AB  - Waitea circinata var. zeae (anamorf Rhizoctonia zeae) je značajan patogen uglavnom onokotiledonih biljaka, koji je prisutan u mnogim delovima sveta, a održava se u zemljištu duži niz godina. Poslednjih godina krug domaćina W. circinata var. zeae širi se i na neke dikotiledone biljke. U okviru nedavnih istraživanja u Srbiji W. circinata var. zeae je utvrđen kao prouzrokovač truleži korena i korenovog vrata, i propadanja biljaka kupusa i uljane repice na lokalitetima Futog i Rimski Šančevi. U radu su sistematizovani publikovani podaci o rasprostranjenosti, krugu domaćina, načinu održavanja i širenja, kao i suzbijanju W. circinata var. zeae, kao i rezultati istraživanja koja su obavljena u Srbiji.

Biocontrol of Botrytis cinerea and promotion of tomato growth by local soil-borne Bacillus isolates

AB  - Grey mould, which is caused by Botrytis cinerea Pers., is among the most damaging diseases of cultivated plants worldwide. The use of fungicides dominates in the protection of tomatoes against grey mould. Due to the emergence of resistance of pathogens to the active ingredients of fungicides, the possibility of using bacteria of the genus Bacillus was studied. A total of 666 isolates were isolated from different agricultural soils, and a preliminary screening dual culture
test with Alternaria dauci was done. A total of 77 isolates were dual culture tested against Fusarium tricinctum and F. proliferatum. Based on preliminary dual culture testing, eight Bacillus isolates were selected for testing their biological potential against two isolates of B. cinerea. The effect of Bacillus spp. isolates was studied for seed germination and plant growth promotion (PGP) on the seedlings of tomato (Solanum lycopersicum L.). Using molecular identification (16S rDNA), five bacterial isolates were identified as Bacillus subtilis, two isolates as B. amyloliquefaciens, and one
isolate as B. pumilus. Using the Internal transcribed spacer (ITS) region, two fungal isolates from an infected leaf and a tomato fruit were identified as B. cinerea. The results of the dual culture test showed that eight bacterial isolates could significantly inhibit the growth of B. cinerea mycelia from 50% to 80%. Two isolates B. amyloliquefaciens 28.3 and B. amyloliquefaciens 21/IV increased seed germination by 85.66% and 86.16%, respectively, compared to the control (82.66%). All bacterial isolates positively affected the morphological parameters of the seedlings. In the greenhouse experiment, three isolates B. subtillis 20.10, B. amyloliquefaciens 28.3, and B. amyloliquefaciens 21/IV and their mixture increased all measured PGP parameters. Isolates Bac 20.10 and Bac 28.3 reduced the severity of grey mould disease by 27.47% and 30.36%, respectively. These results show that Bacillus isolates have the potential for biological control of grey mould; also, that they have a positive effect on the growth of tomato plants.

Antagonistički potencijal sojeva Bacillus spp. prema prouzrokovaču sive truleži (Botrytis cinerea) paradajza u in vitro uslovima

