i pokazao da iz oplemenjivackog programa ne treba automatski odbacivati genotipove sa malom masom korena. Svi ispitivani oprašivači Instituta za ratarstvo i povrtarstvo u ovom istraživanju su imali negativne vrednosti opštih kombinacionih sposobnosti za masu korena i trebalo bi ih koristiti kao domaćine za uvođenje nove germplazme.

Effect of azotobacter chroococcum application on production characteristics of sugar beet and microorganisms in sugar beet rhizosphere

AB  - Effects of Azotobacter chroococcum inoculation on sugar beet production characteristics and microbiological status in the soil were studied on three different methods of sugar beet inoculation with Azotobacter chroococcum strains. The variety Drena was used in the study. Experiments were carried out at Rimski Sancevi in 2008, 2009 and 2010. Five strains of Azotobacter chroococcum were used as microbial inoculants. Three inoculation methods were applied: (A) strain incorporation into the soil before planting, (B) strain application on soil surface before the first inter-row cultivation and (C) seed inoculation before planting. All five Azotobacter chroococcum strains, in each of the three application methods, showed a positive effect on the production characteristics of sugar beet. The highest yield increases in root (6.45 t ha(-1)) and sugar (0.63 t ha(-1)) was obtained on seed inoculated with strain 10.

Influence of harvest dates on sugar beet quantitative traits

AB  - Results of the investigation regarding influence of harvest dates on quantitative traits of sugar beet are presented in this paper. The three-year trial was conducted on five varieties developed by Institute of Field and Vegetable Crops, Novi Sad. Analysis of variance showed significant differences between harvest dates in 2008 and 2010 for all analyzed traits, but in 2009, due to severe drought differences were not found. With the delay of harvest date, root and raw sugar yield increased, while sugar content was dependent on climatic conditions (precipitation) between two harvest dates. There were no differences among the analyzed genotypes for measured traits in 2010. Interaction between genotype and harvest date has not been confirmed. Variety Drena had the highest root and raw sugar yields in 2008 and 2010.
AB  - U radu su predstavljeni rezultati trogodišnjeg ispitivanja uticaja tri roka vađenja šećerne repe na kvantitativna svojstva korena pet komercijalno gajenih sorata Instituta za ratarstvo i povrtarstvo u Novom Sadu. Analizom varijanse utvrđene su značajne razlike između rokova vađenja u 2008. i 2010. za sva ispitivana svojstva. Zbog suše u 2009. nisu utvrđene razlike između različitih datuma vađenja. Odlaganjem roka vađenja prinos korena i prinos polarizacionog šećera se linearno povećavao u 2008. i 2010. godini. Sadržaj šećera je varirao u zavisnosti od vremenskih uslova (padavine) između dva različita roka vađenja. U pogledu ispitivanih svojstava između genotipova nije uočena razlika samo u 2010. godini. Nije potvrđeno postojanje interakcije između genotipa i roka vađenja za ispitivana svojstva. Development and application of qRT-PCR for sugar beet gene expression analysis in response to in vitro induced water deficit

AB  - Sugar beet is a significant industrial crop, often grown in the areas where summer drought can severely limit root yield and sugar content. In order to improve development of sugar beet cultivars with increased drought tolerance it is necessary to understand plant response to water stress at the genomic level. Since recent research efforts have focused on the molecular response of the plant in order to identify water deficit inducible genes, the aim of this investigation was to develop qRT-PCR methodology for the quantification of gene expression in sugar beet under conditions of water deficiency in vitro. Sugar beet genotypes, selected for different response to water deficit, were grown and multiplied in vitro. Axilary shoots were placed on micropropagation media with 0%, 3% and 5% PEG, for 28 days. To determine reaction of sugar beet genotypes to in vitro induced water deficit changes in number of axillary shoots, shoot fresh weight and dry matter content were measured. Total RNA was extracted from leaves and reverse transcribed into cDNA, which served as matrix in real-time PCR reaction using TaqMan technology. The housekeeping gene for glutamine synthetase was used as endogenous control, while the genes for alpha amylase and osmotin-like protein were target genes. The relative quantification values for each target gene were calculated by the 2(-Delta Delta Ct) method. Selected candidate genes differed in relative gene expression among genotypes and applied PEG treatments. The obtained results indicated that qRT-PCR protocol was efficient and accurate, showing the potential to be used in further expression analysis of candidate genes involved in sugar beet reaction to water stress.

