Variability of agronomic traits in vegetable pea (Pisum sativum L.) genotypes

AB  - In this study, a total of 12 vegetable pea genotypes of different growing seasons were subjected to phenotypic characterization. The vegetable pea genotypes are a part of the collection maintained at the Institute of Field and Vegetable Crops Novi Sad. The plant material included 10 promising lines and two released cultivars, Tamiš and Dunav. The trial was carried out in 2022. It was set up at the Rimski Šančevi site, as a randomized block design in five replications. A total of 14 agronomic traits were analyzed. The obtained research results revealed divergence in the investigated plant material. The statistical significance of all sources of variation was determined by the LSD test. The height of the first fertile node was found to be the most variable feature, with a coefficient of variation of 40.54%. The tested genotypes were clustered into two groups and two subgroups within the second group. The correlation analysis of the examined quantitative traits revealed the presence of several statistically significant positive and negative correlations. Some of the most significant positive correlations were established between the grain weight per plant and the number of grains per plant and the yield of technologically mature grain, while the pod width and the number of fertile nodes per plant had the most negative correlations with the other tested traits.
AB  - U toku ovog istraživanja izvršena je fenotipska karakterizacija 12 genotipova povrtarskog graška, različite dužine vegetacije, koji pripadaju kolekciji Instituta za ratarstvo i povrtarstvo u Novom Sadu. Biljni materijal obuhvatao je 10 perspektivnih selekcionih linija i dve priznate sorte tamiš i dunav. Ogled je izveden tokom 2022. godine, postavljen je po slučajnom blok sistemu, u pet ponavljanja na lokalitetu Rimski Šančevi. Analizirano je 14 agronomskih osobina. Dobijeni rezultati istraživanja ukazuju na postojanje divergentnosti u ispitivanom biljnom materijalu. Analizom varijanse ispitivanih agronomskih osobina utvrđena je statistička značajnost svih izvora varijacije. Kao najvarijabilnija osobina istakla se visina prvog rodnog nodusa sa koeficijentom varijacije od 40,54%. Ispitivani genotipovi su primenom klaster analize grupisani u dve grupe sa dve podgrupe u okviru druge grupe. Korelacionom analizom ispitivanih kvantitativnih svojstava utvrđeno je postojanje nekoliko statistički značajnih pozitivnih i negativnih međuzavisnosti. Neke od najznačajnijih pozitivnih korelacija utvrđene su između osobina: masa zrna po biljci i prinos tehnološki zrelog zrna, kao i broja zrna po biljci i prinosa tehnološki zrelog zrna, dok svojstva širina mahune i suma rodnih nodusa po biljci imaju najviše negativnih korelacija sa ostalim ispitivanim osobinama.

AB  - Biostimulants are used for seed quality and performance improvement. However, the impact of biostimulants on parsnip seed quality has not been examined. Slow germination of parsnip is often further impeded by stress conditions. This study therefore aimed to assess whether treatments with different biostimulants could enhance seed germination and the initial seedling growth of parsnip. Amino acid and micronutrient-based biostimulants (0.2% solutions (v/v) of Technokel Amino Mix® and Megafol®) improved germination of parsnip seeds under optimal conditions, while biostimulant treatments with humic acid, fulvic acid and micronutrients (0.02% solution (v/v) of Organiko®) enhanced the initial growth. Our findings validate the potential of biostimulant application as a seed treatment. Determination of the main physiological causes of parsnip germination enhancement is needed in further research.

Bacillus Species: Excellent Biocontrol Agents against Tomato Diseases

AB  - Tomatoes encounter many pathogens, such as fungi and bacteria, which reduce the yield and
quality of plants and lead to large losses in production. The application of plant protection products
(PPPs) is still an important and most effective measure to control plant diseases. However, the use of chemicals in agriculture contributes to environmental pollution and biodiversity loss, and it can also threaten non-target living organisms. Biological control is a widely accessible, environmentally safe, and cost-efficient alternative for the prevention and suppression of plant diseases. Bacillus species with antimicrobial and plant growth-promoting effects are most frequently used as biocontrol agents to increase the resilience of agricultural production against biotic stresses. The present review discusses the antagonistic mechanisms and the biocontrol potential of Bacillus spp. against tomato diseases caused by different pathogens. The main mechanisms of Bacillus spp. include the production of antimicrobial compounds (antibiotics, extracellular enzymes, siderophores, and volatile compounds), competition for nutrients and space, and induced systemic resistance (ISR). Although Bacillus-based PPPs have been developed and commercialised worldwide for various crops and pathogens, the efficiency issues are still subject to debate. Additionally, a combined strategy for controlling tomato diseases based on Bacillus spp. and other available methods (conventional or natural-based) is a promising research field.

