Antioxidant properties of soybean seedlings grown in different soil types

AB  - The purpose of this work was to assess change in antioxidant ability and accumulation of antioxidants in soybean seedlings, as well as some morphological parameters (plant height, shoot and root mass) during abiotic stress. Activity of antioxidant enzymes [superoxide dismutase (SOD; EC 1.15.1.1) and peroxidases (GPX-guaiacol peroxidase and PPX-pyrogallol peroxidase, EC 1.11.1.7)], phenylalanine ammonia lyase (PAL; EC 4.3.1.5), as well as content of non-enzymatic antioxidants (total phenolics, tannins and flavonoids) were analyzed in leaves of soybean seedlings grown in different soil types [mollic vertisol (humic); calcic chernozem (calcic) and haplic fluvisol (calcaric)]. Seedlings grown in fluvisol had significantly higher activity of SOD (7.5-8.6%) and PAL (39.2-43.0%) in leaves, as well as total phenolics and tannins content (8.1-80.7%) in comparison to those grown in chernozem and vertisol. However, soybean grown in chernozem had the most favourable conditions for growth and the highest total flavonoids content (2.3 mg rutin g-1 dry weight). Regardless the difference in response to higher and lower level of soil fertility, soybean seedlings grown in vertisol and fluvisol had similar morphological properties, which showed the great ability of their antioxidant systems to acclimate to change in soil quality.
AB  - Cilj ovog rada bio je da se utvrdi da li akumulacija antioksidanata i promene u antioksidantnoj sposobnosti u klijancima soje utiču na morfološke parametre (visinu biljke i masu nadzemnog i podzemnog dela biljke). Analizirane su promene u aktivnosti antioksidantnih enzima [superoksid-dismutaze (SOD; EC 1.15.1.1) i peroksidaza (GPX-gvajakol i PPX-pirogalol peroksidaze, EC 1.11.1.7)], fenilalanin amonijum-lijaze (PAL; EC 4.3.1.5), kao i sadržaju neenzimskih antioksidanata (ukupnih fenola, tanina i flavonoida) u listu klijanaca soje gajene na različitim tipovima zemljišta [ritska crnica, černozem i fluvisol]. U listu klijanaca soje gajenih na fluvisolu zabeležena je značajno veća aktivnost superoksid-dismutaze (7.5-8.6%) i fenilalanin amonijum-lijaze (39.2-43.0%, kao i sadržaj ukupnih fenola i tanina (8.1-80.7%) u odnosu na klijance gajene na černozemu i ritskoj crnici. Ipak, klijanci soje gajeni na černozemu imali su najbolje uslove za rast i razvoj, kao i najveći sadržaj flavonoida (2.3 mg rutina g-1 suve mase). Pored toga, morfološke karakteristike klijanaca soje gajenih na ritskoj crnici i fluvisolu se nisu značajno razlikovale, što navodi na zaključak na izuzetno izraženu adaptabilnost antioksidantnog sistema ovih biljaka na promene u kvalitetu zemljišta.

Correlation between lipid peroxidation and phenolics content in leaves and roots of sugar beet infected with Rhizoctonia solani

AB  - The correlation between intensity of lipid peroxidation and changes in antioxidant capacity of sugar beet plants (cv. `Drena') infected with Rhizoctonia solani Kuhn isolate ( AG 2- 2 IIIB group) was studied. Successful inoculation was confirmed by the presence of infection cushions in a cross section of leaf petioles. On the 7th day of the experiment, phenylalanine ammonia-lyase ( PAL; EC. 4.3.1.5) activity was in negative correlation with intensified lipid peroxidation process in leaves of sugar beet plants ( r= - 0.99). Also, in leaves and roots of inoculated sugar beet plants, total flavonoids content ( 35% and 20%, respectively) and 1,1- diphenyl- 2-picrylhydrazyl (DPPH)-scavenging activity ( 80% and 55%, respectively) were significantly reduced. Necrotic processes resulting from R. solani infection of sugar beet plants was followed by induction of plant phenolics metabolism; however, antioxidant capacity of these plants was reduced.