Estimation of genetic variation among related sugar beet genotypes by using RAPD

AB  - In marker assisted breeding programs, determination of genome polymorphism and development of suitable molecular markers is of the greatest importance. The aim of this research was development of RAPD markers, which will enable quick and cost efficient DNA polymorphism analysis among closely related sugar beet genotypes. The research was conducted on twelve sugar beet genotypes from population of closely related genotypes. Reactions with eight RAPD primers and five primer mixtures resulted in stable and reproducible bands in all samples, with 44 polymorphic and 14 monomorphic loci, and average of 6.13 bands per primer. In two-primer reactions nine new polymorphic bands were detected. Polymorphism information content (PIC) for each primer was calculated, while genetic variation was estimated by calculation of the number of polymorphic loci and their percentage, observed number of alleles, effective number of alleles, and Nei's gene diversity. An unweighted pair group arithmetic mean method (UPGMA) cluster analysis showed that samples were divided in two groups with relatively high coefficient of similarity. The presented results showed that RAPD markers can be suitable for genetic diversity analysis in breeding material with high levels of homology and homozygosity.
AB  - U programima oplemenjivanja uz pomoć molekularnih markera, određivanje polimorfizma genoma i razvoj odgovarajućih markerskih sistema je od najveće važnosti. Cilj ovog rada je bio razvoj RAPD marker sitema sa ciljem njegove primene u brzoj i jeftinog analizi DNK polimorfizma u populacijama srodnih genotipova šećerne repe. Predstavljena istraživanja su sprovedena na dvanaest srodnih genotipova šećerne repe. Kao rezultat reakcija sa osam RAPD prajmera i pet mešavina prajmera dobijeni su stabilni i ponovljivi produkti reakcija kod svih genotipova, sa 44 polimorfna i 14 monomorfnih lokusa, odnosno prosečno 6.13 traka po prajmeru. U reakcijama sa dva prajmera detektovano je devet novih polimorfnih traka. Informativnost prajmera i prajmerskih mešavina je određena pomoću PIC (polymorphism information content) vrednosti, dok je genetska varijabilnosti procenjena određivanjem broja polimorfnih lokusa i njihovim procentom, uočenim brojem alela, efektivnim brojem alela, i genetskog diverziteta po Nei -u. UPGMA (unweighted pair group arithmetic mean method) klaster analiza je pokazala da su uzorci bili grupisani u dve glaven podgrupe sa značajnim koeficijentom sličnosti unutar njih. Predstavljeni rezultati pokazuju da RAPD markeri mogu omogućiti analizu genetske divergentnosti u oplemenjivakom materijalu kod koga se očekuje povećana homologija i stepen homozigotnosti.

Correlations of root traits in monogerm sugar beet from open pollination and their variability

AB  - The aim of this study was to analyze six open-pollinated monogerm sugar beet genotypes for the important root traits, to examine their hybrids with one (CMS) tester and to estimate the correlation between the traits. Root weight, dry matter content, dry matter yield per root, circumference of root and height of the root head were analyzed. All of the tested hybrids showed better results for root weight and dry matter yield per root than their parents. It was not possible to predict superior hybrid combination based solely on the characteristics of parents. Parents with the lowest root weight per se produced the test hybrid with the highest root weight. Parents showed a positive correlation between root weight and dry matter content and this could help sugar beet breeders to choose the most suitable selection criteria.
AB  - Cilj ovog rada je bilo ispitivanje šest novih jednokličnih genotipova šećerne repe poreklom iz slobodne oplodnje, kao i njihovi hibridi za najvažnija korenska svojstva. Određeni su koeficijenti korelacije između svojstava. Nakon završene vegetacije analizirani su: masa korena, sadržaj suve materije, prinos suve materije po korenu, obim korena i visina glave korena. Utvrđeno je povećanje mase korena i prinosa suve materije po korenu kod svih test hibrida u poređenju sa roditeljima. Za razliku od hibrida, kod roditelja je ustanovljena pozitivna korelacija između mase korena i sadržaja suve materije. Na osnovu karakteristika roditelja per se nije moguće predvideti superiorno hibridno potomstvo, jer su roditelji sa najmanjom masom korena dali hibride sa najvećom masom korena.

AB  - In this study we analyzed 10 monogerm and 10 multigerm sugar beet genotypes from the collections of the Institute of Field and Vegetable Crops in Novi Sad. The appearance of sugar beet root surface was evaluated as a property that has an impact on the soil removal and root washing in the sugar factory. The genotypes were tested with nonparametric Kruskal-Wallis's test and cluster analysis as a method of multivariate analysis. Based on these results, genotype 12 had the lowest values for the root surface (the least furrow of the root), which will further be used in sugar beet breeding programs.
AB  - U ovom istraživanju je analizirano 10 monogermnih i 10 multigermnih genotipova šećerne repe koji se nalaze u kolekciji Instituta za ratarstvo i povrtarstvo u Novom Sadu. Ispitivana je površina korena šećerne repe kao svojstvo koje ima uticaj na iznošenje zemljišta pri vađenju, kao i pranju korena tokom procesa prerade. Primenjeno je neparametarsko testiranje sa Kruskal-Wallisovim testom i analiza grupisanja kao metod multivarijacione analize. Na osnovu dobijenih rezultata, najmanju ocenu površine korena (najmanje brazdi na korenu) imao je genotip 12 koji će se koristiti za poboljšanje ovog svojstva kod drugih genotipova.

Meiose analyses of sugar beet triploids

AB  - During the seed crop growing there can be also problems with uncontrolled pollination if in the previous years there was seed shedding during the harvest, which can cause growth and flowering of diploid, triploid and tetraploid plants and that can present serious problem for the next sugar beet seed crop. Meiosis of triploid hybrids is irregular and can result in development of gametes with different chromosome number. Sugar beet triploids in metaphase I have 9 univalents and 9 bivalents and therefore can be source of aneuploid gametes. After cytological analysis that was performed on 2124 meiocytes, in 14.64% different types of irregularities were detected, such as dislocated chromosomes and chromosome bridges.
AB  - U semenskoj proizvodnji triploida šećerne repe u toku žetve dolazi do osipanja semena. Zbog toga u narednih nekoliko godina niču triploidne i tetraploidne biljke. Cilj rada je bio da se uradi citogenetska analiza nepravilnosti u mejozi kod triploidne hibridne sorte šećerne repe. Triploidi šećerne repe u Metafazi I imaju 9 univalenata i 9 bivalenata i mogu da budu izvor aneuploidnih gameta. Pojava aneuploida kao i njihova posledica delimična ili potpuna sterilnost, jesu problemi koji prate semensku proizvodnju šećerne repe. Acetokarmin squash metodom pregledano je ukupno 2124 mejocita, kod kojih su u 14,64% ustanovljene različite nepravilnosti. Konstatovane su nepravilnosti tipa dislociranih hromozoma, ubrzanih (izbeglih) i usporenih (izostalih), kao i hromozomski mostovi.

