AB  - As a part of the ECOBREED project, farmer participatory trials for organic soybean production were set up in Serbia, Germany, Romania, Austria and Slovenia (seasons 2021 and 2022). By setting up trials direct with organic producers, it was enabled that farmers assess the adaptability of the soybean varieties for organic production in specific agro-ecological conditions at their farm locations. This is one practical tool that enables organic producers to contribute and to be involved in the process of creating new soybean varieties, especially in crops composite population (CCPs) observations together with breeders. ECOBREED farmer participatory trials for organic soybean comprised field testings of soybean varieties from three breeding institutions from Serbia, Austria and Romania (Serbia-Institute of Field and Vegetable Crops, Austria-Saatzucht Gleisdorf, Romania-NARDI-Funduela). The network of organic participatory farms was established in 2021 at 17 locations in 5 European countries.

AB  - Leafy kale (Brassica oleracea var. acephala L.) is one of the most
important Brassicaceae species with health-promoting properties and great
diversity. To investigate variability within and among accessions, 12 accessions
(each accession comprised four individuals/genotypes) from the Balkan region
were genotyped at 12 simple sequence repeats (SSRs) loci. The selected SSR
markers originated from the genomes of B. oleracea (eight), B. rapa (two) and
B. napus (two). DNA was extracted from seeds, cotyledons, shoots, and young
leaves, depending on the germination energy of each seed, using magnetic
extraction method. Genetic analysis included eight accessions from Serbia (24
genotypes), two accessions from Montenegro (eight genotypes), and one
accession from Bosnia and Herzegovina and Croatia (with four genotypes each).
Accession-specific polymorphism was found at the Ol12-FO2 and Ol10-F11a
loci, where genotypes were qualitatively distinguished by horizontal agarose gel
electrophoresis (binary detection). Among 12 loci, the average polymorphic
information content (PIC) was 0.71, with the marker Ol13-C12 proving to be the

most informative (PIC = 0.89). However, the analysis of molecular variability
(AMOVA) showed 11% diversity between accessions, 19% diversity between
genotypes, and 70% diversity within genotypes, explaining the molecular
diversity of native Balkan leafy kale germplasm. According to the genetic
structure, four genetic groups were formed with an average expected
heterozygosity of 0.70 between clusters. In order to exploit the genetic diversity,
it would be advisable to evaluate these accessions at phenotypic level and use
their potential in breeding programmes.
AB  - Raštan (Brassica oleracea var. acephala L.) je jedna od najvažnijih vrsta iz porodice kupusnjača koja povoljno utiče na zdravlje ljudi i odlikuje se velikom raznovrsnošću. Da bi se istražila varijabilnost unutar i između prinova, 12 prinova (svaka prinova se sastojala od četiri individue/genotipa) sa teritorije Balkanskog poluostrva je genotipizovano koristeći 12 SSR markera. Odabrani SSR markeri potiču iz genoma B. oleracea (osam), B. rapa (dva) i B. napus (dva). DNK je izolovana iz sjemena, kotiledona, izdanaka i mladih listova, u zavisnosti od energije klijanja svakog sjemena, metodom magnetne ekstrakcije. Genetičkom analizom obuhvaćeno je osam prinova iz Srbije (24 genotipa), dvije prinove iz Crne Gore (osam genotipova) i jedna prinova iz Bosne i Hercegovine i Hrvatske (sa po četiri genotipa). Polimorfizam specifičan za prinove pronađen je na lokusima Ol12-FO2 i Ol10-F11a, gdje su genotipovi kvalitativno razlikovani horizontalnom elektroforezom u agaroznom gelu (binarna detekcija). Kod ukupno 12 lokusa, prosječan sadržaj polimorfne informacije (PIC) bio je 0,71, pri čemu se kao najinformativniji pokazao marker Ol13-C12 (PIC = 0,89). Međutim, analiza molekularne varijabilnosti (AMOVA) pokazala je 11% raznovrsnosti između prinova, 19% raznovrsnosti između genotipova i 70% raznovrsnosti unutar genotipova, objašnjavajući molekularnu raznovrsnost izvorne germplazme raštana sa teritorije Balkanskog poluostrva. AB  - Grass pea (Lathyrus sativus L.) is a self-pollinating legume that is well adapted to changing climatic conditions. In our study, the collection of 25 accessions from different parts of Europe (7 from BiH, 2 from Romania, 4 from Portugal, 8 from Serbia, 2 from Bulgaria, 1 from Greece and 1 from Slovenia) was genetically assessed by 15 species-specific nSSR markers (Simple Sequence Repeat). Extraction of gDNA was performed from 3 to 10 individual young plants/genotypes of each accession using an automated magnetic procedure. PCR reaction mixtures and amplification were optimized for each primer pair under two-step touch-down PCR conditions. Fragment analysis was performed on a genetic analyzer (ABI 3130XL) together with the internal standard ROX500. Accurate allele lengths were recorded from the electropherograms in GeneMapper6.0 software. The diversity parameters and genetic structure of the Lathyrus collection was observed using various population genetic programmes such as Arleqin, MSToolkit, GenAlEx, Populations, TreeView, and Structure.

