AB  - Oil and protein content in wheat grain are quality indicators important for food processing, cosmetics
and pharmaceutical industries. These and many other quality parameters could be signiticantly
influenced by genotype, growing conditions and cultivation practices, especially fertilization. The
objective of this study was to investigate effects of genotype, growing season, mitrogen (N)
fertilization and their interactions on the oil and protein content in 24 varieties of winter wheat
(Triticum aestivum, L.). Field trials with two N rates (low nitrogen N4s and high nitrogen Nii) were
conducted at the location of Rimski Sanéevi, Serbia, during two growing seasons. The oil was
extracted from wheat bran obtained by laboratory mill MLU 202. Classical Rushkovsky method was
used to determine oil content, while protein content was determined by the ICC 105/2 method.
Significant variability was found among the genotypes for both analysed traits. In two growing
seasons and at different N rates, the oil content varied from 2.02% to 5.58%, with the average value
of 3.96% and coefficient of variation (CV) 9.2%. The protein content ranged from 10.7% to 17.7%
with the CV of 3.1%. All sources of variation (genotype - G, year - Y and N fertilization - F) and their
interactions had significant effects on oil and protein content, except the Y x F interaction on the
latter. Regarding the oil content, the effect of the year was stronger than the effect of nitrogen. In
contrast, protein content was more affected by the N fertilization than by the year. Principal
component analysis was used for grouping genotypes according to their stability and reaction to
different growing environments. Cvs. Cipovka, Dragana and Simonida were identified as very stable
with high oil content in different growing conditions, while cvs. Bankut 1205 and Banatka were
identified as potential sources of high protein content. The identified genotypes can serve as parents
in wheat breeding for higher oil and protein content.

AB  - Wheat bran is specifically rich in dietary fibre, essential fatty acids and contains important quantities of starch, protein, vitamins, dietary minerals and phytic acid. It is also well known medicinally for its powerful anticarcinogenic and antioxidant properties. Wheat bran is rich source of essential fatty acids, such as linoleic acid (18:2n6) and linolenic acid (18:3n3). Inoleic acid and fatty acid of n6 and n3 series play an important role in the odulation of human metabolism. Linoleic acid is undoubtedly one of the most location of Rimski Sančevi (Vojvodina Province, Serbia) to evaluate oil tents and fatty acid profiles in a collection of 25 wheat genotypes.

High-performance liquid chromatography determination of tocopherols in wheat bran

AB  - Natural vitamin E is significant phytochemical compound with antioxidant activity
and potential benefit for human health. The physiological role of vitamin E is in
its ability to react with free radicals in cell membranes and preventing lipid
peroxidation. Vitamin E is composed of eight different structurally related
compounds, vitamers (a-, B-, y- and 65-tocopherols and a-, B-, y- and otocotrienols) which are known to occur in nature. Among cereals, wheat grain
and wheat bran represent rich sources of tocopherols. The aim of this study was
to investigate tocophérols composition (a-, B-, y- tocopherols) in selected 20
wheat genotypes. The samples consisted the bran portion of the outer layer of
wheat grain that were the by-product of milling on the MLU 202 laboratory flour
mill. Tocopherols were determined in the wheat bran samples by using the
simple analytical method which includes extraction with hexane and normalphase high-performance liquid chromatography with fluorescence detection. The
mobile phase consisted a mixture of hexane:ethyl acetate (70:30, viv). Total
tocopherols content in wheat bran extracts ranged from 28.08 in cv. Partizanka
to 107.49 mg/kg in cv. Skopljanka The average contents of a-, B- and ytocopherols were in the range from 3.16 (a-tocopherol in cv. Partizanka) to
81.04 mg/kg (y- tocopherol in cv. Skopljanka). Major vitamers for those tested
genotypes were a- and y- tocopherols. The results have shown that there were
significant differences in tocopherols composition and content among the
genotypes, indicating the great genetic potential for improvement.

