Correlation between phenolic content and antioxidant activity in fruits of Prunus avium infected with Monilia laxa

AB  - Accumulation of phenolic polymers reinforce plant cell walls, limiting fungal penetration. Some of these
compounds, such as isochlorogenic acid, are particularly active in limiting fungal extension. Two main features of
phenolic compounds are that they are produced in greater quantities by resistant plant genotypes, and that they
have the ability to limit plant pathogens growth. Thus, they could be considered as potential markers of resistance
to Monilinia laxa in sweet cherrie breeding programs.
We investigated the content of the total phenols (TP), total tannins (TT) and anthocyanins (A) in the fruits of ten
sweet cherrie cultivars, before and after the infection with M. laxa, under the field conditions. In addition to this,
we determined the total antioxidant capacity of the fruit extracts using the DPPH-free radical scavenging assay and
FRAP method. The correlations between phenolic classes and antioxidant activity have been calculated, as well.
Results showed that the TP content ranged from 3.71 to 7.52 mg gallic acid equivalent (GAE) g-1 dry weight
depending on the genotype and the infection. TT content was lower and it ranged from 0.52 to 2.64 mg GAE g-1 dry
weight. Anthocyanins content ranged from 0.29 to 2.71 mg cyanidin-3-glucoside g-1 dry weight. DPPH values
ranged from 38.47 to 63.50 % of neutralised radicals, while FRAPvalues were 14.73-33.69 mg trolox g-1 fresh
weight. No positive correlation was found for antioxidant tests and phenolic compounds investigated. Still, TP and
TT were higher in infected fruits of five specimen (Junska rana, Summit, Mershant, Priusodebnaja and Burlat), and
A levels were higher in the same cultivars except for the cv. Summit. These cultivars accumulate defensive
compounds under the pathogen induced stress and may be considered as more tolerant genotypes with more
desired traits for production, selection and breeding programmes.

Changes in superoxide-dismutase (SOD) activity in auxin treated Prunus sp. softwood cuttings under oxidative stress

AB  - Oxidative metabolism of normal cells and different stress situations generate highly reactive oxygen
species (ROS). The ROS have been implicated in a number of physiological disorders in plants. Such factors as certain mechanical injuries are known to induce ROS formation in most aerobic organisms. Antioxidant defence systems have co-evolved with aerobic metabolism to counteract oxidative damage from ROS. This includes antioxidant enzymes such as superoxide-dismutase (SOD; EC 1.15.1.1). SOD is of the greatest importance since it catalyzes the dismutation of the superoxide radical (O2.-) to O2 and H2O. Thus, the aim of this study was to measure the intensity of SOD in the leaves of six cherry rootstock selections in order to investigate the effect of exogenously applied auxins on the mechanical injury induced oxidative stress during the process of rooting of softwood cuttings.
The experiment was carried out in the plastic house under a fogging system with 95-99% average relative
humidity. The 15-20 cm long terminal cuttings were treated with the mix of auxins (IBA, 0.5% and α-NAA, 0.8%)
powder prior to insertion into the white sphagnum and perlite substrate mixture. Leaves of six promising rootstock selections of Prunus cerasus L., P. mahaleb L. and P. fruticosa Pall., and one standard rootstock PHL-A were sampled on the 1st, 3rd and 7th day after inserting softwood cuttings into the rooting substrate. The SOD activity was measured by monitoring the inhibition of nitroblue-tetrazolium (NBT) reduction at 560 nm.
Results showed that generation of ROS occurred in all investigated genotypes, but they greatly differed in
susceptibility towards oxidative stress induces by mechanical injury. Application of auxins on standard vegetative rootstock PHL-A, as well as P. mahaleb selection M6 showed positive effect on lowering SOD activity after the 3rd and 7th collecting day compared to control. On the other hand, selections of P. cerasus OV22 and P. fruticosa SV2 expressed high SOD activity during all collecting days which was from 3.6-54.9% higher compared to control.
Enhanced SOD activity in leaves of softwood cuttings indicates induction of the enzymatic antioxidant system in
both auxin treated and untreated cutting. According to these results SOD activity could be used as one of the
biochemical parameters in further rootstock selection for sweet and sour cherries.