Effect of Azotobacter chroococcum application type on microorganisms in the rhizosphere and sugar beet yield

AB  - Plant growth promoting rhizobacteria (PGPR) are a group of diverse soil bacteria which stimulate the growth of the host plant. It has been shown that Azotobacter chroococcum may be used as biofertilizer for increasing the yield and improving technological characteristics of sugar beet. The effect of three different ways of inoculation of sugarbeet with Azotobacter chroococcum on the production features of sugar beet and microbiological status of rhizosphere soil has been tested. One variety of sugar beet, Drena, was included in the tests in the region Rimski Šančevi, Novi Sad, during 2008 and 2009. Five strains of Azotobacter chroococcum (1, 5, 8, 10 and 14) were used as microbiological fertilizers. There were three variations of inoculation: (A) incorporation in the soil, (B) before the fi rst cultivation, and (C) applying the liquid culture of strain on the seed before sowing. The highest increase in yield of sugar beet roots 6.25 t ha-1 and yield of white sugar 0.91 t ha-1 was achieved with strain 10 with variant of inoculation of soil before the first cultivation. In both years, a positive effect of Azotobacter chroococcum was observed on the total number of microorganisms in the rhizosphere.
AB  - Ispitan je efekat tri različita načina inokulacije šećerne repe sa sojevima Azotobacter chroococcum na parametre prinosa šećerne repe i mikrobiološki status zemljišta. U ispitivanjima je korišćena sorta Drena, a eksperiment je bio lociran na Rimskim Šančevima u toku 2008. i 2009. Kao mikrobiološko đubrivo korišćeno je pet sojeva Azotobacter chroococcum. Inokulacija je izvršena na tri načina: (A) inkorporacijom sojeva u zemljište pre setve, (B) inkorporacija sojeva u zemljište pred prvu međurednu kultivaciju i (C) nanošenjem inokuluma na seme pre setve. Najveće povećanje u prinosu korena od 6,25 t ha-1 i u prinosu kristalnog šećera od 0,91 t ha-1 dobijeno je sa sojem 10 na varijanti inokulacije zemljišta pre prve međuredne kultivacije. U obe godine dobijen je pozitivan efekat na ukupan broj mikroorganizama i broj azotobaktera u rizosferi.

abstract = "Plant growth promoting rhizobacteria (PGPR) are a group of diverse soil bacteria which stimulate the growth of the host plant. It has been shown that Azotobacter chroococcum may be used as biofertilizer for increasing the yield and improving technological characteristics of sugar beet. The effect of three different ways of inoculation of sugarbeet with Azotobacter chroococcum on the production features of sugar beet and microbiological status of rhizosphere soil has been tested. One variety of sugar beet, Drena, was included in the tests in the region Rimski Šančevi, Novi Sad, during 2008 and 2009. Five strains of Azotobacter chroococcum (1, 5, 8, 10 and 14) were used as microbiological fertilizers. There were three variations of inoculation: (A) incorporation in the soil, (B) before the fi rst cultivation, and (C) applying the liquid culture of strain on the seed before sowing. The highest increase in yield of sugar beet roots 6.25 t ha-1 and yield of white sugar 0.91 t ha-1 was achieved with strain 10 with variant of inoculation of soil before the first cultivation. In both years, a positive effect of Azotobacter chroococcum was observed on the total number of microorganisms in the rhizosphere., Ispitan je efekat tri različita načina inokulacije šećerne repe sa sojevima Azotobacter chroococcum na parametre prinosa šećerne repe i mikrobiološki status zemljišta. U ispitivanjima je korišćena sorta Drena, a eksperiment je bio lociran na Rimskim Šančevima u toku 2008. i 2009. Kao mikrobiološko đubrivo korišćeno je pet sojeva Azotobacter chroococcum. Inokulacija je izvršena na tri načina: (A) inkorporacijom sojeva u zemljište pre setve, (B) inkorporacija sojeva u zemljište pred prvu međurednu kultivaciju i (C) nanošenjem inokuluma na seme pre setve. Najveće povećanje u prinosu korena od 6,25 t ha-1 i u prinosu kristalnog šećera od 0,91 t ha-1 dobijeno je sa sojem 10 na varijanti inokulacije zemljišta pre prve međuredne kultivacije. U obe godine dobijen je pozitivan efekat na ukupan broj mikroorganizama i broj azotobaktera u rizosferi.",