Changes in the lipid peroxidation intensity in auxin treated cherry rootstocks softwood cuttings

AB  - In order to investigate the effect of phytohormones exogenous auxins on the level of the mechanical injury induced oxidative stress, intensity of lipid peroxidation (LP) was measured in rootstocks of cherry softwood cuttings. Basal part of the cuttings and leaves of five rootstocks (Mahaleb 1 and 2, Gisela 5, European ground cherry (EGC) and “Oblačinska” sour cherry) were sampled 0, 2, 4 and 6 days after cutting. Cuttings were treated with 0.5% solutions of three auxins: α-naphthylacetic acid (NAA), indolebutyric acid (IBA) and combination of these two (INCIT K). Results obtained for the LP intensity varied depending on the rootstock, plant organ and auxin applied. The best LP-lowering effect of auxins occurred in all genotypes when treated with NAA. In the leaves, the LP peaked on the 2nd day, while the best LP-lowering effect was recorded in European ground cherry rootstocks on the 4th day, independently of auxin applied. The highest MDA production in leaves was recorded in Mahaleb 2 and European ground cherry rootstocks treated with IBA (60-90% higher than control) two days after the cuttings were made. It has been established that in the most of the rootstocks examined auxins showed lowering effect on LP which points to their positive effect, not only on rooting of softwood cuttings, but on their antioxidant protection system, as well.
AB  - U radu je ispitan nivo oksidativnog stresa izazvanog mehaničkom povredom biljke prilikom proizvodnje reznica, u bazalnom delu i listovima pet podloga za višnju i trešnju (Magriva 1 i 2, Gizela 5, Stepska (EGC) i “Oblačinska” višnja). Istovremeno, reznice su tretirane 0.5% rastvorima tri auksina (α-naftilsirćetna kiselina, NAA, indol- buterna kiselina, IBA i kombinacija ovih hormona, INCIT K), u periodu od 60 min, kako bi se utvrdio efekat auksina na antioksidantni status reznica višnje. Uzorci su uzimani nakon 0, 2, 4 i 6 dana. Kontrolu su činile reznice držane u vodi bez hormona. Intenzitet lipidne peroksidacije (LP) je meren kao produkcija malondialdehida (MDA), spektrofotometrijski na 532 nm, i izražen je u nmol MDA g-1 sveže mase.
Dobijeni rezultati su varirali u zavisnosti od podloge, organa biljke i primenjenih auksina. Utvrđeno je da je intenzitet LP u bazalnom delu bio najveći prvog dana, na- kon ozleđivanja, a da su svi ispitivani auksini snižavali produkciju MDA u narednim danima. Najbolji efekat pokazala je Magriva 1 tretirana sa NAA. U listovima, LP je bila najveća 2. dana, a najbolji efekat na smanjenje pokazali su primenjeni auksini u Stepskoj višnji (4. dana). Najveća produkcija MDA u listovima zabeležena je u Magrivoj 2 i Stepskoj višnji tretiranih sa IBA, 2. dana nakon odsecanja reznica (60-90% više od kontrole). Utvrđeno je da u većini podloga pod dejstvom ispitivanih hormona dolazi do smanjenja intenziteta LP što ukazuje na pozitivan efekat primenjenih auksina, ne samo na ožiljavanje reznica nego i na njihov antioksidantni sistem.

Cholic acid changes defense response to oxidative stress in soybean induced by Aspergillus niger

AB  - The oxidative stress and antioxidant systems in soybean leaves and roots infected with plant pathogen Aspergillus niger were studied following treatment with different concentrations of cholic acid. Several oxidative stress parameters were analyzed: production of superoxide (O-2 (center dot-)) and hydroxyl radicals ((OH)-O-center dot), lipid peroxidation (LP), and superoxide dismutase (SOD; EC 1.15.1.1) activity, as well as the content of reduced glutathione (GSH). Results showed that inoculation with A. niger led to the increase of O-2 (center dot-) production and GSH quantities in leaves and (OH)-O-center dot in roots. The highest activity of SOD occured in infected plants treated with cholic acid in concentrations of 40 and 60 mg L-1 which ultimately led to a decrease in O-2 (center dot-) production. Inoculation with Aspergillus in combination with elevated cholic acid concentrations also increased (OH)-O-center dot production which is correlated with increased LP. These results may support the idea of using cholic acid as an elicitor to trigger hypersensitive response in plant cells. Use of cholic acid may also actively contribute to soybean plants defense response against pathogen attack.