AB  - The effect of exogenously applied auxins on mechanical injury induced oxidative stress was studied in 6 promising rootstock selections of Prunus cerasus L., P. mahaleb L., and P. fruticosa Pall. Investigated selections were included in low vigorous rootstock breeding programme for sweet and sour cherries. The standard rootstock PHL-A was used as a control, due to successful rooting. Leaves of investigated rootstocks were collected on: 0, 1st, 3rd and 7th day of inserting softwood cuttings into the rooting substrate under a fogging system with 95-99% average relative humidity. Exogenously applied auxin mixture consisted of 0.8% α-naphthylacetic acid (NAA) and 0.5% indolebutyric acid (IBA). Mechanical injuries during rooting period represent one of the factors that induce stress in softwood cuttings. Intensity of LP is used as a valuable biomarker of plant response to various abiotic factors. Differences in intensity of LP between auxin-treated and -untreated cuttings were examined. Almost all of investigated selections had lower LP intensity after auxin application (11.4-47.2%) between 1st and 3rd day. However, the most prominent change was in leaves of PHL-A (49.2%). The best LP-lowering effect were recorded in leaves of P. fruticosa, SV4 selection (56.9%) on 7th day and in P. mahaleb, M4 selection, on 1st and 3rd day (5.9, 5.8%), in comparison to untreated softwood cuttings. As for OV21 selection, LP intensity significantly increased in both treated and untreated cuttings on the 1st, 3rd and 7th day, but auxin-treated cuttings showed lower LP values, except on 7th day, reaching 221.3 nmol MDA equivalents g-1 fresh weight. Intensity of LP during vegetative propagation of selected genotypes could be used as one of the biochemical parameters in further rootstock selection for sweet and sour cherries.

AB  - In order to investigate the effect of phytohormones exogenous auxins on the level of the mechanical injury induced oxidative stress, intensity of lipid peroxidation (LP) was measured in rootstocks of cherry softwood cuttings. Basal part of the cuttings and leaves of five rootstocks (Mahaleb 1 and 2, Gisela 5, European ground cherry (EGC) and “Oblačinska” sour cherry) were sampled 0, 2, 4 and 6 days after cutting. Cuttings were treated with 0.5% solutions of three auxins: α-naphthylacetic acid (NAA), indolebutyric acid (IBA) and combination of these two (INCIT K). Results obtained for the LP intensity varied depending on the rootstock, plant organ and auxin applied. The best LP-lowering effect of auxins occurred in all genotypes when treated with NAA. In the leaves, the LP peaked on the 2nd day, while the best LP-lowering effect was recorded in European ground cherry rootstocks on the 4th day, independently of auxin applied. The highest MDA production in leaves was recorded in Mahaleb 2 and European ground cherry rootstocks treated with IBA (60-90% higher than control) two days after the cuttings were made. It has been established that in the most of the rootstocks examined auxins showed lowering effect on LP which points to their positive effect, not only on rooting of softwood cuttings, but on their antioxidant protection system, as well.
AB  - U radu je ispitan nivo oksidativnog stresa izazvanog mehaničkom povredom biljke prilikom proizvodnje reznica, u bazalnom delu i listovima pet podloga za višnju i trešnju (Magriva 1 i 2, Gizela 5, Stepska (EGC) i “Oblačinska” višnja). Istovremeno, reznice su tretirane 0.5% rastvorima tri auksina (α-naftilsirćetna kiselina, NAA, indol- buterna kiselina, IBA i kombinacija ovih hormona, INCIT K), u periodu od 60 min, kako bi se utvrdio efekat auksina na antioksidantni status reznica višnje. Uzorci su uzimani nakon 0, 2, 4 i 6 dana. Kontrolu su činile reznice držane u vodi bez hormona. Intenzitet lipidne peroksidacije (LP) je meren kao produkcija malondialdehida (MDA), spektrofotometrijski na 532 nm, i izražen je u nmol MDA g-1 sveže mase.
Dobijeni rezultati su varirali u zavisnosti od podloge, organa biljke i primenjenih auksina. Utvrđeno je da je intenzitet LP u bazalnom delu bio najveći prvog dana, na- kon ozleđivanja, a da su svi ispitivani auksini snižavali produkciju MDA u narednim danima. Najbolji efekat pokazala je Magriva 1 tretirana sa NAA. U listovima, LP je bila najveća 2. dana, a najbolji efekat na smanjenje pokazali su primenjeni auksini u Stepskoj višnji (4. dana). Najveća produkcija MDA u listovima zabeležena je u Magrivoj 2 i Stepskoj višnji tretiranih sa IBA, 2. dana nakon odsecanja reznica (60-90% više od kontrole). Utvrđeno je da u većini podloga pod dejstvom ispitivanih hormona dolazi do smanjenja intenziteta LP što ukazuje na pozitivan efekat primenjenih auksina, ne samo na ožiljavanje reznica nego i na njihov antioksidantni sistem.