AB  - Wheat and other cereals are important sources of dietary fibre and antioxidants. Most of the bioactive components are found in the outer layers and aleurone layers of the seed, but also the flour contains some. The major dietary fibre components in the wheat grain are cell wall polysaccharides such as arabinoxylan and β-glucan, which account for about 70% and 20%, respectively, of the total cell wall polysaccharides in the starchy endosperm. Barley and oats are rich in β-glucan, while wheat and rye are richer in arabinoxylan. Arabinoxylan has two forms, i.e. water-soluble (WE) and insoluble (WU) fractions, which differ in their health benefits. Antioxidants (e.g., alkylresorcinol) delay or inhibit oxidation processes. Alkylresorcinol is formed in the organism and acts against free radicals, which are responsible for the cell degradation.

AB  - Grass pea (Lathyrus sativus L.) is a legume with high protein content and good quantities of essential amino acids. It has high tolerance to drought, high adaptability to extreme conditions, disease resistance and low input requirement for cultivation, which makes it very useful for cultivation by resource-poor farmers. Potential usage of grass pea is limited due to the presence of anti-nutritional factors including protease inhibitors, especially trypsin inhibitors. They have a major impact on nutritional value, by inhibiting pancreatic serine proteases (thus impairing protein digestion), but are also likely to protect legume seed against attack by predators. Although once extensively grown throughout Balkan Peninsula, grass pea was nearly forgotten. In recent years there was an effort to reintroduce and increase the interest of researchers and farmers in this crop. The aim of this study was to establish whether genotype and environment have significant influences on trypsin inhibitor activity (TIA) in grass pea and to determine whether TIA is correlated with seed protein content.

AB  - Family Brassicaceae is abundant in species used in food production, but also contains many industrial species, vegetables and ornamentals. Green collard (Brassica oleracea var. acephala L.) was traditionally grown in Balkan Peninsula and its wide green leaves, rich in vitamins A and C, are used for cooking. There is great diversity in B. oleracea acephala germplasm that reflects on agronomic, morphological and molecular characteristics. In this study, was assessed genetic variation of 38 accessions of green collard out of which 3 accessions were standard/commercial varieties and 35 were locally grown populations, collected in Western Balkan: 18 from Bosnia and Herzegovina, 8 from Serbia (Vojvodina), 7 from Croatia and two from Montenegro. Genetic diversity was assessed using twelve species-specific SSR markers (Simple Sequence Repeats) originating from B. oleracea (8), B. rapa (2) and B. napus (2). Each accession was represented with four individual samples.