AB  - Poljoprivredna proizvodnja je u velikoj meri uslovljena klimatskim faktorima. Scenario globalne promene klime predviđa da će naredne godine biti sušne, sa toplijom klimom, koja će imati negativan uticaj na visinu prinosa ratarskih biljaka. Alternativne biljne vrste-ABV, zahvaljujući tolerantnosti prema suši i visokim temperaturama, imaju perspektivu u strukturi ratarske proizvodnje. Za dobijanje visokog i stabilnog prinosa semena alternativnih biljnih vrsta neophodno je pored kvalitetnog sertifikovanog semena primeniti i odgovarajuću sortnu tehnologiju gajenja, prilagođenu zahtevima pojedinih sorti, klimatskim uslovima lokaliteta gajenja i osobinama zemljišta. U ovoj studiji su ispitivani prinosi sorti: prosa (Biserka i Rumenka), heljde (Novosadska), sirka za zrno (Alba i Gold), sirka metlas a (Prima i Reform), konoplje (Helena i Marina), facelije (NS Priora), uljanog lana (NS Primus i NS Marko) i uljane tikve (Olivija i Olinka). Ostvareni prosečni prinosi ABV u 2022. godini bili su na nivou višegodišnjeg proseka. Visoke temperature i nedostatak padavina u kritičnim fazama rasta biljaka su uslovili prosečne i nešto niže prinose. Evidentne su velike i fluktuacije prinosa u zavisnosti od primenjenih agrotehničkih mera u toku vegetacionog perioda: vremena setve i navodnjavanja.

abstract = "Poljoprivredna proizvodnja je u velikoj meri uslovljena klimatskim faktorima. Scenario globalne promene klime predviđa da će naredne godine biti sušne, sa toplijom klimom, koja će imati negativan uticaj na visinu prinosa ratarskih biljaka. Alternativne biljne vrste-ABV, zahvaljujući tolerantnosti prema suši i visokim temperaturama, imaju perspektivu u strukturi ratarske proizvodnje. Za dobijanje visokog i stabilnog prinosa semena alternativnih biljnih vrsta neophodno je pored kvalitetnog sertifikovanog semena primeniti i odgovarajuću sortnu tehnologiju gajenja, prilagođenu zahtevima pojedinih sorti, klimatskim uslovima lokaliteta gajenja i osobinama zemljišta. U ovoj studiji su ispitivani prinosi sorti: prosa (Biserka i Rumenka), heljde (Novosadska), sirka za zrno (Alba i Gold), sirka metlas a (Prima i Reform), konoplje (Helena i Marina), facelije (NS Priora), uljanog lana (NS Primus i NS Marko) i uljane tikve (Olivija i Olinka). Ostvareni prosečni prinosi ABV u 2022. godini bili su na nivou višegodišnjeg proseka. Visoke temperature i nedostatak padavina u kritičnim fazama rasta biljaka su uslovili prosečne i nešto niže prinose. Evidentne su velike i fluktuacije prinosa u zavisnosti od primenjenih agrotehničkih mera u toku vegetacionog perioda: vremena setve i navodnjavanja.",

AB  - Heat stress seriously affects the production of cool-season food legume crops such as
garden peas. Seed priming is a widely used technique that increases germination and improves
plant growth and development, resulting in better field performance and higher yield of crops. In
the current study, we investigated three seed priming treatments—hydropriming (dH2O),
osmopriming (2.2% w/v CaCl2), and hormopriming (50 mg L-1 salicylic acid - SA)—and their effect
on germination, initial seedling development, and physiological traits of two novel garden pea
cultivars, under optimal conditions and heat stress. Seed priming with H2O, CaCl2, and SA enhanced
garden pea performance under both optimal and stress conditions via significant improvements in
germination energy, final germination, mean germination time, mean germination rate, seedling
vigor index, shoot length, root length, fresh seedling weight, dry seedling weight, shoot elongation
rate, root elongation rate, relative water content, chlorophyll content, and membrane stability index,
as compared to control. The highest effect on the examined parameters was achieved by
osmopriming and hormopriming in both cultivars, suggesting that these treatments could be used
to improve the heat stress tolerance of garden pea, after extensive field trials

AB  - Backgraund: The presence or absence of fungi on the seed surface is an important aspect that determines the quality of seeds. Since the data on the presence of Fusarium spp. and other seed-borne pathogens on vegetable pea in Serbia are scarce, the aim of this study was to isolate seed-borne pathogens and identify the disease-causing Fusarium spp. from the infected pea seed.
Methods: Colony morphology and microscopic characters of isolated Fusarium species were recorded from the cultures grown on PDA and CLA, respectively. In order to confirm the morphological identification at the species level, a single-spore culture of the representative isolates was subjected to genomic DNA extraction and gene amplification using the translational elongation factor 1α(TEF-1α) region. The PCR products were sequenced and the sequences were compared against the GenBank nucleotide database by using the BLAST alignment.
Result: Based on morphological and microscopic characteristics, as well as molecular identification by sequencing the TEF gene, the presence of Fusarium equiseti and F. proliferatum was confirmed on the representative isolates Ps18, Ps19 and Ps1, Ps36 obtained from vegetable pea seeds, respectively. According to our knowledge and research, this is the first report of F. equiseti and F. proliferatum on vegetable pea seeds in Serbia.