AB  - Although sugar beet has been grown for only 250 years or so, breeding directions in this plant have changed more than in any other crop. Many type of changes occurred because of the influence of growers, consumers or pathogens that brought about a decrease in sugar beet growing areas. Work on sugar beet breeding at the Institute of Field and Vegetable Crops in Novi Sad began in 1939. The main goal of the sugar beet breeding program is to develop hybrids with high sugar yields tolerant to the prevalent pathogens and stress. During the last fifty years of intensive work at the Institute's Sugar Beet Department, 33 varieties of this crop have been registered in Serbia and 17 abroad.
AB  - Iako se šećerna repa gaji tek oko 250 godina, izmena pravaca oplemenjivanja su bila mnogo izraženija u odnosu na druge ratarske biljke. Razne promene koje su se desile kod šećerne repe pod uticajem bilo proizvođača, potrošača ili raznih patogena dovele su u pitanje proizvodnju ove biljne vrste u svetu. U Institutu za ratarstvo i povrtarstvo u Novom Sadu na oplemenjivanju šećerne repe se uspešno radi od 1939 godine. Osnovni pravac u oplemenjivačkom programu je stvaranje hibrida sa visokim prinosom šećera po hektaru i hibrida tolerantnih prema najznačajnijim patogenima i stresnim uslovima. Tokom proteklih 50 godina intenzivnog rada u Odeljenju za šećernu repu stvoreno je 50 sorata (33 u zemlji i 17 u inostranstvu).

Effect of biofertilizer application in sugar beet

AB  - Field trials were carried out in 2007 and 2008 on a number of commercial plots in Vojvodina in order to determine how the biofertilizer NS-Betafixin affects root yields, white sugar yields, and microbial abundance in the sugar beet rhizosphere. The use of NS-Betafixin resulted in a significant increase of yield compared with the control (untreated soil). The total number of microorganisms and azotobacter abundance increased as well, while the number of fungi and actinomycetes decreased. Overall, the application of NS-Betafixin can be regarded as a good tool for improving sugar beet yields both quantitatively and qualitatively. It may also reduce fertilizer application and production costs and is environmentally friendly.
AB  - Poljski ogledi su izvedeni tokom 2007. i 2008. godine na većem broju proizvodnih parcela u Vojvodini kako bi se utvrdio efekat primene mikrobiološkog đubriva NS-Betafixina na prinos korena, prinos kristalnog šećera, kao i brojnost mikroorganizama u rizosferi šećerne repe. Primena NS-Betafiksina dovela je do značajnog poboljšanja prinosa u poređenju sa kontrolom, tj. netretiranim zemljištima, a takođe je došlo do povećanja brojnosti mikroorganizama (ukupan broj i broj azotobaktera) i do smanjenja brojnosti gljiva i aktinomiceta na tretiranom zemljištu. Uopšteno, moglo bi se zaključiti da je biođubrenje, tj. primena NS-Betafixina dobar alat u poboljšanju prinosa šećerne repe kako kvantitativno tako i kvalitativno, može da redukuje primenu đubriva, tj. cenu proizvodnje, a uz to doprinosi poboljšanju životne sredine.

AB  - Rhizosphere appears as the most important niche for microbial activity. It is of direct relevance to nutrient cycling affecting plant nutrition. Rhizosphere can be defined as the root surface and the soil adhering to the root. The aim of this research was to determine the effect of inoculation with Azotobacter chroococcum on the number of microorganisms in the rhizosphere of sugar beet in dependence of fertilization. Field trials were conducted on a chernozem soil at the Rimski Šančevi Experiment Field of the Institute in 2002, 2003 and 2004. The sugar beet hybrid variety Sara was used. Inoculation of sugar beet was performed with a liquid culture of Azotobacter chroococcum strains, which was incorporated into the soil just before planting in the concentration of 109/ml. Differences were registered in population size depending on both the nitrogen dose and fertilisation method. On average, a higher percentage of increase was in the total number of microorganisms (35-118%) than in azotobacter population size (49-52.8%).
AB  - Cilj ovih istraživanja bio je utvrđivanje uticaja inokulacije sa azotobakterom na brojnost mikroorganizama u rizosferi u zavisnosti od doze azota i vrste đubrenja kod šećerne repe. Poljski ogledi su izvedeni u toku 2002, 2003. i 2004. godine na Rimskim Šančevima sa hibridnom sortom Sara stvorenom u Institutu za ratarstvo i povrtarstvo. U radu je određivan ukupan broj mikroorganizama na zemljišnom agaru (razređenja 106), broj gljiva na Čapekovom agaru (razređenja 104), aktinomiceta na sintetičkom agaru (razređenja 104) i azotobaktera na podlozi Fjodora (razređenja 102). Inokulacijom je povećan ukupan broj mikroorgainazama (35-118%), broj azotobaktera (49-52,8%), i gljiva (7,5-19,7%). Broj aktinomiceta je smanjen inokulacijom (1,6-5,4%). Najveći broj azotobaktera dobijen je na varijanti bez dodatka azota, a najmanji sa 200 kg N/ha. Najveći broj gljiva dobijen je sa 100 kg N/ha, a aktinomiceta sa 150 kg N/ha.

