Sclerotinia wilt occurrence on sunflower in Vojvodina, Serbia

AB  - Sclerotinia sclerotiorim is a facultative parasite that can cause diseases on more than 400 plant species. Sclerotinia wilt presence was observed in Vojvodina during the period 2007-2009 at seven sites (Bečej, Kikinda, Kula, Pančevo, Sombor, Vršac and Zrenjanin). Assessment was done on 8 hybrids (Baća, Bačvanin, Branko, Duško, NS-H-111, Sremac, Šumadinac and Velja). Disease intensity was calculated according to McKinney's formula. AMMI analysis was used in order to estimate stability of hybrid tolerance to Sclerotinia wilt in different environments. Average Sclerotinia wilt occurrence on sunflower was the lowest in 2007 and the highest in 2009. Sclerotinia wilt was not observed in Zrenjanin and Vršac in 2007, nor in Pančevo in 2008. The lowest average Sclerotinia wilt occurrence during 3-year period was in Sombor and Pančevo. Hybrids Duško, NS-H-111, Sremac and Velja can be recommended for future growing on sites where Sclerotinia wilt of sunflower occurs.
AB  - Fakultativni parazit Sclerotinia sclerotiorum izaziva bolesti na više od 400 biljnih vrsta. Pojava bele truleži korena suncokreta na teritoriji Vojvodine je praćena tokom trogodišnjeg perioda (2007-2009) u sedam lokaliteta (Bečej, Kikinda, Kula, Pančevo, Sombor, Vršac i Zrenjanin), a u svakom lokalitetu je bilo ocenjeno osam hibrida (Baća, Bačvanin, Branko, Duško, NS-H-111, Sremac, Šumadinac i Velja). Stepen osetljivosti biljaka je izračunat po McKinney formuli. AMMI analizom je vršena procena stabilnosti hibrida na tolerantnost prema beloj truleži korena u različitim lokalitetima. Prosečna zastupljenost ovog oboljenja u 2007. godini je bila najniža, a u 2009. godini najviša. Bela trulež korena suncokreta u 2007. godini nije registrovana u Zrenjaninu i Vršcu, a u 2008. godini u Vršcu i Pančevu. Najniža prosečna pojava ove bolesti u trogodišnjem periodu je bila u Somboru i Pančevu. Na osnovu dobijenih rezultata hibridi Duško, NS-H-111, Sremac i Velja se mogu preporučiti za gajenje u lokalitetima na kojima se javlja bela trulež korena suncokreta.

Downy mildew and its control

AB  - Sunflower (Helianthus annuus) is the most important oil crop in Serbia, that produces high quality oil for human consumption and technical purposes as well. Sunflower is attacked by more than 40 different diseases of which only a certain number causes serious problems in the production process. Among these, the most important are phytopathogenic fungi. One of the most damaging diseases is downy mildew which is caused by Plasmopara halstedii fungus (Farl.) Berl.et Toni. (syn. Plasmopara helianthi Novit.). In conditions that favour its development it can reduce the yield of the sunflower by 50%. Many physiological races of this fungi hinder the control of the disease and that's why agrotechnical, chemical and quarantine measures should be applied. As the dominant race in our country is race 730, the testing of the selected material is aimed at creating hybrids of high resistance to this particular race of downy mildew. Other races are also taken into consideration and evaluated. As downy mildew occurred during the vegetation period in 2008, this research was designed to investigate the efficiency of fungicides, applied in case of secondary infection. We have measured oil and protein contents, germination and pathogen presence in seed of treated plants. Crops infested with Plasmopara helianthi and treated with Ridomil, developed healthy seed with unchanged oil and protein contents in comparison with non-treated healthy crop. According to given results, we can confirm that, when secondary infection of downy mildew appears, treatment with fungicides are reliable. Following NS hybrids in Serbian commercial sunflower production that contain Pl-6 gene, conferring resistance to most important races of downy mildew are recommended: Sremac, Duško, Plamen, Kazanova,Velja and Rimi PR.
AB  - Suncokret (Helianthus annuus) je najvažnija uljana kultura, od koje se dobija kvalitetno ulje za ljudsku ishranu i tehničku upotrebu. Suncokret napada više od 40 raznih prouzrokovača bolesti od kojih samo određen broj stvara velike probleme u proizvodnji (Aćimović, 1998). Značajno mesto zauzimaju fitopatogene gljive. Jedna od ekonomski najštet- nijih bolesti je plamenjača, koju prouzrokuje gljiva Plasmopara halstedii (Farl.) Berl. et Toni. (syn. Plasmopara helianthi Novot.). U povoljnim uslovima za njen razvoj, može da smanji prinos suncokreta i do 50%. Zbog prisustva velikog broja fizioloških rasa u okviru ove gljive, otežana je borba protiv ovog patogena. Zato se koriste agrotehničke mere, hemijske mere i karantinske mere borbe. U našoj zemlji dominira rasa 730, tako da je testiranje sortimenta usmereno u pravcu stvaranja otpornih hibrida na ovu rasu plamenjače. Ne zanemaruje se i mogućnost prisustva i drugih rasa. Zbog pojave plamenjače u toku vegetacije 2008. godine, predmet naših istraživanja je i hemijska mera borbe. One su opravdane u slučaju sekundarnih infekcija. U ovom radu na biljkama tretiranim fungicidima ispitan je kvalitet semena na sadržaj ulja i belančevina, klijavost i prisustvo patogena u semenu. Dobijeni rezultati su potvrdili naše pretpostavke o opravdanosti primene hemijske zaštite kod sekundarne pojave plamenjače.

