Subcritical Water Extraction as an Effective Technique for the Isolation of Phenolic Compounds of Achillea Species

AB  - The genus Achillea has significant medical potential due to the presence of highly bioactive compounds in its chemical composition. To take advantage of plants’ biomedical potential, it is of great importance to use a proper extraction process. This study aimed to determine and compare the preliminary chemical composition of five different Achillea species extracted with two conventional (infusion and maceration) and two non-conventional (ultrasound-assisted extraction (UAE) and Subcritical water extraction (SWE)) techniques. The extracts were prepared using the previously described procedures for infusion, maceration, UAE and SWE extraction. For all extracts, the extraction yield (dry extract (DE)) was determined. The analyzed extracts were preliminarily chemically characterized spectrophotometrically in terms of total phenolic content (TPC) and total flavonoid content (TFC). The obtained results showed that non-conventional techniques delivered higher values of TPC and TFC than conventional. There is a statistically significant increase in DE and TPC content when applying SWE for all observed Achillea species. The highest DE value, 48.80 ± 1.76%, was observed for A. asplenifolia. The highest TPC values were observed after applying SWE: 93.63 ± 1.01 mg GAE/g DE for A. millefolium, and 90.12 ± 0.87 mg GAE/g DE for A. crithmifolia. The results for TFC revealed a statistically significant difference in values, with A. nobilis subsp. nelreichii as the sample with the highest content of TFC (11.11 ± 0.22 mg QE/g DE) when using UAE. Consequently, it could be concluded that SWE is a superior non-conventional extraction technique, and A. nobilis subsp. nerleichii presents as the most promising plant.

AB  - Main conclusion Vavilovia formosa (Stev.) Fed. is a scientifically valuable common ancestor of the plant tribe Fabeae and also important in breeding and agronomy studies of the cultivated Fabeae, but it is close to extinction. A concerted academic and geovernmental effort is needed to save it. Since 2007, an informal international group of researchers on legumes has been working to increase awareness of Vavilovia formosa (Stev.) Fed., a relict and endangered wild-land relative to crop plant species. A majority of the modern botanical classifications place it within the tribe Fabeae, together with the genera vetchling (Lathyrus L.), lentil (Lens Mill.), pea (Pisum L.) and vetch (Vicia L.). V. formosa is encountered at altitudes from 1,500 m up to 3,500 m in Armenia, Azerbaijan, Georgia, Iran, Iraq, Lebanon, Russia, Syria and Turkey. This species may be of extraordinary importance for broadening current scientific knowledge on legume evolution and taxonomy because of its proximity to the hypothetical common ancestor of the tribe Fabeae, as well as for breeding and agronomy of the cultivated Fabeae species due to its perenniality and stress resistance. All this may be feasible only if a concerted and long-term conservation strategy is established and carried out by both academic and geovernmental authorities. The existing populations of V. formosa are in serious danger of extinction. The main threats are domestic and wild animal grazing, foraging, and early frosts in late summer. A long-term strategy to save V. formosa from extinction and to sustain its use in both basic and applied research comprises much improved in situ preservation, greater efforts for an ex situ conservation, and novel approaches of in vitro propagation.

AB  - The development of agriculture was a key turning point in human history, a central part of which was the evolution of new plant forms, domesticated crops. Grain legumes were domesticated in parallel with cereals and formed important dietary components of early civilizations. First domesticated in the Near East, pea has been cultivated in Europe since the Stone and Bronze Ages. In this study, we present a molecular analysis of ancient DNA (aDNA) extracted from carbonized pea seeds recovered from deposits at Hissar, in southeast Serbia, that date to the eleventh century B.C. Four selected chloroplast DNA loci (trnSG, trnK, matK and rbcL) amplified in six fragments of 128-340 bp with a total length of 1,329 bp were successfully recovered in order to distinguish between cultivated and wild gathered pea. Based on identified mutations, the results showed that genuine aDNA was analyzed. Moreover, DNA analysis resulted in placing the ancient sample at an intermediate position between extant cultivated [Pisum sativum L. and wild P. sativum subsp. elatius (Steven ex M. Bieb.) Asch. et Graebn.]. Consequently, based on a combination of morphological and molecular data, we concluded that the material represents an early domesticated pea. We speculate that Iron Age pea would be of colored flower and pigmented testa, similar to today's fodder pea (P. sativum subsp. sativum var. arvense (L.) Poir.), possibly of winter type. This is the first report of successful aDNA extraction and analysis from any legume species thus far. The implications for pea domestication are discussed here.

Moreover, DNA analysis resulted in placing the ancient sample at an intermediate position between extant cultivated [Pisum sativum L. and wild P. sativum subsp. elatius (Steven ex M. Bieb.) Asch. et Graebn.]. Consequently, based on a combination of morphological and molecular data, we concluded that the material represents an early domesticated pea. We speculate that Iron Age pea would be of colored flower and pigmented testa, similar to today's fodder pea (P. sativum subsp. sativum var. arvense (L.) Poir.), possibly of winter type. This is the first report of successful aDNA extraction and analysis from any legume species thus far. The implications for pea domestication are discussed here.",

AB  - An increasing interest in cultivation and improvement of faba bean (Vicia faba L.), an important food and feed grain legume whose ex siz genetic resources comprise cultivated forms only, triggers both search for novel genotypes and their molecular characterization. Due to the poor representation of faba bean genotypes from the central Balkans in worldwide ex situ collections, we collected seeds of 13 faba bean landraces from this region, preserved them at the French National Institute for Agricultural Research (INRA) ex situ collection and used them also for amplification of ten nuclear microsatellites (simple sequence repeats — SSRs). Given the self-pollinating nature of faba bean and low heterozygosity levels in this crop, we assumed that relatively low number of alleles may be expected in each landrace. Therefore, we first demonstrated that all alleles obtained upon amplification of ten SSRs in 11 individuals from one landrace could be detected when DNAs from these individuals were pooled and such a bulk DNA was used as a template for PCR reactions, and then tested all SSRs in bulk DNA templates from each out of ten remaining landraces comprising six to 15 genotypes.