abstract = "In order to investigate the effect of phytohormones exogenous auxins on the level of the mechanical injury induced oxidative stress, intensity of lipid peroxidation (LP) was measured in rootstocks of cherry softwood cuttings. Basal part of the cuttings and leaves of five rootstocks (Mahaleb 1 and 2, Gisela 5, European ground cherry (EGC) and “Oblačinska” sour cherry) were sampled 0, 2, 4 and 6 days after cutting. Cuttings were treated with 0.5% solutions of three auxins: α-naphthylacetic acid (NAA), indolebutyric acid (IBA) and combination of these two (INCIT K). Results obtained for the LP intensity varied depending on the rootstock, plant organ and auxin applied. The best LP-lowering effect of auxins occurred in all genotypes when treated with NAA. In the leaves, the LP peaked on the 2nd day, while the best LP-lowering effect was recorded in European ground cherry rootstocks on the 4th day, independently of auxin applied. The highest MDA production in leaves was recorded in Mahaleb 2 and European ground cherry rootstocks treated with IBA (60-90% higher than control) two days after the cuttings were made. It has been established that in the most of the rootstocks examined auxins showed lowering effect on LP which points to their positive effect, not only on rooting of softwood cuttings, but on their antioxidant protection system, as well., U radu je ispitan nivo oksidativnog stresa izazvanog mehaničkom povredom biljke prilikom proizvodnje reznica, u bazalnom delu i listovima pet podloga za višnju i trešnju (Magriva 1 i 2, Gizela 5, Stepska (EGC) i “Oblačinska” višnja). Istovremeno, reznice su tretirane 0.5% rastvorima tri auksina (α-naftilsirćetna kiselina, NAA, indol- buterna kiselina, IBA i kombinacija ovih hormona, INCIT K), u periodu od 60 min, kako bi se utvrdio efekat auksina na antioksidantni status reznica višnje. Uzorci su uzimani nakon 0, 2, 4 i 6 dana. Kontrolu su činile reznice držane u vodi bez hormona. Intenzitet lipidne peroksidacije (LP) je meren kao produkcija malondialdehida (MDA), spektrofotometrijski na 532 nm, i izražen je u nmol MDA g-1 sveže mase.
Dobijeni rezultati su varirali u zavisnosti od podloge, organa biljke i primenjenih auksina. Utvrđeno je da je intenzitet LP u bazalnom delu bio najveći prvog dana, na- kon ozleđivanja, a da su svi ispitivani auksini snižavali produkciju MDA u narednim danima. Najbolji efekat pokazala je Magriva 1 tretirana sa NAA. U listovima, LP je bila najveća 2. dana, a najbolji efekat na smanjenje pokazali su primenjeni auksini u Stepskoj višnji (4. dana). Najveća produkcija MDA u listovima zabeležena je u Magrivoj 2 i Stepskoj višnji tretiranih sa IBA, 2. dana nakon odsecanja reznica (60-90% više od kontrole). Utvrđeno je da u većini podloga pod dejstvom ispitivanih hormona dolazi do smanjenja intenziteta LP što ukazuje na pozitivan efekat primenjenih auksina, ne samo na ožiljavanje reznica nego i na njihov antioksidantni sistem.",