AB  - Paradajz (Solanum lycopersicum L.) je značajna povrtarska vrsta čiju proizvodnju može
ugroziti veliki broj bolesti umanjujući prinos i kvalitet plodova. Siva trulež paradajza, koju
prouzrokuje gljiva Botrytis cinerea Pers., izaziva značajne štete na paradajzu, ali i na više od
1400 drugih domaćina širom sveta. Upotreba fungicida i dalje predstavlja najvažniju meru
kontrole ovog patogena. Međutim, zabrinutost javnosti zbog ostataka fungicida u plodovima i
pojava rezistentnosti patogena dovele su do stroži regulative u pogledu upotrebe fungicida.
Stoga se pojavila potreba za izučavanjem alternativnih mera suzbijanja patogena, a jedna od njih
je primena antagonističkih bakterija roda Bacillus kao bioloških agensa za kontrolu B. cinerea.
Ciljevi istraživanja bili su ispitivanje antagonističkog potencijala tri autohtona soja
Bacillus spp., poreklom iz Srbije, kao i njihove smeše prema izolatu B. cinerea 463-19 na
listovima i plodovima paradajza u in vitro uslovima. Listovi i plodovi paradajza dezinfikovani
su površinski 70% etil alkoholom u trajanju od 40 sekundi, a zatim tretirani prskanjem
suspenzije pojedinačnog soja ili smešom sojeva Bacillus spp. približne koncentracije 107
ćel/ml. Suspenzije su pripremljene od bakterijske kulture stare 24h gajene u LB (engl.
Lysogeny broth) podlozi. Nakon sušenja micelijarni diskovi B. cinerea, veličine 9 mm, dobijeni
od kulture stare 14 dana gajene na PDA (engl. Potato Dextrose Agar-krompir dekstrozni agar)
podlozi, postavljeni su u blizini lisnih nerava, ili na povređenu pokožicu ploda paradajza.
Intezitet oboljenja (engl. disease severity-DS %) na listovima izračunat je prema formuli:
DS(%)=((∑(broj zaraženih listova x intenzitet oboljenja))/(4xbroj ocenjenih listova))x100. Na
plodovima paradajza ocenjivan je procenat smanjenja razvoja truleži. Listovi i plodovi
inkubirani su pri 25 °C (12 h dan/12 hnoć), a ogled je ocenjen nakon 3 dana. Za svaki tretman
u ogledu korišćeno je po 10 listova ili 6 plodova, u tri ponavljanja. U kontroli plodovi i listovi
tretirani su sterilnom destilovanom vodom (SDV) ili preparatom Switch (aktivne materije
ciprodinil i fludioksonil) u koncentraciji 0,23%.
Listovi paradajza tretirani sojevima B. subtillis 20.10 i B. amyloliquefaciens 21/IV ispoljili
su manji intenzitet oboljenja (DS=47,5% i DS=50,8%) u poređenju sa listovima tretiranim SDV
(DS=89,1%) i preparatom Switch (DS=52,5%). Za soj B. amyloliquefaciens 28.3 zabeležena je
vrednost DS=56,6%, a za smešu sojeva DS=73,3%. Na plodovima tretman smešom sojeva dao
je najbolje rezultate i inhibirao razvoj truleži za 31,3% u odnosu na SDV. Pojedinačni tretman
sojevima Bacillus takođe je smanjio razvoj truleži na plodovima: B. amyloliquefaciens 21/IV za
25,7% i B. amyloliquefaciens 28.3 za 21,44%, dok je soj B. subtillis 20.10 umanjio razvoj truleži
za zanemarljivih 1,3%. U poređenju sa preparatom Switch, smeša sojeva umanjila je razvoj
truleži na plodovima za 39,9%, dok je pojedinični tretman sojem B. subtillis 20.10 umanjio
razvoj truleži za 13,7%, B. amyloliquefaciens 28.3 za 31,2%, a B. amyloliquefaciens 21/IV za
35%. Rezultati ukazuju da u in vitro uslovima sva tri soja Bacillus spp., pojedinačno kao i u
smeši, ispoljavaju antagonistički potencijal prema B. cinerea, kako na listovima tako i na plodovima čime je uspešno izveden prvi korak ka potencijalnoj primeni ovih bioloških agensa
u zaštiti paradajza od sive truleži.

AB  - Waitea circinata Warcup & P.H.B. Talbot (multinuclear Rhizoctonia spp.) is a widespread, soil-borne plant pathogen causing root rot mainly of monocotyledonous plants. Among five described varieties, the most common and widespread is W. circinata var. zeae Toda, Mushika, T. Hayak., Tanaka, Tani & Hyakum. Besides monocotyledonous hosts, a natural infection of a limited number of dicotyledonous plants, including soybeans, beans, peas, sugar beets and carrots were recorded recently. During 2017, cabbage and oilseed rape plants showing symptoms of root and root neck rot, sampled at the localities of Futog and Rimski Šančevi (Serbia), were proved to be infected with W. circinata var. zeae. This was the first record of natural infection of plants from Brassicaceae family worldwide. Two selected isolates from cabbage (299-17) and oilseed rape (300-17) formed fast-growing orange colonies with red sclerotia and multinucleate hyphae with a characteristic branching pattern. The identification was further confirmed by BLAST analysis of sequences of ITS rDNA, RPB2, LSU and β-tubulin genomic regions. Koch’s postulates were fulfilled after inoculation of cabbage and oilseed rape seedlings, while an experimental host range was determined by inoculations of 14 different plant species from 8 families.