AB  - Wheat is one of the most widespread plant species, primarily because of its adaptability and
diverse uses. Increased application of wheat oil in food, beauty and pharmaceutical industries
has led to an increased importance of oil content as an indicator of quality. Wheat oil has
beneficial effects on human health due to its high content of tocopherols and polyunsaturated
fatty acids. The most important chemical traits of tocopherols are their antioxidant activities
and biological function in vitamin E activities. The aim of this study was to determine the variability of oil and tocopherols content in 24 genetically divergent wheat cultivars from
different selection cycles during a two-year period. The oil was extracted from wheat bran
obtained by using laboratory mill MLU 202. The classic Rushkovsky method was used to determine the oil content, while tocopherols (a, B y) were simultaneously determined using
hexane extraction and liquid chromatography with detection of fluorescence. The average oil content during two years ranged from 4.1% to 4.3%, while the minimum and maximum
values ranged from 3.2% to 4.9%. The coefficient of variation (CV) value was 8.6%. The
average content of a tocopherols during two years ranged from 9.1 to 13.1 mg/kg bran (min.
and max. values: 3.8-18.5 mg/kg bran), 8 tocopherols 6.5-8.2 mg/kg bran (min. and max.
values: 2.9-11.1 mg/kg bran) and y tocopherols 31.1-53.0 mg/kg bran (min. and max. values:
29.2-67.4 mg/kg bran). The CVs for the contents of a, B and y tocopherols were 8, 4.6 and 3.4%, respectively. The effect of genetic factors and environment (and their interaction), were
highly significant for all analysed parameters, and highly significant correlations were found
between them (correlations varied from 0,871** between oil content and y tocopherols to 0.990** between o and B tocopherols). Cluster analysis showing the degree of similarities was used to group the analysed varieties into clusters, which can serve as the basis for the selection of parental components to further the wheat breeding for higher oil content and quality. The results obtained from wheat bran have shown lower oil content and higher
content of y tocopherols in relation to the direct extraction from the wheat germ. Varieties with higher oil and tocopherol content can be used as a good raw material for whole-wheat
products with important benefits to the human health.

diverse uses. Increased application of wheat oil in food, beauty and pharmaceutical industries
has led to an increased importance of oil content as an indicator of quality. Wheat oil has
beneficial effects on human health due to its high content of tocopherols and polyunsaturated
fatty acids. The most important chemical traits of tocopherols are their antioxidant activities
and biological function in vitamin E activities. The aim of this study was to determine the variability of oil and tocopherols content in 24 genetically divergent wheat cultivars from
different selection cycles during a two-year period. The oil was extracted from wheat bran
obtained by using laboratory mill MLU 202. The classic Rushkovsky method was used to determine the oil content, while tocopherols (a, B y) were simultaneously determined using
hexane extraction and liquid chromatography with detection of fluorescence. The average oil content during two years ranged from 4.1% to 4.3%, while the minimum and maximum
values ranged from 3.2% to 4.9%. The coefficient of variation (CV) value was 8.6%. The
average content of a tocopherols during two years ranged from 9.1 to 13.1 mg/kg bran (min.
and max. values: 3.8-18.5 mg/kg bran), 8 tocopherols 6.5-8.2 mg/kg bran (min. and max.
values: 2.9-11.1 mg/kg bran) and y tocopherols 31.1-53.0 mg/kg bran (min. and max. values:
29.2-67.4 mg/kg bran). The CVs for the contents of a, B and y tocopherols were 8, 4.6 and 3.4%, respectively. The effect of genetic factors and environment (and their interaction), were
highly significant for all analysed parameters, and highly significant correlations were found
between them (correlations varied from 0,871** between oil content and y tocopherols to 0.990** between o and B tocopherols). Cluster analysis showing the degree of similarities was used to group the analysed varieties into clusters, which can serve as the basis for the selection of parental components to further the wheat breeding for higher oil content and quality. The results obtained from wheat bran have shown lower oil content and higher
content of y tocopherols in relation to the direct extraction from the wheat germ. Varieties with higher oil and tocopherol content can be used as a good raw material for whole-wheat
products with important benefits to the human health.