AB  - Genetic diversity and structure of common bean (Phaseolus vulgaris L.) germplasm from Serbia, comprising 118 landraces and 18 cultivars, was assessed with the application of 27 Single Sequence Repeats (SSR) markers. Thirteen accessions from Agricultural Institute of Slovenia were used as references for gene pool determination. Main parameters of genetic diversity were calculated for each SSR loci, i.e. number of different and rare alleles, number of effective alleles, Shannon’s information index, observed and expected heterozygosity and polymorphic information content. A total of 445 allelic variants, with 16.5 alleles per locus on average, were detected. Mean gene diversity (He = 0.79) indicated sufficient reservoir of genetic variation preserved in studied bean germplasm. Landraces displayed higher variability compared to cultivars (405 in relation to 233 allelic variants). Genetic structure and relatedness of accessions was assessed by model-based method and hierarchical clustering method in combination with genetic distance calculation. The Bayesian clustering model implemented in STRUCTURE software, on the primary level (K = 2), revealed clear separation of accessions into two groups, corresponding to gene pool affiliation. Mesoamerican gene pool (M) was represented with 23.5% of accessions, while Andean (A) was larger, composed of 68.4% of studied germplasm. Small group (8.1%) showed admixed genetic structure between two gene pools. Additional variation in respect to two recognized gene pools was revealed (K = 3), whose basis was acknowledged to be within Andean gene pool. Further subdivision of accessions (K = 8), mainly according to the seed forms, was observed. Genetic distance analysis associated with Neighbour-joining clustering method revealed grouping pattern of landraces and cultivars corresponding to the gene pool and their seed phenotypes. Classification and structuring of the bean accessions according to and beyond the gene pool of origin should facilitate conservation strategies and breeding of this material. Combining the information of phenotypic variation obtained in previous research and molecular data reveled in this study will assist in selection of parental components for breeding, or in the choice of smaller sample in order to further acknowledge their breeding value. In addition, obtained results of this work should serve as an additional information on common bean germplasm variation in Western Balkans and beyond, in Europe.

AB  - Genetic diversity and structure of common bean (Phaseolus vulgaris L.) germplasm from Serbia, comprising 118 landraces and 18 cultivars, was assessed with the application of 27 Single Sequence Repeats (SSR) markers. Thirteen accessions from Agricultural Institute of Slovenia were used as references for gene pool determination. Main parameters of genetic diversity were calculated for each SSR loci, i.e. number of different and rare alleles, number of effective alleles, Shannon’s information index, observed and expected heterozygosity and polymorphic information content. A total of 445 allelic variants, with 16.5 alleles per locus on average, were detected. Mean gene diversity (He = 0.79) indicated sufficient reservoir of genetic variation preserved in studied bean germplasm. Landraces displayed higher variability compared to cultivars (405 in relation to 233 allelic variants). Genetic structure and relatedness of accessions was assessed by model-based method and hierarchical clustering method in combination with genetic distance calculation. The Bayesian clustering model implemented in STRUCTURE software, on the primary level (K = 2), revealed clear separation of accessions into two groups, corresponding to gene pool affiliation. Mesoamerican gene pool (M) was represented with 23.5% of accessions, while Andean (A) was larger, composed of 68.4% of studied germplasm. Small group (8.1%) showed admixed genetic structure between two gene pools. Additional variation in respect to two recognized gene pools was revealed (K = 3), whose basis was acknowledged to be within Andean gene pool. Further subdivision of accessions (K = 8), mainly according to the seed forms, was observed. Genetic distance analysis associated with Neighbour-joining clustering method revealed grouping pattern of landraces and cultivars corresponding to the gene pool and their seed phenotypes. Classification and structuring of the bean accessions according to and beyond the gene pool of origin should facilitate conservation strategies and breeding of this material. Combining the information of phenotypic variation obtained in previous research and molecular data reveled in this study will assist in selection of parental components for breeding, or in the choice of smaller sample in order to further acknowledge their breeding value. In addition, obtained results of this work should serve as an additional information on common bean germplasm variation in Western Balkans and beyond, in Europe.

