Detection of Xanthomonas axonopodis pv. phaseoli and Pseudomonas savastanoi pv. phaseolicola on bean seed using a Milk-tween medium

AB  - Bean production is threaten by phytopathogenic bacteria causing agents of blights, Xanthomonas axonopodis pv. phaseoli (Xap) and Pseudomonas savastanoi pv. phaseolicola (Psp). Since there is no satisfactory chemical control for the disease, the recommended measures are preventive and include use of healthy seed, crop rotation, deep plowing and use of resistant cultivars. In this work we involved a detection method for isolation of Xap and Psp from bean seed to semi-selective medium Milk Agar Tween (MT). On this medium, Xap formed yellow, mucoid and convex colonies with two hydrolysis zones (less milk and more enlightened), and Psp formed whitish-cream, flat and round colonies. The identification of Xap and Psp was confirmed using the ELISA and PCR. Due to its selectivity, easy preparation and possibility of simultaneous detection of bacteria Xap and Psp, MT medium can be recommended for routine test of seed health for local seed or seed from import.
AB  - Proizvodnju pasulja ugrožavaju fitopatogene bakterije prouzrokovači plamenjača Xanthomonas axonopodis pv. phaseoli (Xap) i Pseudomonas savastanoi pv. phaseolicola (Psp). S obzirom da do sada nije postignuta zadovoljavajuća hemijska zaštita za suzbijanje ovih bakterija, mere koje se preporučuju preventivnog su karaktera i uključuju upotrebu zdravog semena za setvu, plodored, duboko zaoravanje ostataka i korišćenje otpornih sorti. U ovom radu je vršena detekcija Xap i Psp na semenu pasulja metodom izolacije na poluselektivnu podlogu Milk Tween Agar (MT). Na ovoj podlozi Xap obrazuje žute, sluzaste i ispupčene kolonije sa dve zone hidrolize (manjim mlečnim i većim prosvetljenim), a Psp obrazuje kolonije beličasto-krem, ravne i okrugle. Identifikacija izolata Xap i Psp je potvrđena primenom ELISA i PCR. Podloga MT se zbog svoje selektivnosti, jednostavne pripreme i mogućnosti istovremene detekcije bakterija Xap i Psp može preporučiti prilikom rutinskog ispitivanja zdravstvenog stanja semena pasulja za setvu proizvedenog kod nas ili prilikom kontrole semena iz uvoza.

Response of different beans against common bacterial blight disease caused by xanthomonas axonopodis pv. Phaseoli

AB  - Common bacterial blight (CBB) in beans, caused by the bacterium Xanthomonas axonopodis pv. phaseoli (Xap), is an economically important disease worldwide which reduces crop yields and seed quality. Since there is no satisfactory chemical control for the disease, the use of resistant cultivars is an important management strategy. Sources of immunity are not yet recognized, but tolerance has been reported in several genetic stocks. The main objective of this study was to determine resistance to CBB on twenty-two local and foreign beans grown in Serbia. Two inoculation methods with Xap (spraying and multiple needles) as well as naturally infected plants in field conditions were taken for evaluation. The experiments were conducted in randomized complete blocks with three replications. Reaction to Xap was assessed as a diseased leaf area and the disease severity index was calculated. The analysis of variance (ANOVA) for disease ratings of leaf reactions indicated significant interactions between cultivars and lines. Results indicate that none of evaluated beans was immune and was found to be resistant to CBB. HR 45, Oreol and XAN 159, -208, -273 were weakly susceptible; Biser, Dobrudzanski rani, KB 100, -101, Medijana, Naya Nayahit, Panonski gradistanac, Panonski tetovac, and Sremac were susceptible while Balkan, Belko, Dobrudzanski rani 7, Dvadesetica, Galeb, Maksa, Slavonski zutozeleni, and Zlatko were highly susceptible cultivars and lines. The five weakly susceptible can be identified and recommend as possible sources of tolerance in plant breeding program.

AB  - Laboratory assays were conducted to detect and identify Xanthomonas axonopodis pv. phaseoli on bean seeds collected from naturally contaminated commercial crops. Semi-selective media, viz. Xanthomonas campestris pv. phaseoli agar and milk tween agar were used to detect the bacterium from whole bean seed extract. Colonies of the bacterium were yellow, mucoid and convex on these media and zones of hydrolysis formed around them. The pathogenicity of the strains was confirmed by stem inoculation of young bean plants. Identification of the strains was proved by using rapid methods (ELISA and PCR). The results showed the presence of seed-borne X. axonopodis pv. phaseoli in 20 out of the 23 assayed bean cultivars that are commercially grown in Serbia. These results indicate that the seed of the bean cultivars grown in our country is highly infected by the bacterium.