Influence of fertilization and nitrate-nitrogen position in soil profile on the sugar beet root yield and quality

AB  - Researches, which have lasted for two years, were carried out on long-term trial field at Rimski Šančevi, Novi Sad, Serbia. In this trial, the eight fertilization variants of N, P2O5 and K2O increased amounts were studied. Sugar beet root and tops yields were determined, as well as the elements of technological sugar beet root quality. Based on these results, percentage of sugar utilization and refined sugar yield was defined. In the spring, before applying of N fertilizer, amount of nitrate nitrogen in the soil and its influence on yield and quality was determined. The highest root yield in 2002 was produced at the variant N100 P150 K150, and in 2003 at the variant N150 P150 K150. However, in both years, referring to the variant N100 P100 K100, the differences were not statistically significant. Increasing of nitrogen amounts had negative effects on refined sugar yield. Amounts of NO3-N in the soil in spring, before sugar beet sowing, in 2002 had significant influence on root yield and refined sugar yield. In the year 2003, which was highly dry, high correlation ratio were gained between amounts of NO3-N in the soil and root quality parameters, but it wasn't significant between nitrogen amounts and root and refined sugar yield.
AB  - Dvogodišnja istraživanja uticaja rastućih količina NPK hraniva na prinos i kvalitet korena šećerne repe izvedena su na stacionarnom poljskom ogledu na Rimskim Šančevima. U proleće, pre primene N đubriva, praćena je količina nitratnog azota po slojevima zemljišta, te njegov uticaj na navedena svojstva. U obe godine, razlika u prinosu korena postignutog pri najvećim količinama NPK hraniva nije bila statistički značajna u odnosu na varijantu N100 P100 K100. Povećanje količine azota delovalo je negativno na prinos rafinisanog šećera. Količina NO3-N u zemljištu u proleće pre setve u 2002. godini imala je značajan pozitivan uticaj na prinos korena i rafinisanog šećera. U izrazito sušnoj 2003. godini visoki koeficijenti korelacije dobijeni su između količine NO3-N u zemljištu i parametara kvaliteta korena, ali nisu bili značajni između količine azota i prinosa korena i rafinisanog šećera.

abstract = "Researches, which have lasted for two years, were carried out on long-term trial field at Rimski Šančevi, Novi Sad, Serbia. In this trial, the eight fertilization variants of N, P2O5 and K2O increased amounts were studied. Sugar beet root and tops yields were determined, as well as the elements of technological sugar beet root quality. Based on these results, percentage of sugar utilization and refined sugar yield was defined. In the spring, before applying of N fertilizer, amount of nitrate nitrogen in the soil and its influence on yield and quality was determined. The highest root yield in 2002 was produced at the variant N100 P150 K150, and in 2003 at the variant N150 P150 K150. However, in both years, referring to the variant N100 P100 K100, the differences were not statistically significant. Increasing of nitrogen amounts had negative effects on refined sugar yield. Amounts of NO3-N in the soil in spring, before sugar beet sowing, in 2002 had significant influence on root yield and refined sugar yield. In the year 2003, which was highly dry, high correlation ratio were gained between amounts of NO3-N in the soil and root quality parameters, but it wasn't significant between nitrogen amounts and root and refined sugar yield., Dvogodišnja istraživanja uticaja rastućih količina NPK hraniva na prinos i kvalitet korena šećerne repe izvedena su na stacionarnom poljskom ogledu na Rimskim Šančevima. U proleće, pre primene N đubriva, praćena je količina nitratnog azota po slojevima zemljišta, te njegov uticaj na navedena svojstva. U obe godine, razlika u prinosu korena postignutog pri najvećim količinama NPK hraniva nije bila statistički značajna u odnosu na varijantu N100 P100 K100. Povećanje količine azota delovalo je negativno na prinos rafinisanog šećera. Količina NO3-N u zemljištu u proleće pre setve u 2002. godini imala je značajan pozitivan uticaj na prinos korena i rafinisanog šećera. U izrazito sušnoj 2003. godini visoki koeficijenti korelacije dobijeni su između količine NO3-N u zemljištu i parametara kvaliteta korena, ali nisu bili značajni između količine azota i prinosa korena i rafinisanog šećera.",

Maintenance and improvement of self-sterile sugar beet pollinators using tissue culture and recurrent selection

AB  - In vitro preservation makes it possible to maintain genotypes in an unchanged condition for a large number of years. With self-sterile genotypes, this method enables the use of recurrent selection for identifying genotypes with better combining abilities than the population average. By recombining selected superior genotypes, it is possible to obtain a new population with improved quantitative traits and combining abilities. The objective of the present paper was to develop a new population of the pollinator cultivar Lara from the progenies of superior genotypes kept in in vitro conditions. The monogerm hybrid cultivar Lara was developed through crosses using a monogerm male-sterile family with a narrow genetic base characterized by good combining abilities, tolerance to Cercospora and the possession of the Holly gene conferring resistance to rhizomania. The pollinator is a diploid multigerm population having tolerance to Cercospora and Rizor genes for tolerance to rhizomania. Used in our study were phenotypically superior pollinator genotypes contained in individual plants within the population. The newly developed homogenous clonal population was kept in a gene bank. A high propagation rate was obtained by stimulating axillary buds from the vegetative tissue of the inflorescence. Based on the hybrids' production characteristics, superior pollinator genotypes were propagated vegetatively and a new population of the pollinator Lara was developed.
AB  - U in vitro uslovima moguće je dugi niz godina sačuvati genotipove u neizmenjenom stanju. Ova metoda kod autosterilnih genotipova omogućuje pri menu rekurentne selekcije u identifikaciji genotipova sa boljim kombinacionim sposobnostima od proseka populacije. Rekombinacijom odabranih superiornih genotipova dobijena je nova populacija sa poboljšanim kombinacionim sposobnostima. Monogermna hibridna sorta Lara nastala je ukrštanjem monogermne muško sterilne familije uske genetičke osnove, dobrih kombinacionih sposobnosti, tolerantna na cerkosporu i sa Holly genom otpornosti prema rizomaniji. AB  - Vegetative in vitro multiplication is one of the most efficient methods for sugar beet (Beta vulgaris L.) propagation. The usual steps in this procedure are sterilization of explant, multiplication, rhizogenesis and acclimatization. In the paper is presented development of regeneration and multiplication techniques from different explants. It gives a detailed description of further micropropagation steps and presents necessary conditions for their realization. The paper also discuss different ways of micropropagation application especially in sugar beet breeding, but in other plant sciences as well.