AB  - The flora of Pčinja valley, as situated in southern Balkan Peninsula, is comparatively rich in native legume species, representing important genomic and breeding resources for legume crops and forages. According to their taxonomical diversity, legume family belongs to the group of most important plant families in the flora of Serbia, and at the same time the second best represented in the flora of Pčinja valley. Considering elements different in origin and ecological requirements this family comprises impressive list of 100 species and subspecies that represents more than 40% of their total number in Serbia. At the same time such a large number of representatives are distributed across the area of hardly 0.17% of whole country area. The genus Trifolium, referring 32 taxa, is the most abundant among all other genera in the flora of the valley. The following genera: Vicia (15), Lathyrus (11), Medicago (7), Chamacytisus (5) and Astragalus (4) are also considerably rich at the family level. The life forms spectrum points at ecological group of annual representatives taking the largest portion of the all legume flora of valley.

AB  - A unique example of 2,572 carbonized pea seeds was recovered from the 11th century B.C. deposits at Hissar, a multilevel settlement of the Brnjica cultural group (1,350-1,000 B.C.) in southeast Serbia. Five hundred seeds maintained pea-like hilum, only few had preserved intact smooth-surfaced testa and the majority of seeds had no seed coat. Fortunately, only few seeds of other pulses were observed in the sample and their identity was clearly determined. Applying the exclusion principle, all the “naked”, 3-4 mm large seeds, mainly broad ellipsoid and less often globose, with flattenings or concavities, were determined to correspond to cultivated pea (Pisum sativum L.). Additionally, a high thousand seed weight of charred seeds (24.4 g) suggested cultivated status. To confirm our finding, we processed two samples with molecular tools. A sequence analysis of four chloroplast DNA loci (trnSG, trnK, matK and rbcL) in total length of 1329bp, showed intermediate position to cultivated P. sativum and wild P. sativum subsp. elatius.

AB  - European Legumes and Their Traditional Food and Feed Products SEELEGUMES) has been carried out within the EU SEE-ERA.NET Plus programme, gathering together 19 partnering organisations from 14 European countries. One of the greatest achievements of the project was a re-discovery of numerous crop wild relatives of cultivated annual legumes in local floras, with a specific emphasis on "tall" pea (Pisum sativum L. subsp. elatius Steven ex M. Bieb.) Asch. & Graebn.). During 2011 and 2012, five expeditions have been made to the upper flow of the river Pčinja and on the slopes of the mountain Kozjak, where a population of Pisum sativum L. subsp. elatius was monitored in situ, from the stages of full bloom in late May 2011, full seed maturity in late June 
2011, autumn emergence in October 2011, spring revival in March 2012 and the next reproductive stage in early June 2012. A morphological and chemical analysis of the forage and grain dry matter confirmed a considerable potential of Pisum sativum L. subsp. elatius for cultivation, while a prominent resistance to low temperatures has also been assessed.

AB  - Large-flowered vetch (Vicia grandiflora Scop.) is widely present in many Eurasian wild floras, where it has significance in improving grassland communities and environment-friendly uses as a cover crop and green manure. The main goal of this study was to assess the possibility of transforming large-flowered vetch into a field crop and assess the basics for its breeding. A small-plot trial was carried out in three consecutive growing seasons 2005-2008 in Novi Sad, including twelve large-flowered wild populations collected at various sites in Serbia and France from 2002 to 2004 and multiplied ex situ during subsequent years. The population MM 02/01 of Serbian origin had the highest three-year average proportion of plants surviving winter (0.96). The smallest average number of days from sowing to first flower was in the population MM 03/10 (199 days). The population MM 03/08 produced the highest forage dry matter (8.0 Mg ha(-1)) yield stage between full flowering and forming first pods. The highest seed yield was in the population MM 03/10 (2,084 kg ha(-1)). Forage dry matter yield was significantly correlated to main stem length (r = 0.932), number of fertile nodes (r = 0.710), number of internodes (r = 0.640) and number of leaves (r = 0.604). Seed yield was significantly correlated to number of pods (r = 0.930), number of seeds (r = 0.883), number of stems (r = 0.882) and number of fertile nodes (r = 0.768). Our study confirmed that large-flowered vetch may represent a stable and quality source of forage in temperate regions such as the Balkans, as well as that it may have its place in various crop rotations and farming systems. At the same time, the existing phenotypic variability offers a solid basis for further improvement of this species.

abstract = "Large-flowered vetch (Vicia grandiflora Scop.) is widely present in many Eurasian wild floras, where it has significance in improving grassland communities and environment-friendly uses as a cover crop and green manure. The main goal of this study was to assess the possibility of transforming large-flowered vetch into a field crop and assess the basics for its breeding. A small-plot trial was carried out in three consecutive growing seasons 2005-2008 in Novi Sad, including twelve large-flowered wild populations collected at various sites in Serbia and France from 2002 to 2004 and multiplied ex situ during subsequent years. The population MM 02/01 of Serbian origin had the highest three-year average proportion of plants surviving winter (0.96). The smallest average number of days from sowing to first flower was in the population MM 03/10 (199 days). The population MM 03/08 produced the highest forage dry matter (8.0 Mg ha(-1)) yield stage between full flowering and forming first pods. The highest seed yield was in the population MM 03/10 (2,084 kg ha(-1)).