AB  - Paradajz je jedna od najvažnijih povrtarskih biljaka čiju proizvodnju može ugroziti veći broj prouzrokovača bolesti, smanjujući prinos i kvalitet ploda. Jedan od najznačajnijih patogena paradajza je gljiva Botrytis cinerea - prouzrokovač sive truleži paradajza. Uprkos raznim preventivnim metodama, primena fungicida i dalje predstavlja najvažniju meru kontrole ovog patogena. Međutim, kontinuirana primena hemikalija izaziva zabrinutost potrošača zbog ostataka preparata na plodovima paradajza i njegovog štetnog uticaja na zdravlje ljudi. Jedna od alternativnih mera u zaštiti biljaka od prouzrokovača sive truleži jeste primena različitih antagonističkih bakterija, između ostalih i iz roda Bacillus. Osim uloge u biokontroli, poznato je da pomenute bakterije mogu stimulisati i rast biljaka. Ciljevi istraživanja bili su izolacija autohtonih sojeva Bacillus spp. iz zemljišta, ispitivanje njihove antifungalne aktivnosti prema dva izolata B. cinerea i proučavanje njihovog uticaja na klijavost semena paradajza i morfološke parametre klijanaca. Antifungalni efekat Bacillus spp. testitan je u in vitro uslovima, primenom metode dvojne kultivacije, u tri ponavljanja. Nakon 7 dana inkubacije pri 28ºC izračunat je procenat inhibicije rasta micelije (Percentage of Growth Inhibition, PGI) i zona inhibicije. Ispitivanje uticaja antagonističkih bakterija na klijavost semena paradajza izvršeno je standardnim testom klijavosti. Seme paradajza, sorte Novosadski jabučar, inokulisano je potapanjem u suspenziju bakterija (108 ćel/ml), dok je seme u kontroli potapano u 0,2% rastvor kalijum nitrata (KNO3). Efekat je ocenjen naklijavanjem 100 semena u Petri posudi (R=140 mm) u četiri ponavljanja. Očitavanje klijavosti semena izvršeno je nakon 14 dana. Iz svakog ponavljanja, slučajnim izborom odabrano je po 10 klijanaca i merena je dužina nadzemnog dela i korena (cm), kao i sveža i suva masa nadzemnog dela i korena (g). Primenom morfoloških i molekularnih metoda, pet bakterijskih sojeva identifikovano je kao Bacillus subtilis, dva soja kao B. amyloliquefaciens i jedan kao B. pumilus. U testu dvojne kultivacije svih osam sojeva Bacillus spp. ispoljili su snažan antifungalni efekat prema oba izolata B. cinerea, sa PGI vrednostima od 50% do 80% i zonom inhibicije od 1,33 mm do 17,33 mm. Najveći procenat klijavosti semena dobijen je inokulacijom sojevima B. amyloliquefaciens (85,66% i 86,16%) u odnosu na kontrolu (82,66%). Jedan soj B. subtillis povećao je dužinu nadzemnog dela za 25,50%, dok je drugi soj iste vrste povećao dužinu korena za 17,62%. Dalje, sojevi B. amyloliquefaciens, B. pumilus, kao i tri soja B. subtillis statistički značajno su povećali svežu masu nadzemnog dela (12,90 - 22,22%). Sojevi B. pumilus i B. subtillis povećali su i svežu masu korena (2,56 - 5,12%). Inicijalni rezultati ukazuju da autohtoni izolati Bacillus spp. iz zemljišta imaju snažan potencijal za biokontrolu i podsticanje rasta biljaka paradajza.

AB  - Proučavanja fitopatogenih gljiva i pseudogljiva u našoj zemlji imaju dugu tradiciju, a početak se uglavnom povezuje sa osnivanjem Poljoprivredno-šumarskog fakulteta davne 1919. godine. Fitopatogene gljive su danas u svetu široko poznati patogeni koji pričinjavaju oko 70% šteta od svih biljnih bolesti. U Srbiji, fitopatogene gljive i pseudogljive takođe redovno nanose velike štete u biljnoj proizvodnji pa su među proizvođačima prepoznate kao važni uzročnici gubitaka koji se mogu sprečiti ili ublažiti samo uz podršku nauke. Po svemu, istraživanja fitopatogenih gljiva i pseudogljiva danas predstavljaju jednu dobro profilisanu i značajnu oblast primenjenih istaživanja.