AB  - Grass pea (Lathyrus sativus L.) is a less common and nearly forgotten crop in many
countries, including Slovenia and Serbia. In the present study, genetic analysis and
seed characteristics of the collection of 22 grass pea accessions from several
Southeast European plant gene banks were examined. The collection was produced
in an open-field experiment at two countries/locations, i.e. Slovenia/Ljubljana –
Jablje and Serbia/Novi Sad – Rimski Šančevi, during the 2019 growing season. For
analysis of genetic diversity, specific high polymorphic SSR markers were applied.
Initial screening at 12 species-specific SSR loci using binary data revealed a great
genetic diversity among the grass pea accessions within a collection and three genetic
groups were formed. Differences in seed size were measured using descriptors for
seed length, width and thickness, and 100-seed weight. Furthermore, several seed
colour characters using IBPGR descriptors were visually assessed. The average seed
length, width and thickness of the seeds in the collection were 9.0 mm, 8.3 mm and
5.4 mm, respectively, while 100-seed weight ranged considerably from 11.7 g to
38.9 g. The highest differences between growing location were seen for 100-seed
weight where the coefficient of variability reached 23.5%. The examined grass pea
accessions had less monochromatic (one colour) and more colourful seeds, with two
or three colours.

AB  - The ECPGR (European Cooperative Programme for Plant Genetic Resources) Grain legumes working group cover a range of species, which have enormous benefits for environment, food security and as well as the breeding material. However, despite its huge potential lots of species, including Lathyrus sp., are not well represented in European collection and enough exploited for different purposes. As starting point of EUGrainLeg project the activities act in two directions: i) AEGIS progress and improved data on European grain legumes plant genetic resources, and ii) improvement of group strategy work – in view of subgroup establishment based on species interest, in order to facilitate experience and knowledge exchange. The task of establishing the European grain legume collection needs updates and improvements, taking in account the large number of species managed inside the group. The significant diversity of Lathyrus sp. exist, but there are difficulties/gaps in order to identify accessions as AEGIS accessions, recording to registered data.

AB  - In the last decade several thousand accessions were collected in different parts of Europe and are stored in national gene banks; at present the Phaseolus Database as part of the EURISCO Web catalogue contains over 46000 records. Main objective of the study (ECPGR_SMARTLEG project) was proper phenotypic and genetic evaluation of European Phaseolus accessions with emphasis to Phaseolus coccineus germplasm providing new data to EURISCO and AEGIS. During the project, we provided three different types of datasets including morpho-agronomic traits of plants, morphometric seed characteristics and genetic profiles of P. coccineus accessions from eight European geographic origins; Slovenia, Romania, Bosnia and Herzegovina, Serbia, Italy, Slovakia, Macedonia and Austria. Before sowing, the morphometric characterization of P. coccineus seeds was performed according to adopted Community Plant Variety Office-Technical Protocol and Phaselieu/AIS descriptors observing 14 different seed characteristics.

abstract = "In the last decade several thousand accessions were collected in different parts of Europe and are stored in national gene banks; at present the Phaseolus Database as part of the EURISCO Web catalogue contains aver 46000 records. Main objective of the study (ECPGR_SMARTLEG project) was proper phenotypic and genetic evaluation of European Phaseolus accessions with emphasis to Phaseolus coccineus germplasm providing new data to EURISCO and AEGIS. During the project, we provided three different types of datasets including morpho-agronomic traits of plants, morphometric seed characteristics and genetic profiles of P. coccineus accessions from eight European geographic origins; Slovenia, Romania, Bosnia and Herzegovina, Serbia, Italy, Slovakia, Macedonia and Austria.",

Genetic diversity of the autochtonous Phaseolus bean germplasm originating from five South east European countries

AB  - Genetic differentiation of 167 Phaseolus vulgaris L. accessions originating from five South east European countries was performed using 13 SSR markers. Analysis included 21 accessions from Bosnia and Herzegovina, 15 accessions from Croatia, 73 accessions from Macedonia, 38 accessions from Serbia and 20 accessions from Slovenia. Calculated mean number of alleles per locus was 9.8. The average polymorphic information content over all loci reached value of 0.710 where the most informative locus was GATS91 (0.926). These results showed that selected set of SSR markers are highly informative and applicable for studies of genetic diversity within germplasm collected in the five South east European countries, which was also confirmed with the calculated value of probability of identity (1.831x10). High mean value of expected heterozygosity (0.731) and Shannon’s information index (1.486) reflects high genetic diversity within accessions from five countries, where accessions from Macedonia are genetically the most uniform, due to 17completely identical accessions included.