AB  - Nanopriming is a newly developed seed technology that improves seed germination, initial plant growth, and crop yield by enabling plants to withstand a variety of abiotic stresses. The objective of this study was to evaluate the effectiveness of comprehensive metal-based (Co, Mn, Cu, Fe, Zn, Mo, and Se) nanopriming as compared to hydro- and non-primed seeds of three different pea cultivars in a germination test. Seed priming with nanoparticles (NPs) improved field pea quality via significant increase in germination energy (cv. E-244), final germination (cv. E-244, cv. Dukat), shoot length (cv. E-244, cv. Partner), root length (cv. E-244, cv. Dukat, cv. Partner), fresh shoot weight (cv. Partner), dry shoot weight (cv. Partner), seedling vigor index (cv. E-244, cv. Partner), and chlorophyll content (cv. Dukat, cv. Partner), as compared to both hydropriming and the control. Moreover, nanopriming led to significant improvements in shoot length, fresh shoot length, dry shoot length, seedling vigor index (cv. Dukat), and dry root weight (cv. E-244) as compared to the control only. In general, the highest effect on the examined parameters was achieved by nanopriming, indicating that this treatment may be utilized to raise field pea quality performance. To optimize the method, it is necessary to conduct extensive laboratory and field trials.

AB  - Salinity is one of the greatest challenges of successful agricultural production worldwide. However, seed priming might be efficient practice for enhancing seed germination and initial plant growth and development. This experiment was undertaken with the aim of assessing the impact of different priming methods on seed quality parameters and early growth of garden pea (Pisum sativum L.) cultivars under salinity stress. Pea seeds of three different cultivars were primed with water (hydropriming), KNO3 solution (0.5%), and PEG 6000 solution (-0.5 MPa) for 24h in the dark. Unprimed and primed seeds were germinated between filter papers saturated with 120 mM NaCl using the germination test. The results clearly showed that the examined parameters of primed pea seeds were substantially greater than the parameters of unprimed seeds under saline stress. Moreover, seed priming with KNO3 and PEG solutions were more effective in improving seed quality and initial growth in garden peas than hydropriming.

abstract = "Salinity is one of the greatest challenges of successful agricultural production worldwide. However, seed priming might be efficient practice for enhancing seed germination and initial plant growth and development. This experiment was undertaken with the aim of assessing the impact of different priming methods on seed quality parameters and early growth of garden pea (Pisum sativum L.) cultivars under salinity stress. Pea seeds of three different cultivars were primed with water (hydropriming), KNO3 solution (0.5%), and PEG 6000 solution (-0.5 MPa) for 24h in the dark. Unprimed and primed seeds were germinated between filter papers saturated with 120 mM NaCl using the germination test. The results clearly showed that the examined parameters of primed pea seeds were substantially greater than the parameters of unprimed seeds under saline stress. Moreover, seed priming with KNO3 and PEG solutions were more effective in improving seed quality and initial growth in garden peas than hydropriming.",

AB  - The objective of this study was to examine the effects of different priming treatments on seed quality and initial growth and development of garden pea. For this purpose, three garden pea cultivars, developed at the Institute of Field and Vegetable crops, Novi Sad were examined. The laboratory experiment was conducted under optimal conditions. Seeds were primed in water (hydropriming), 0.5% KNO3 solution, and -0.49 MPa PEG solution for 24 hours; non-primed seeds were controls. The results showed that the percentage of germination, shoot and root length, and mean germination rate significantly increased after the tested priming treatments, while the percentage of abnormal seedlings and mean germination time were significantly decreased after the priming treatments compared to the control. The increase of fresh and dry seedling biomass was significant only in cv.1 and cv.2. Despite the genetic diversity of pea cultivars, the results indicated that the examined seed priming treatments enhanced seed quality and vigour of garden pea cultivars.
AB  - Cilj ovog istraživanja bio je da se ispitaju efekti različitih tretmana prajminga na kvalitet semena i početni rast i razvoj povrtarskog graška. U tu svrhu ispitane su tri sorte povrtarskog graška stvorene u Institutu za ratarstvo i povrtarstvo u Novom Sadu. Laboratorijski ogled je izveden u optimalnim uslovima. Seme je potapano u vodu (hidroprajming), 0,5% rastvor KNO3 i -0,49 MPa rastvor PEG tokom 24 sata; netretirano seme je bilo kontrola. Rezultati su pokazali da su se procenat klijanja, dužina nadzemnog dela i korena izdanaka, kao i srednja stopa klijanja značajno povećali u ispitivanim tretmanima prajminga, dok su se procenat atipičnih izdanaka i srednje vreme klijanja značajno smanjili u tretmanima prajminga u poređenju sa kontrolom. Povećanje sveže i suve biomase izdanaka bilo je značajno samo kod sorti cv.1 i cv.2. Uprkos genetskoj raznolikosti sorti graška, ovi rezultati su pokazali da su ispitivani tretmani prajminga semena efikasne metode u smislu povećanja kvaliteta i vigora semena sorti povrtarskog graška.