Waitea circinata var. zeae Causing Root Rot of Cabbage and Oilseed Rape

AB  - Cabbage, a widely used and popular vegetable, and oilseed rape, the second most valuable oilseed crop in the world, are two important species from the Brassicaceae family. Two geographically separated outbreaks of cabbage and oilseed rape root rot with estimated incidence of 15 and 20%, respectively, were recorded during 2017 in the Vojvodina region, Serbia. Twelve hyphal-tip isolates were obtained from symptomatic cabbage and oilseed rape plants and identified as Waitea circinata var. zeae based on morphological and molecular features. This indicates that W. circinata var. zeae has expanded its host range to the Brassicaceae family. Sequence analyses of internal transcribed spacer (ITS) and large subunit of the ribosomal DNA, RPB2, and B-tubulin genes revealed the highest similarity with multiple W. circinata var. zeae. Neighbor-joining analyses of ITS sequences resulted in a phylogenetic tree with one welldefined branch of W. circinata var. zeae, with two separate groups. All Serbian isolates and the majority of isolates originating from natural infection of dicotyledonous plants grouped together in group I. Following artificial inoculation, W. circinata var. zeae isolates caused mild to medium root necrosis of seedlings of 2 monocotyledonous and 12 dicotyledonous plant species, implying a wider host range than was known for W. circinata var. zeae. Additionally, this is the first occurrence of W. circinata var. zeae on dicotyledonous host plants in Europe. Because cabbage and oilseed rape are important crops grown worldwide, the occurrence of this new soilborne pathogen with a broad host range imposes the necessity for changes in routine disease control practices, particularly crop rotation.

AB  - Rhizoctonia spp. comprise soil-borne fungi widely distributed in temperate climate worldwide which are annually causing significant yield loses to a range of cultivated plants. A species status of Rhizoctonia spp. has not yet been established and the isolates are classified based on nuclei status as multinucleate and binucleate Rhizoctonia spp., and further divided into 13 anastomosis groups (AG) (AG-1-13) of multinucleate Rhizoctonia and 23 AG (AG-A-W) of binucleate Rhizoctonia. In this paper we are reporting morphological and molecular features of binucleate Rhizoctonia spp. AG-A causing black root rot on strawberry and multinucleate R. solani AG-2-2 causing root and crown rot of sugar beet. During 2015 and 2016 at several strawberry and sugar beet producing areas, plants expressing prominent root and crown necrosis and rot have been observed with disease incidence estimated at 20-30%.

AB  - Species of the genus Alternaria are among the most important plant pathogens that occur worldwide on many host plants, as well as plants of the Apiaceae family, including important vegetables such as carrots, parsley, celery, parsnips and fennel, and popular herbs such as anise, cumin, dill, coriander and others. These plants are attacked by different species of phytopathogenic fungi of the genus Alternaria, and this paper offers current review of nine different species. However, there is still insufficient information on both these species and the nu-merous interactions between them and their host plants and environmental factors. This information are necessary as the basis for adequate disease control measures and thus enabling successful and profitable cultivation of these high valued crops. Therefore, we present pathogen profiles of A. dauci, A. radicina, A. carotiincultae, A. petroselini, A. selini, A. smyrnii, A. alternata, A. longipes and A. burnsii as pathogens of Apiaceae plants, as well as their full current names and synonyms, taxonomic position, distribution and symptoms these pathogens cause to their host plants, as well as their biology and mode of transmission and spread, with special emphasis on control measures.
AB  - Vrste roda Alternaria spadaju među najvažnije biljne patogene koji se pojavljuju širom svijeta na mnogim biljkama domaćinima, uključujući i biljke iz familije Apiaceae. Među njima posebno su značajne povrtrarske biljke kao što su mrkva, peršun, celer, paštrnak i komorač, kao i brojne i često upotrebljavane začinske biljke, kao što su anis, kim, mirođija, korijander i druge. Ove biljke napadaju različite vrste fitopatogenih gljiva iz roda Alternaria, pri čemu je u ovom radu opisano devet vrsta. Međutim, još uvijek je nedovoljno informacija kako o ovim vrstama, tako i o brojnim interakcijama između njih i njihovih biljaka domaćina i faktora spoljne sredine. Ove informacije su neophodne kao osnova za preduzimanje adekvatnih mjera za njihovo suzbijanje i omogućavanje uspješnog i profitabilnog uzgoja ovih na tržištu izuzetno traženih i cijenjenih biljnih vrsta. Zbog toga su u ovom radu prikazane vrste roda Alternaria prourokovači bolesti povrtarskih i začinskih biljaka iz familije Apiaceae, pre svega A. dauci, A. radicina, A. carotiincultae, A. petroselini, A. selini, A. smyrnii, A. alternata, A. longipes i A. burnsii, te njihovi puni aktuelni nazivi i sinonimi, taksonomska pozicija, rasprostranjenost i simptomi koje prouzrokuju na biljkama domaćinima, kao i njihova biologija i način prenošenja i širenja, uz poseban naglasak na mjere za njihovo suzbijanje.