Current status of broomrape (Orobanche cumana Wallr.) in Serbia

AB  - Broomrape is the major issue in sunflower production in Serbia. Since its first occurrence, the species Orobanche cumana Wallr. had been responsible for considerable yield loss in some areas. In Serbia broomrape is present in the Vojvodina Province. Distribution and virulence groups of parasite are constantly monitored. Only two distinct broomrape populations currently exist in this part of country. Race B is dominant in the south of the Vojvodina Province and race E in the north. During 2008, sets of 8 commercial hybrids with different broomrape resistance genes were sown in regions with sunflower production. Hybrids were sown on 34 different locations. Presence of broomrape was confirmed on 6 sites. Occurrence of parasitic plants was not detected on cultivars resistant to race E. Sunflower fields in regions where broomrape was not previously detected, were surveyed. The survey revealed occurrence of broomrapes in new regions. Infested areas are located in Bor County near the state border with Romania and Bulgaria.

Tolerance of several Novi Sad sunflower hybrids to phomopsis stem canker under natural infection

AB  - Sunflower diseases have a major impact on sunflower yield and quality of seed. When conditions during vegetation are favorable, the intensity of damage can be very high. Therefore, surveys of economically important diseases are necessary. A survey of incidences and severity of phomopsis stem canker was performed on 9 locations during 2007 and 2008. A total of 8 hybrids per location was examined. Lodging of plants as a symptom of stem canker disease was not detected. The mean disease incidence was less than 5%, depending on hybrid, location and year. Generally, higher deasease incidence was recorded in the first year of survey than in the second. The hybrids Velja, NS-H-111 and Baća demonstrated an elevated level of resistance. Despite significant differences in disease severity that was found among the hybrids tested, all of them were highly resistant to phomopsis stem canker.
AB  - Suncokret predstavlja našu najvažniju biljnu vrstu za dobijanje kvalitetnih biljnih ulja. Napada ga više od 40 raznih prouzrokovača bolesti od kojih samo određeni broj predstavlja problem u proizvodnji. Praćenje njihove pojave je značajno sa aspekta suzbijanja jer doprinosi stabilnosti proizvodnje. Pojava mrko-sive pegavosti stabla praćena je 2007. i 2008. godine u mreži demonstracionih ogleda. U prvoj godini posmatranja zabeležena je veća pojava bolesti na svim lokalitetima u odnosu na drugu godinu. Među hibridima u obe godine posmatranja najveća otpornost prema Phomopsis heliathi zabeležena je kod hibrida NS-H-111, Velja i Baća. Uprkos značajnim razlikama u intenzitetu pojave mrko-sive pegavosti stabla između hibrida, nije zabeležena jača pojava bolesti iz čega proizilazi da posmatrani hibridi poseduju visok stepen otpornosti prema Ph. helianthi u agroekološkim uslovima Vojvodine.

