AB  - Investigation about effect of high amounts of nitrogen and source of nutrition on winter triticale productivity was carried out in a stationary field trial in the Sumadija region, in Central Serbia. This study was conducted over a period of three years (2009-2011) on a vertisol soil to evaluate the effect of different fertilization methods on grain yield and yield components of winter triticale variety. Treatments included control and six fertilization variants with different combinations of NPK fertilizers (N, NK, NP1, NP2, NP1K, and NP2K). Investigation showed a considerable variation of grain yield depending on mineral nutrition. The highest and statistically significant average grain yields were achieved under NP1K (4.105 t ha(-1)) and NP2K (4.050 t ha(-1)) treatments. The combined use of NP1K fertilizers (120 kg N ha(-1), 60 kg P2O5 ha(-1), 60 kg K2O ha(-1)) and NP2K treatment (120 kg N ha(-1), 100 kg P2O5 ha(-1), 60 kg K2O ha(-1)) represented the excellent base for optimum stock of major nutrients, resulting in maximum grain yield. Thousand grain weight of triticale significantly varied across years, the highest average 1000-grain weight of winter triticale investigated was achieved in the NP1K variant (44.81 g). Grain yield was significantly positively correlated with 1000-grain weight only in all years.

Comparative analysis of laboratory and field procedures for testing genetic purity of sunflower parent lines

AB  - One of the main features that confer high quality to the seed is its genetic purity. Genetic impurities in sunflower seed lot can originate from seed due to self-pollination, uncontrolled cross-pollination or mechanical mixing of seeds. Genetic purity of 18 seed lots of sunflower parental lines is controlled by comparative analysis of morphological traits and electrophoretic analysis 4 isoenzyme systems, according to the UPOV guidelines for DUS test of sunflower lines and hybrids. In 83.3% of the total analysed samples, results in the field and laboratory were in agreement. In parental line R-8 5% genetic impurity was found by isoenzyme analysis, which was not detected based on morphological characteristics, while electrophoretic analysis of samples L-10 and L-11 showed less genetic impurities in comparison with the analysis in the field.
AB  - Jedna od važnih komponenti kvaliteta semena jeste njegova sortna, odnosno genetska čistoća. Uzroci nesortnih primesa mogu poticati od semena nastalog usled nekontrolisane samooplodnje, stranooplodnje ili usled mehaničkog mešanja semena. Genetska čistoća 18 partija roditeljskih linija suncokreta kontrolisana je uporednom analizom morfoloških osobina i elektroforetskom analizom 4 izoenzimska sistema, prema UPOV uputstvu za DUS analizu linija i hibrida suncokreta. U 83,3% od ukupno ispitanih uzoraka, rezultati u polju i laboratoriji su bili u saglasnosti. Kod roditeljske linije R-8 analizom izoenzima pronađeno je 5% genetske nečistoće koja nije konstatovana na osnovu morfoloških karakteristika, dok je elektroforetska analiza uzoraka L-10 i L-11 pokazala manju genetsku nečistoću u poređenju sa analizom u polju.

AB  - Genetically modified soybean was explored in food samples, labeled as "non-GM" and unlabeled, using multiplex PCR and Real-time PCR techniques. In total 110 (2006) and 237 food samples (2007) containing soybean were analyzed. The 7% of the products were shown to consist of material derived from the Roundup Ready soybean in 2006 and 13% of the foods samples in 2007. The most of analyzed samples were correctly labeled, but ten of food products are demonstrated to contain material above the threshold levels for labeling of 0.9%. This is the first report of the validity of labels on imported food samples from different countries and uptake of GM food derived from soybean into the food chain in Serbia.

Genetic purity of sunflower hybrids determined on the basis of isozymes and seed storage proteins

AB  - Maintenance of genetic uniformity of lines and hybrids is a prerequisite for successful production and placement of commercial hybrid seed on the market. Genetic purity of seed as specific seed trait is of great significance for seed science. Protein markers, seed storage proteins and isozymes, which are commonly used for the estimation of genetic purity, were used in this work to estimate genetic purity in sunflower hybrids. Analysis of helianthinin revealed tree zymogram patterns within and between sixteen sunflower hybrids. All of the 6 enzymatic systems analyzed, MDH, PGM, PHI, PGD, IDH and ACP, were polymorphic. A comparative analysis of genetic purity level of the sunflower hybrids was performed using electrophoretic methods. The methods of electrophoretic separation of isozymes and seed storage proteins were in agreement, with differences ranging from 1% to 5% in 81% of the samples. The level of polymorphism obtained by both methods was not distinct enough to be used in genotype identification.

Detection of triplex PCR for the modified qualitative soybean and maize genetically

AB  - A molecular screening method based on multiplex PCR that involves amplification of specific soybean or maize sequences from plant DNA (lectin or zein) and the amplification of 35S promoter and NOS terminator,for the detection of genetically modified soybean and maize was developed.