Conventional and real-time PCR assays for detection and identification of Rhizoctonia solani AG-2-2, the causal agent of root rot of sugar beet

AB  - Soil-borne fungi belonging to the genus Rhizoctonia are considered to be among the most destructive sugar beet pathogens. Although multinucleate R. solani AG-2-2 is frequently detected as the main causal agent of root rot of sugar beet worldwide, several binucleate (AG-A, AG-E and AG-K) and multinucleate Rhizoctonia (R. solani AG-4, AG-5 and AG-8) have also been included in the disease complex. Due to their soil-borne nature and wide host range, the management of Rhizoctonia root rot of sugar beet is highly demanding. Identification of Rhizoctonia AG associated with root rot of sugar beet is the essential first step in determining a successful disease management strategy. In this paper we report a highly specific and sensitive real-time PCR protocol for detection of R. solani AG-2-2 which showed a high level of specificity after testing against 10 different anastomosis groups and subgroups, including AG-2-1 as the most closely related. Moreover, a similar conventional PCR assay showed the same specificity but proved to be at least a 100 times less sensitive. Future research will include further testing and adaptation of this protocol for direct detection and quantification of R. solani AG-2-2 in different substrates, including plant tissue and soil samples.
AB  - Smatra se da su gljive iz roda Rhizoctonia koje se održavaju u zemljištu, najdestruktivniji patogeni šećerne repe. Mada je širom sveta višejedarna R. solani AG-2-2 najčešće označena kao najvažniji prouzrokovač truleži korena šećerne repe, nekoliko dvojedarnih (AG-A, AG-E i AG-K) kao i višejedarnih Rhizoctonia (R. solani AG-4, AG-5 i AG-8) takođe mogu da učestvuju u kompleksu bolesti. Kako se održavaju u zemljištu i imaju širok krug domaćina, suzbijanje vrsta Rhizoctonia koje izazivaju trulež korena šećerne repe veoma je zahtevno. Identifikacija anastomoznih grupa prouzrokovača oboljenja predstavlja neophodan prvi korak u uspostavljanju uspešnog suzbijanja. U ovom radu razvijen je visokospecifičan i osetljiv real-time PCR protokol za detekciju i identifikaciju R. solani AG-2-2 koji se pokazao kao visokospecifičan nakon testiranja koja su obuhvatila 10 različitih AG grupa i podgrupa, uključujući i AG-2-1 kao najsrodniju. Sličan konvencionalni PCR protokol ispoljio je istu specifičnost, ali i najmanje 100 × manju osetljivost. Buduća istraživanja uključiće dalje testiranje i adaptaciju ovog protokola za direktnu detekciju i kvantifikaciju R. solani AG-2-2 u različitim uzorcima, uključujući biljno tkivo i zemljište.