AB  - In the forty years of seminars of agronomists, the genetic composition of sugar beet hybrids and varieties has been completely changed, mainly due to development of different scientific methods. Every change lead to increase of genetic yield potential for important quantitative and qualitative traits, or improvement of efficiency and profitability of production. Sugar beet breeding in Institute of field and vegetative crops always managed to follow European and world trends, in order to create hybrid and varieties whose performance was equal to hybrids made by international companies. Increase in yield potential of 2% per year in the last few decades, is mostly due to genotype improvement, which presents huge success of scientists involved in sugar beet genetics and breeding.
AB  - U proteklih četrdeset godina u vremenu trajanja seminara agronoma zahvaljujući rezultatima fundamentalne nauke mnogo češće nego kod drugih gajenih biljaka kod šećerne repe je nekoliko puta dolazilo do kompletne promene genetičke kompozicije gajenih sorti šećerne repe. Svaka promena dovodila je do povećanja genetičkog potnecijala rodnosti za najvažnije kvantitativne karakteristike ili do povećanja ekonomičnosti i profitabilnosti proizvodnje. Oplemenjivanje šećerne repe u Naučnom institutu za ratarstvo i povrtarstvo je uglavnom vrlo efikasno pratilo evropske i svetske trendove u smislu stvaranja sorti koje po svojim karakteristikama nisu zaostajale za sortama multinacionalnih kompanija. U zadnjih nekoliko dekada povećanje potencijala rodnosti koje se duguje isključivo poboljšanom genotipu iznosi 2% godišnje što predstavlja nesumnjivo veliki uspeh genetičara i oplemenjivača šećerne repe.

AB  - Coat protein gene of beet necrotic yellow vein virus (BNYVV) was isolated from inoculated sugar beet roots and leaves of Chenopodium quinoa and Tetragonia expansa, by RT-PCR and imuno capture RT-PCR. Specific primers were made to complement coat protein gene and untranslated leader sequence, so that two fragments were obtained: long (731 bp), which contained coat protein gene and leader sequence, and short (587 bp), with coat protein gene. Fragments were cloned in two plant transformation vectors: pCAMBIA 3301M and pCAMBIA 1304M, which were modified by removing multicloning site and NcoI restriction site at the 5' end of the reporter genes. Vector pC3301M had bar gene which confers resistance against the herbicide gluphosinate ammonium as selectable marker, and pC1304M had gene for resistance to antibiotic hygromycin. Three constructs were made from each vector: CPL, containing coat protein gene with leader sequence; CPS with gene for coat protein, and CPSas with coat protein gene in antisense orientation. All constructs were transfered to Agrobacterium tumefaciens strain LBA4404.

Construction of plant transformation vectors carrying beet necrotic yellow vein virus coat protein gene (ii)- plant transformation

AB  - Fragments containing the coat protein gene of beet necrotic yellow vein virus were cloned in two plant transformation vectors: pCAMBIA3301M with the bar gene as selectable marker, and pCAMBIA1304M, with resistance to hygromycin. Three constructs were made of each vector: CPL, containing coat protein gene with leader sequence; CPS with coat protein gene, and CPSas with coat protein gene in antisense orientation. Vectors pC3301MCPL, pC3301MCPS. and pC3301MCPSas were used in Agrobacterium—mediated transformation of Nicotiana tabacum (tobacco), Nicotiana excelsior and Nicotiana benthamiana. Regenerants that developed roots on selective media were tested for the presence of CP fragments and the bar gene, but most regenerants were nontransformed (50-83% escapes). After all rooted plants had been selfed, and T1 seed germinated on selective media, only plants descending from one N. excelsior regenerant transformed with pC3301MCPS were positive for presence of bar gene and CPS fragment. Tobacco and Nicotiana benthamiana were transformed with constructs pC1304MCPS and pC1304MCPSas. Transformation efficiency was much higher and approximately 50% of regenerants that rooted on media with 20 mg l−1 hygromycin were positive for the presence of CP fragments. All T1 plants were positive for presence of CP fragments.

AB  - In 2001 and 2002 seven sugar beet varieties with different level of rhizomania resistance were tested on five locations, for root yield, sugar content and yield stability. Analyzed traits depended on agroecological conditions and varied from 44.96 t/ha to 95.00 t/ha for root yield, 11.30% to 16.56% for sugar content and 5.08 t/ha to 13.54 t/ha for sugar content. Varieties Mara and Sara had the highest sugar content. High general adaptability for root and recoverable sucrose yield had variety Drena, while variety Sara had significantly higher stability for those traits.
AB  - Tokom 2001. i 2002. godine na 5 lokaliteta je ispitivano 7 sorti šećerne repe različitog nivoa tolerantnosti prema rizomaniji. Utvrđen je prinos korena sadržaj šećera i prinos šećera i po modelu Eberhart i Russell (1966) izračunata stabilnosti sorti. U zavisnosti od faktora spoljne sredine prinos korena je varirao od 44,96 t/ha do 95,00 t/ha, sadržaj šećera od 11,30% do 16,56% i prinos polarizacionog šećera od 5,08 t/ha do 13,54 t/ha. U sadržaju šećera prednost su pokazale sorte Mara i Lara. Visoku opštu adaptabilnost za prinos korena i prinos šećera pokazala je sorta Drena, dok je sorta Sara pokazala natprosečnu stabilnost za ova svojstva.