abstract = "The oxidative stress and antioxidant systems in soybean leaves and roots infected with plant pathogen Aspergillus niger were studied following treatment with different concentrations of cholic acid. Several oxidative stress parameters were analyzed: production of superoxide (O-2 (center dot-)) and hydroxyl radicals ((OH)-O-center dot), lipid peroxidation (LP), and superoxide dismutase (SOD; EC 1.15.1.1) activity, as well as the content of reduced glutathione (GSH). Results showed that inoculation with A. niger led to the increase of O-2 (center dot-) production and GSH quantities in leaves and (OH)-O-center dot in roots. The highest activity of SOD occured in infected plants treated with cholic acid in concentrations of 40 and 60 mg L-1 which ultimately led to a decrease in O-2 (center dot-) production. Inoculation with Aspergillus in combination with elevated cholic acid concentrations also increased (OH)-O-center dot production which is correlated with increased LP. These results may support the idea of using cholic acid as an elicitor to trigger hypersensitive response in plant cells. Use of cholic acid may also actively contribute to soybean plants defense response against pathogen attack.",

AB  - Changes in antioxidant systems in soybean [Glycine max (L) Merr., Fabaceae] genotypes infected with Sclerotinia sclerotiorum were studied 12, 24, 48 and 72 h after inoculation. Generation of superoxide and hydroxyl radicals was evaluated together with the production of malonyldialdehyde, main end product of lipid peroxidation. Several enzymatic and non-enzymatic parameters were monitored as well, such as the activity of antioxidant enzymes superoxide dismutase and pyrogallol and guaiacol peroxidases, reduced glutathione, soluble proteins and total carotenoids content. Results showed that genotypes expressed oxidative burst as well as different antioxidant systems in response to biotic stress caused by pathogen invasion. It has been confirmed that, although hypersensitive cell death is efficient against biotrophic pathogens, it does not protect soybean plants against infection by the necrotrophic pathogen such as S. sclerotiorum. Still, some genotypes showed distinctive and combined activity of several biochemical parameters which may point to further directions in exploring host-pathogen relations and lead to selection and production of new genotypes with higher levels of tolerance

Polyphenol contents and antioxidant activity of soybean seed extracts

AB  - The antioxidant activity and contents of various polyphenol classes in the seeds of 20 soybean hybrids were evaluated. Total polyphenols, tannins and proanthocyanidins were determined after extraction of plant material with 70% aqueous acetone. In addition, flavonoid content was determined. Antioxidant activity of seed extracts was evaluated by DPPH free radical scavenging activity assay. A positive linear correlation between antioxidant activity and contents of total phenols, tannins and proanthocyanidins was established. The highest antioxidant activity was observed in the extracts of hybrids which have higher levels of all polyphenol classes examined. The most of the single-cross hybrids were poor in tannins which recommend them as good source for ensiled livestock feed. Results suggested that polyphenol content should be considered as an important feature of the soybean seed.

AB  - The contents and antioxidant ability of various classes of phenolic compounds present in the seeds of twenty soybean hybrids were evaluated. Total phenolics, tannins and proanthocyanidins were determined spectrophotometrically, after extraction of seeds with 70% aqueous acetone. In addition, the flavonoid contents were determined. The antioxidant activity of aqueous acetone extracts was evaluated by the 1,1-diphenyl-2picrylhydrazyl (DPPH) free radical scavenging activity assay. The highest contents of total phenolics were found in Serbian cultivar 1511 and Chinese cultivar LN92-7369, which also displayed the highest total antioxidant activity. Conversely, genotypes poor in phenolics also showed low levels of DPPH-radical scavenging activity. The results suggested that besides protein and oil contents, the phenolic contents should be also considered as an important characteristic feature of soybean seeds, and as a potential selection criterion for antioxidant activity in soybean.

AB  - Tokom 1995. godine obavljena su poljska i laboratorijska istraživanja sa ciljem utvrđivanja međuzavisnosti aktivnosti enzima metabolizma azota na prinos zrna i sadržaj rastvorljivih proteina soje. Aktivnosti sva tri ispitivana enzima (NG, NR i GDH) bila su u negativhoj korelaciji sa prinosom zrna, i u pozitivnoj korelaciji sa sadržajem rastvorljivih proteina. Path-koeficijent analiza pokazala je da je direktan efekat aktivnosti NG pozitivan u odnosu na oba ispitivana svojstva, dok su direktni efekti NR i GDH negativni. Dobijeni rezultati pokazali su da aktivnost NG ima primarnu ulogu u procesima metabolizma azota u biljkama soje.