AB  - Strawberry production is a popular, fast-growing agricultural business in Serbia. Its cultivar selection has been changing fast, following market demands. One of the limiting factors of strawberry production worldwide is black root rot, primarily caused by binucleate Rhizoctonia. Recently, outbreaks of black root rot of strawberry have occurred in Serbia and the estimated disease incidence was up to 30%. Isolates of binucleate Rhizoctonia AG-A were recovered from symptomatic strawberry plants, and characterized on the bases of morphological, molecular and pathogenic features. Despite their uniform morphological characteristics, the isolates demonstrated genetic variability within ITS rDNA, grouping into three different phylogenetic sub-clusters which comprise AG-A isolates originating from Italy, Israel, Japan and the USA. The binucleate Rhizoctonia AG-A from Serbia exhibited uniform virulence on strawberry after inoculation of daughter plants and detached leaf petioles, as well as on seedlings of bean, carrot and sunflower, while they were non-pathogenic to wheat, maize, tomato, pepper, tobacco, cucumber, lettuce, peas, cabbage, rapeseed and sugar beet.
AB  - Proizvodnja jagode je popularna grana poljoprivrede u Srbiji koja se brzo razvija. Sastav sorti uključenih u proizvodnju podleže stalnim promenama u skladu sa zahtevima tržišta. Jedan od ograničavajućih faktora u proizvodnji jagode širom sveta je crna trulež korena, bolest koju najčešće izazivaju dvojedarne vrste roda Rhizoctonia. U Srbiji je poslednjih godina nekoliko puta zabeležena pojava crne truleži jagode uz učestalost koja je iznosila do 30%. Iz biljaka jagode sa simptomima izdvojeni su izolati dvojedarne Rhizoctonia AG-A i okarakterisani na osnovu morfološih i molekularnih osobina i patogenosti. Uprkos uniformnim morfološkim osobinama, izolati su ispoljili genetičku varijabilnost na osnovu sekvence ITS rDNK, grupišući se u tri različita filogentska sub-klastera koji su obuhvatali isolate iz Italije, Izraela, Japana i SAD. Izolati dvojedarnih Rhizoctonia AG-A poreklom iz Srbije ispoljili su uniformnu virulentnost na jagodi nakon inokulacija živića i fragmenata lisnih peteljki, kao i na sejancima pasulja, mrkve i suncokreta, dok nisu bili patogeni za pšenicu, kukuruz, paradajz, papriku, duvan, krastavac, salatu, grašak, kupus i šećernu repu.

The presence of turnip yellows virus in oilseed rape (Brassica napus L.) in Serbia

AB  - A total of 86 oilseed rape samples from six crops in different localities were collected during 2014 and analyzed for the presence of Turnip yellows virus (TuYV), Cauliflower mosaic virus (CaMV) and Turnip mosaic virus (TuMV) using commercial double-antibody sandwich (DAS)-ELISA kits. TuYV was serologically detected in 60 collected samples (69.77%), and none of the samples tested were positive for CaMV and TuMV. Six selected TuYV isolates were successfully transmitted by Myzus persicae to three test plants, confirming the infectious nature of the disease. In the selected ELISA-positive samples, the presence of TuYV was further confirmed by RT-PCR and sequencing. A comparison of the obtained sequence with those available in GenBank confirmed the presence of TuYV in oilseed rape samples. An analysis of P0 gene sequence data for a subset of these isolates showed they clustered with the known TuYV and were distinct from Beet western yellows virus (BWYV) isolates.
AB  - Tokom 2014. godine, sa šest lokaliteta gajenja uljane repice, ukupno je sakupljeno 86 uzorka koji su serološki testirani na prisustvo virusa žutice postrne repe [Turnip yellow virus (TuYV)], virusa mozaika karfiola [Cauliflower mosaic virus (CaMV)] i virusa mozaika postrne repe [Turnip mosaic virus (TuMV)], korišćenjem komercijalno dostupnih kitova za DAS-ELISA test. Prisustvo TuYV dokazano je u 60 (69.77%) prikupljenih uzoraka, dok prisustvo CaMV i TuMV nije dokazano ni u jednom od testiranih uzoraka. Za dalja istraživanja odabrano je šest uzoraka prirodno zaraženih biljaka uljane repice poreklom iz različitih lokaliteta koji su uspešno preneti vašima na tri različite test biljke, čime je potvrđena infektivna priroda oboljenja. AB  - During 2009 and 2010, a survey was conducted in pepper crops to detect the possible presence of Pepper mild mottle virus (PMMoV) in Serbia. A total of 239 pepper samples from 39 crops at 26 localities were collected and analyzed for the presence of PMMoV, Cucumber mosaic virus (CMV), Potato virus Y (PVY), and Alfalfa mosaic virus (AMV), using DAS-ELISA test. Although it was detected in a small percentage, PMMoV could pose a threat to pepper production in Serbia due to its rapid seed-borne spread. Presence of PMMoV was confirmed by serological and biological detection, followed by conventional reverse transcription RT-PCR, using primers specific for the RNA-dependent RNA polymerase (RdRp) and the coat protein (CP) genes. Molecular identification confirmed that the Serbian isolates belong to PMMoV pathotypes P1,2 which do not break the resistance gene L-3. Reconstructed phylogenetic tree confirmed the allocation of the Serbian isolates together with the majority of PMMoV isolates which belong to pathotypes P-1,P-2. This study represents the first serological and molecular characterization of PMMoV infection of pepper in Serbia, and provides important data on the population structure. The obtained data could have great influence on pepper production in Serbia as well as future pepper resistance breeding in the country.