Current status of broomrape (Orobanche cumana Wallr) in Serbia

AB  - Broomrape is the parasitic plant that presents the major problem in sunflower production in Serbia. Since its first appearance in our country the species Orobanche cumana Wallr had been responsible for considerable yield loss in some sunflower growing areas. In Serbia broomrape is mostly present in Vojvodina province, where distribution and virulence groups of parasite are constantly monitored. Only two distinct broomrape populations currently exist in this part of country. According to previous research race B is dominant in south of the Vojvodina province and race E in the north. During 2008 set of 8 commercial hybrids with different broomrape resistance genes were sown in regions of Vojvodina and Central Serbia with sunflower production, on 34 different locations. Presence of broomrape was confirmed on 6 sites but occurrence of parasitic plants was not detected on hybrids resistant to race E. The survey of sunflower fields in regions where broomrape was not previously detected was also done. Survey revealed occurrence of broomrape in the new region. Infested areas are placed in Bor County near state border with Romania and Bulgaria

AB  - The large number of various species of fungi has been isolated from the sunflower seed samples. The most important pathogens found on the sunflower are: Plasmopara helianthi, Sclerotinia sclerotiorum, Botrytis cinerea and Phoma macdonaldii. The majority of these pathogens are transmitted through the seed. Presence of Phomopsis helianthi in the sunflower seed does not represent threat in the epidemiology of the transmission of pathogens in the nature. The saprophit species as Aspergillus and Penicillium are the potential carieres of mycotoxins. Control of isolated pathogenic and saprophit fungi includes preventive measurements such as the treatment of the seed by fungicides like metalaxyl, benomil and tiram.
AB  - Poznavanje mikopopulacije semena suncokreta je važan činilac u proizvodnji ove gajene biljke. Paraziti semena smanjuju njegovu nutritivnu vrednost, a ulje iz zaraženog semena je nepovoljnih prehrambenih karakteristika i organoleptičkih svojstava. Zbog toga se testira kompletan selekcioni materijal, seme, kao i stvoreni hibridi, na pojavu parazitnih, odnosno saprofitnih mikroorganizama (gljiva i bakterija).

The use of PCR-based markers in the evaluation of resistance to downy mildew in ns-breeding material

AB  - The aim of this work was the application of previously developed and development of new PCR markers for the evaluation of sunflower resistance to downy mildew. Twenty sunflower inbred lines were investigated. Plant resistance to downy mildew was determined by the whole seedling immersion method. Genomic DNA was extracted from the first pair of leaves, and its polymorphism was investigated by RAPD, SSR and several published markers for disease resistance. The presence of the markers Ha-NBS 7/R, Ha-NBS 8/R Ha-NBS 9/R, in Pl6 donor lines (Ha-335, JM-8) and in resistant progeny (Ha-26, G12, G10, G11) confirm that HAP3 could be useful for the detection of the Pl6 gene. DNA polymorphism, which coincided with disease resistance, was revealed with one RAPD (UBC 119 fragment 900-1000 bp) and one SSR primer (ORS37 fragment 600-700 bp). Amplified fragments segregated in the same way i.e., they appeared in 50% of the resistant genotypes. The non-expecting SSR fragment was purified, cloned and sequenced. The results indicated that this fragment is not a part of a coding sequence. Specific primers for the amplification of this fragment have been designed and the investigation of the inheritance of this SCAR marker is under way. None of the applied markers appeared in all resistant genotypes. In order to select lines for making crosses for use in further investigation, the obtained results were also used for the calculation of genetic distances between genotypes (simple matching coefficient) and the construction of a dendrogram (UPGMA method).

Testing two isolates of diaporthe/phomopsis helianthi in a population of sunflower recombinant inbred lines

AB  - Development of new, more aggressive isolates of Phomopsis helianthi in Serbia and Montenegro has been investigated. Two isolates of Phomopsis helianthi Yu 4 and Yu 12, had been found on the stem of NS-H-45, a highly tolerant sunflower hybrid. The isolate Yu 4 was isolated in the northern part of the Vojvodina Province, a region where Phomopsis typically occurs. The isolate Yu 12 comes from the southern part of the Vojvodina Province, a region along the Danube River, which suffered an intensive outbreak of Phomopsis in 1997. A preliminary study of the isolates showed that these two isolates were significantly different for radial growth in Petri dish. A test was thus made with recombinant inbred lines in the F9 generation developed from the cross of LR-4-17 (inbred line with high tolerance to Phomopsis) and HA-89 (susceptible inbred line). Experiments were conducted in the greenhouse inoculating plant leaf tips with the mycelium of the fungus. The test showed that there were no significant differences between the two isolates, Yu 4 and Yu 12. It was concluded that the intensive attack recorded in 1997 along the Danube River occurred due to exceptionally favorable environmental conditions for the development of the disease.