AB  - Fragments of viral cDNA containing the coat protein gene of beet necrotic yellow vein virus were cloned in plant transformation vector pCAMBIA3301M with the bar gene as selectable marker. Vector pC3301MCPL carrying coat protein gene with leader sequence, and pC3301MCPS with coat protein gene, were used in Agrobacterium - mediated transformation of sugar beet. The transformation method used was based on the fact that sugar beet develops axillary shoots in in vitro conditions, when placed on media with citokinins. Since this ability is not genotype or ploidy dependant it is widely used for sugar beet vegetative multiplication. Sterile seedlings, with removed cotyledons and lower half of hypocotyl, were used as starting material. After transformation ex-plants were put on micropropagation medium with cephotaxime and phosphinotricyn (ppt), where axillary shoots started to develop. Since concentration of ppt was not selective enough, after two subcultivations it was increased twofold. Only one sample, transformed with pC3301MCPS preserved morphogenetic potential for micropropagatio, and it was tested for presence of COS fragment and bar gene bz PCR with soecific primers.
AB  - Fragmenti virusne cDNK sa genom za protein omotača virusa nekrotičnog žutila nerava repe su klonirani u vektor za transformaciju biljaka pCAM-BIA3301M koji je sadržao bar gen kao selektivni marker. Vektori pC3301MCPL, sa genom za protein omotača virusa i njegovom lider sekvencom, i pC3301MCPS, sa genom za protein omotača, su korišćeni u tramsformaciji repe pomoću Agrobacterium-a. Metod transformacije se zasniva na sposobnosti repe da u uslovima in vitro razvije aksilarne pupoljke na podlozi sa citokininima. Pošto ova sposobnost ne zavisi od genotipa ili od nivoa plodnosti, postala je standardni metod za vegetativno umnožavanje repe. Kao početni materijal su korišćeni sterilni ponići kojima su odstranjeni kotiledoni i donja polovina hipokotila. Nakon transformacije eskplantati su postavljeni na selektivnu podlogu za mikropropagaciju sa cefotaksimom i fosfinotricinom (ppt) gde je došlo do razvoja bočnih pupoljaka. Po.sto koncentracija fosfinotricina nije bila dovoljno selektivna, ona je nakon dve subkultivacije dvostruko povećana. Samo je jedan uzorak, transformisan vektorom pC3301MCPS, nakon dve subkultivacije sačuvao mofrogenetski potencijal za mikropropagaciju, i bio testiran na prisustvo CPS fragmenta i bar gena PCR reakcijom sa specifičnim prajmerima.

AB  - The objective of this study was to assess the effect of inoculation on Azotobacter population size in dependence of fertilizer dose and fertilization method. Differences were registered in Azotobacter population size which depended on both, nitrogen dose and fertilization method. On average, the highest percentage of increase in Azotobacter population size in relation to the non-inoculated variant, was registered in the variant with nitrogen, liquid manure and harvest residues. The largest increase in Azotobacter population size was obtained in the inoculation variants.

AB  - Increased yield of sugar beet depends on hybrids that are grown agroecological conditions and applied agrotechnic. Last years yield increasing depends only of genotype, in EC is 2% per year and is result of improved yield potential and disease resistance. This is great success for sugar beet genetics and breeding. Rhizomania is caused by Beet necrotic yellow vein (BNYW) which transferred by fungus Polymixa betae. Considering the yield reduction and changes in plant growth and development, rhizomania is economically the most important sugar beet disease. In the paper are described all known types of resistance-tolerance: Alba, Rizor and Holly type, and presented results of three year field trials on four locations. Four hybrid varieties were tested: Sara, Lara, Drena and Darija, accepted by Plant variety commission, Ministry of agriculture and water management Republic of Serbia and one experimental hybrid. There most important quantitative trais were tested and than compared with standard varieties. It could be observed that varieties did not significantly differ among themselves, which was to be expected since all four them are resistant to rhizomania.
AB  - Povećanje rodnosti kod šećerne repe u najvećoj meri zavisi od hibrida koji se gaji, klimatskih uslova i primenjene agrotehnike gajenja. U zadnjih nekoliko dekada povećanje prinosa koje se duguje isključivo genotipu, u zemljama EEC je 2% godišnje, što predstavlja nesumnjivo veliki uspeh genetičara i oplemenjivača šećerne repe. To povećanje prinosa ogleda se ne samo povećanjem genetskog potencijala rodnosti već i poboljšanjem otpornosti prema najznačajnijim bolestima i štetočinama. Jedno od najštetnijih oboljenja koje je do sada utvrđeno i opisano kod šećerne repe je Rizomanija, a najefikasnija mera borbe je stvaranje tolerantnih ili otpornih hibridnih sorti. Najnovije NS-hibridne sorte šećerne repe Sara, Lara, Drena, Darija i NS-Hy-031R koje su otporne prema rizomaniji u ogledima pokazuju visok genetički potencijal za najvažnije kvantitativne osobine.