Presence and distribution of pepper viruses in Serbia

AB  - Pepper is a very popular and profitable crop in Serbia and its production is growing rapidly as well as the importance of diseases caused by viruses. Virus infections interfere with development of pepper plants, reducing yield and fruit quality. More than 45 viruses have been isolated from pepper so far, and in our country the following are considered economically very important: Tobacco mosaic virus (TMV), Cucumber mosaic virus (CMV), Alfalfa mosaic virus (AMV), Tomato spotted wilt virus (TSWV) and Potato virus Y (PVY). During 2009, an investigation was conducted regarding the presence and distribution of pepper viruses in Serbia, which included a survey of different localities of transplant production, and both greenhouse and open field pepper crops. Collected samples were tested utilizing a double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) with polyclonal antisera specific for the detection of economically important pepper viruses: PVY, AMV, CMV, TSWV, TMV and Potato virus X (PVX). In the collected samples, presence of the four viruses (PVY, CMV, TSWV and AMV) was confirmed in single and mixed infections. In tested transplant samples, the most frequently detected virus was CMV (18.18%), while PVY was the most frequent in samples collected in greenhouse and open field pepper crops (32.56% and 51.21% respectively). The presence of TSWV was detected only in greenhouse-grown pepper crops, while the presence of PVX and TMV was not detected during this investigation. The obtained results indicated that PVY and CMV were widely distributed and the most frequent viruses in pepper crops in Serbia. Future investigation should include their detailed biological and molecular characterization, as well as the implementation of appropriate control measures in pepper productions.
AB  - Virusne bolesti paprike mogu ugroziti normalan razvoj biljaka i time značajno umanjiti prinos i kvalitet plodova. U svetu je opisano više od 45 virusa infektivnih za papriku, među kojima su ekonomski najznačajniji virus mozaika duvana (Tobacco mosaic virus, TMV), virus mozaika krastavca (Cucumber mosaic virus, CMV), virus mozaika lucerke (Alfalfa mosaic virus, AMV), virus bronzavosti paradajza (Tomato spotted wilt virus, TSWV) i virus crtičastog mozaika krompira (Potato virus Y, PVY). Prisustvo i rasprostranjenost virusnih zaraza paprike u Srbiji ispitivani su tokom 2009. u različitim lokalitetima proizvodnje rasada, kao i useva paprike gajene u zaštićenom prostoru i na otvorenom polju. Sakupljeni uzorci lišća sa simptomima testirani su DAS-ELISA metodom na prisustvo: PVY, AMV, CMV, TSWV, TMV i virusa mozaika krompira (Potato virus X, PVX). Dobijeni rezultati ispitivanja su ukazali na prisustvo četiri virusa: PVY, CMV, TSWV i AMV, koji su identifikovani u pojedinačnim ili mešanim infekcijama. U testiranim uzorcima paprike iz rasada najzastupljeniji je bio CMV (18,18%), dok je u uzorcima prikupljenim iz plastenika i polja najčešći bio PVY (32,56% i 51,21%). Prisustvo TSWV dokazano je samo na biljkama paprike gajenim u zaštićenom prostoru. Prisustvo PVX i TMV nije ustanovljeno ni u jednom ispitivanom uzorku paprike. Utvrđena značajna rasprostranjenost i zastupljenost PVY i CMV u proizvodnji paprike u Srbiji ukazuje na potrebu detaljne karakterizacije ova dva virusa na biološkom i molekularnom nivou, kao i sprovođenje odgovarajućih mera kontrole.
