Trypsin inhibitor activity in grass pea seeds (Lathyrus sativus L.)

AB  - Grass pea (Lathyrus sativus) is a valuable grain legume known for its high protein content and rich essential amino acid profile. Its exceptional characteristics such as drought tolerance, high adaptability to extreme conditions, disease resistance, and low cultivation inputs, make it particularly suitable for cultivation by resource-poor farmers. However, the potential use of grass pea is limited due to the presence of anti-nutritional factors, including protease inhibitors, especially trypsin inhibitor. This study aimed to develop a rapid and reliable method for measuring TI activity in seeds of grass pea and to investigate the influence of genotype and environment on trypsin inhibitor (TI) activity in seeds of grass pea. A set of 25 grass pea accessions from seven European countries was cultivated in Slovenia at the Agricultural Institute of Slovenia, and a set of 12 grass pea accessions from the Serbian gene bank was grown in Serbia at the Institute of Field and Vegetable Crops. The TI activity varied considerably among the grass pea accessions studied, with values ranging from 26.7 to 90.3 TUI/mg. To further evaluate the effects of environmental conditions on TI activity, eight grass pea accessions originating from Serbia were grown in both Slovenia and Serbia. The TI activity of the accessions grown in Slovenia ranged from 26.7 to 81.0 TUI/mg, while the activity of the accessions grown in Serbia ranged from 40.3 to 57.0 TUI/mg. The correlation of TI activity between grass pea accessions grown in Slovenia and those grown in Serbia was 0.39, with genotype diversity being the largest contributor (55.9%). This study provides a valuable insight into the variability of TI activity in grass pea and shows the possible influence of environmental conditions on this trait. However, since the data are only from a one-year field trial, further studies are needed to fully evaluate the influence of different environmental factors on TI activity.

AB  - Alternaria brassicae i Alternaria brassicicola prouzrokuju ekonomski značajne štete na biljkama iz porodice kupusnjača. AB  - Charcoal rot of sugar beet, caused by Macrophomina phaseolina in recent years has become a main concern for farmers in Serbia. Since control measures are mostly preventive with only partial effect, introduction of alternative measures, such as biological control based on augmentation of mycoparasites, represents the most promising approach. ITS region and tef gene sequence analysis of ten Trichoderma spp. isolates from sugar beet and soybean rhizosphere was done to confirm species identity. All isolates were confirmed to be Trichoderma harzianum.

AB  - Sugar beet is one of the most important industrial crops. Beet sugar now accounts for almost all sugar production in the Europe and for about 20% of total world production. Crop improvement is important in order to enhance productivity, sugar content or other desirable traits, and molecular techniques can help to accelerate such efforts. One of our research interests is focused on to the development of new sugar beet lines and hybrids combining marker assisted selection with conventional breeding methods. DNA markers were used to assess the genetic diversity and distance among breeding lines and correlate them with hybrid performance, in order to define the heterotic groups and predict heterosis effect. Our activities also involve the investigation of different reaction of sugar beet genotypes to reduced water supply, as the major limiting environmental factor in sugar beet growing.

AB  - The genus vetch (Vicia L.) includes over 200 herbaceous annual and perennial species, containing food and forage legumes among them, with a prevailing Euro-Asiatic distribution. There are only few studies published considering the assessment of genetic diversity in Vicia using DNA markers, except for faba bean, where these were used extensively. The development of novel markers requires high costs and is time consuming. Instead of developing new markers, their transfer within genus is an alternative in less studied species. Having that in mind, the goal of This research was to investigate the possibility of transfer of SSR and ISSR markers within Vicia genus. Cross-species amplifications of the 6 SSR and 4 ISSR primers were carried out, using genomic DNA isolated from three accessions of eight Vicia species. Average transferability for SSR markers was 56.25%. Primers VfG14 and GAI1 displayed the highest, while Vig24 had the lowest rate of transfer.

AB  - The development of novel markers usually requires high costs and is time consuming. Instead of developing new markers, their transfer within a genus is an alternative particularly suitable in less studied species. Hence the goal of this study was to evaluate the ability of SSR markers, previously developed from Vicia faba, and ISSR markers to estimate DNA polymorphism within the genus Vicia. Eight Vicia species were used for cross-species amplification. Transferability of SSR markers in the studied species ranged from 25% to 100%, with an average of 56.25%. Although three SSR primers were polymorphic, the nature of the amplification products indicated that their further usefulness is questionable. ISSR primers generated polymorphic products of amplification in all tested Vicia species. The average polymorphism of ISSR primers was 93.75%, which implies that these primers can be used within the genus Vicia.
AB  - Razvoj markera obično zahteva vreme i velika materijalna ulaganja. Umesto razvijanja novih markera za manje proučavane vrste, alternativa može biti transfer sa vrsta unutar istog roda. Cilj ovog rada je bio da se proceni mogućnost primene SSR prethodno razvijenih na Vicia faba i ISSR prajmera u proceni DNK polimorfizma u rodu Vicia. Prenosivost SSR markera se kretala u intervalu 25-100% sa prosečnom vrednošću od 56,25%. Iako su produkti tri SSR prajmera bili polimorfni, zbog dobijene količine i kvaliteta, njihovo korišćenje u daljim istraživanjima je diskutabilno. Svi ispitivani ISSR prajmeri su davali različite profile amplifikacije za svaku ispitivanu vrstu. Prosečni procenat polimorfizma ISSR prajmera je bio vrlo visok (93,75%), što sugeriše da ovi prajmeri mogu biti korišćeni u proceni DNK polimorfizma unutar roda Vicia.