abstract = "Increased yield of sugar beet depends on hybrids that are grown agroecological conditions and applied agrotechnic. Last years yield increasing depends only of genotype, in EC is 2% per year and is result of improved yield potential and disease resistance. This is great success for sugar beet genetics and breeding. Rhizomania is caused by Beet necrotic yellow vein (BNYW) which transferred by fungus Polymixa betae. Considering the yield reduction and changes in plant growth and development, rhizomania is economically the most important sugar beet disease. In the paper are described all known types of resistance-tolerance: Alba, Rizor and Holly type, and presented results of three year field trials on four locations. Four hybrid varieties were tested: Sara, Lara, Drena and Darija, accepted by Plant variety commission, Ministry of agriculture and water management Republic of Serbia and one experimental hybrid. There most important quantitative trais were tested and than compared with standard varieties. It could be observed that varieties did not significantly differ among themselves, which was to be expected since all four them are resistant to rhizomania., Povećanje rodnosti kod šećerne repe u najvećoj meri zavisi od hibrida koji se gaji, klimatskih uslova i primenjene agrotehnike gajenja. U zadnjih nekoliko dekada povećanje prinosa koje se duguje isključivo genotipu, u zemljama EEC je 2% godišnje, što predstavlja nesumnjivo veliki uspeh genetičara i oplemenjivača šećerne repe. To povećanje prinosa ogleda se ne samo povećanjem genetskog potencijala rodnosti već i poboljšanjem otpornosti prema najznačajnijim bolestima i štetočinama. Jedno od najštetnijih oboljenja koje je do sada utvrđeno i opisano kod šećerne repe je Rizomanija, a najefikasnija mera borbe je stvaranje tolerantnih ili otpornih hibridnih sorti. Najnovije NS-hibridne sorte šećerne repe Sara, Lara, Drena, Darija i NS-Hy-031R koje su otporne prema rizomaniji u ogledima pokazuju visok genetički potencijal za najvažnije kvantitativne osobine.",

Induction and micropropagation potential of sugar beet haploids

AB  - The aim of research was obtaining sugar beet haploids via gyno-genesis and their micropropagation. Haploids were obtained by ovule culture from fourteen diploid, monogerm, fertile genotypes. On the tested nutrient media genotypes exhibited different gynogenic potential. Eight haploid plant were chosen for further investigation and after development of first leaves put on micropropagation medium. The presence of cyto-kinin in medium stimulated development of axillary buds, while in some genotypes adventitious buds developed as well. Multiplication rate was not consistent, although number of developed plants grew after each sub-cultivation. Differences in plant multiplication started to differ after four subcultures. By testing of differences between correlation coefficients, i.e. multiplication rate during six subcultivations, it was determined that they significantly differ between tested genotypes.
AB  - Cilj istraživanja je bio dobijanje haploida šećerne repe putem ginogeneze i njihova mikropropagacije. Haploidi su dobijeni kulturom semenog zametka iz Četrnaest diploidnih, monogermnih i muški fertilnih genotipova šećerne repe. Ispitivani genotipovi su na podlogama sa različitom koncentracijom stimulatora rasta ispoljili različit potencijal za ginogenezu. Za dalja ispitivanja je odabrano osam haploida koji su nakon razvića prvog para listova postavljeni na podlogu za mikropropagaciju. Prisustvo citokinina u hranljivoj podlozi je stimulisalo pojavu i razviće bočnih pupoljaka u pazuhu listova, dok je kod nekih genotipova takođe došlo do pojave adventivnih pupoljaka. Porast broja biljaka tokom mikropropagacije nije bio kontinuiran iako je broj novostvorenih biljaka rastao posle svake sub-kultivacije. Razlike u intenzitetu multiplikacije biljaka počele su da se značajno ispoljavaju nakon Četvrte subkultivacije. Testiranjem značajnosti razlika korelacionih koeflcijenata, odnosno intenziteta mikropropagacije ispitivanih genotipova tokom šest subkultivacija, utvrđeno je da se one međusobno statistički značajno razlikuju.

It could be observed that varieties did not significantly differ among themselves, which was to be expected since all three of them are resistant to rhizomania. Regularity in trials were noted but not statistically proved that variety Lara has high sugar content, Sara is normal type variety, and Drena has high root yield.
AB  - U radu su prikazani rezultati trogodišnjih ispitivanja šećerne repe na većem broju lokaliteta. Ispitivane su hibridne sorte šećerne repe otporne prema Rizomaniji koje su priznate od strane Zavoda za biljne i životinjske genetičke resurse, Komisije za priznavanje sorti pod nazivom Sara, Lara i Drena i jedan eksperimentalni hibrid koji se nalazi u ispitivanju. Ispitivane su tri najznačajnije kvantitativne osobine: prinos korena sadržaj šećera i prinos kristalnog šećera i upoređivanje sa standardnim sortama. Između tri priznate sorte nisu ustanovljene značajne razlike, jer se radi o sortama otpornim prema Rizomaniji. Pravilnost koja je u ogledima uočena ali nije statistički potvrđena je da sorta Lara ima naglašen sadržaj šećera, Sara je normalnog tipa a Drena ima naglašen prinos korena.