Estimation of the Genetic Diversity in Tetraploid Alfalfa Populations Based on RAPD Markers for Breeding Purposes

AB  - Alfalfa is an autotetraploid, allogamous and heterozygous forage legume, whose varieties are synthetic populations. Due to the complex nature of the species, information about genetic diversity of germplasm used in any alfalfa breeding program is most beneficial. The genetic diversity of five alfalfa varieties, involved in progeny tests at Institute of Field and Vegetable Crops, was characterized based on RAPD markers. A total of 60 primers were screened, out of which 17 were selected for the analysis of genetic diversity. A total of 156 polymorphic bands were generated, with 10.6 bands per primer. Number and percentage of polymorphic loci, effective number of alleles, expected heterozygosity and Shannon's information index were used to estimate genetic variation. Variety Zuzana had the highest values for all tested parameters, exhibiting the highest level of variation, whereas variety RSI 20 exhibited the lowest. Analysis of molecular variance (AMOVA) showed that 88.39% of the total genetic variation was attributed to intra-varietal variance. The cluster analysis for individual samples and varieties revealed differences in their population structures: variety Zuzana showed a very high level of genetic variation, Banat and Ghareh were divided in subpopulations, while Pecy and RSI 20 were relatively uniform. Ways of exploiting the investigated germplasm in the breeding programs are suggested in this paper, depending on their population structure and diversity. The RAPD analysis shows potential to be applied in analysis of parental populations in semi-hybrid alfalfa breeding program in both, development of new homogenous germplasm, and identification of promising, complementary germplasm.

AB  - Although alfalfa is the most important forage crop grown in the temperate regions, breeding improvements are not easily achieved due to high genetic complexity of this species. Because of allogamy and tetraploidy, cultivated alfalfa synthetic varieties have a high degree of genetic diversity among individual plants in populations. The knowledge of the extent of genomic variability in cultivars bearing desirable agronomic traits is an area that could benefit from molecular genetic analyses. Therefore, the aim of the present work was to evaluate the ability of RAPD markers to detect intra-population variation in fi ve alfalfa varieties. Presented here are the first results of investigating 40 randomly selected RAPD primers in order to establish whether they were able to synthesize PCR products, which would enable detection of polymorphism between the selected varieties and inside them as well.
AB  - Iako je lucerka najznačajnija gajena biljka za stočnu ishranu u umerenim klimatskim regionima, postizanje značajnih napredaka u oplemenjivanju je veoma komplikovano zbog njene složene genetske konstitucije. Usled alogamije i tetraploidije sintetičke sorte lucerke se karakterišu veoma izraženom varijabilnošću individua unutar same sorte. Poznavanje stepena genetske varijabilnosti sorti i populacija sa poželjnim agronomskim svojstvima moglo bi značajno biti unapređeno upotrebom molekularne genetske analize. Stoga je cilj ovog rada bilo ispitivanje mogućnosti RAPD markera da detektuju varijacije unutar populacije kod pet sorti lucerke različitog porekla. Predstavljeni su rezultati početnog ispitivanja 40 slučajnih RAPD prajmera u cilju ispitivanja njihovih mogućnosti da sintetišu PCR produkte koji bi omogućili kako detekciju polimorfizma između ispitivanih sorti, tako i unutar njih.