AB  - A wide occurrence of wilt and rot caused by Fusarium species has been observed in the last few years in many vegetable-growing areas in the Republic of Serbia. Symptoms of rot and wilt of celery, garlic, and onion were noticed and after determination, Fusarium spp. was identified as the main causal agent of the disease. Most of the Fusarium species that cause disease on vegetable crops (garlic, onion, celery) overwinter in soil debris and soil. This study was conducted to isolate and identify Fusarium from soil samples by conventional and molecular methods. Soil samples were taken under the diseased plant, and 25 samples were collected in order to perform isolation of Fusarium species that cause rot and wilt during production.

AB  - Sunflower (Helianthus annuus L.) is one of the most important oil crops worldwide, including Serbia, where it is grown on about 200,000 ha with an average yield of 3 t/ha. Botrytis cinerea Pers. (1794) regularly occurs on sunflower seeds associated with the phenology of the host plant. However, sunflower seed is often affected by the Rhizopus spp., which causes similar symptoms on seed and seedlings as B. cinerea, and it is important to reliably differentiate those species based on morphological and molecular characteristics. The method used in this study for the detection of B. cinerea is described by ISTA method 7-003. The main task of this study was to isolate the causal agent of grey mould and rot of sunflower seed and seedlings. Infected seeds were covered with overgrown dark-grey mycelia with sporulation. In order to isolate the pathogen, the infected seed was transferred onto potato dextrose agar (PDA) and incubated for 7 days at 20°C. For morphological identification, 11 isolates were single-spored and subcultured onto PDA. Strains formed straight, noddy conidiophores, branched at the top and resembling a cluster with conidia. Conidia were ovoid or elliptical avg. 11.6 × 7.5 µm in size.

AB  - Povrće je važan deo ljudske ishrane jer ono prvenstveno sadrži materije korisne za ljudsko zdravlje kao što su: vitamini, minerali, biljna vlakna, antioksidansi, organske kiseline i dr. U svetu je poznato oko hiljadu vrsta povrća, od kojih se gaji oko 150, a najširu upotrebu ima 30-50 vrsta, koliko je zastupljeno u ishrani u Srbiji (Gvozdanovic-Varga i sar., 2016). U Institutu za ratarstvo i povrtarstvo Novi Sad do sada je stvoreno više od 50 sorti povrća priznatih u zemlji i delom u inostranstvu. Sortiment povrća Odeljenja za povrtarske i alternativne biljne vrste čine nove sorte povrća, kao i odomaćene sorte, te se proizvodi seme 27 povrtarskih vrsta i 10 vrsta cveća. Pored rada na novim sortama povrća koje se odlikuju visokim prinosom, vodilo se računa da sorte svojim izgledom i ukusom zadovoljavaju zahteve probirljivog tržišta i da su prilagođene našim agroklimatskim uslovima (Gvozdenovic i sar., 2008). Zadatak ovog rada je da se prikažu karakteristike novopriznatih sorti povrća, koje su dobijene kao rezultat višegodišnjeg rada, u cilju zadovoljenja potreba proizvođača i potrošača za novim, atraktivnim i kvalitetnim sortama
povrtarskih vrsta.

AB  - Seeds are an important source of primary inoculum for the development of many diseases. Due to the very short vegetation period, controlling the causative agent of the disease on the radish is very difficult. Considering this fact, it is necessary to provide sufficient quantities of seeds of adequate quality and health. The aim of this study was to determine the mycopopulation of radish seeds during different years of production. Seeds used for testing were collected during the period between 2017-2021 from different varieties and localities. After conducting the study, the infection index ranged from 1 to 7%. Fungi of the following genus are represented on the seed in different intensities: Alternaria spp., Fusarium spp., Penicillium spp., Rhizopus spp. and the species Aspergillus niger.

AB  - Seed as a highly-valuable resource is preserved in collections for many years. Although the seed is kept under optimal conditions, monitoring of germination and the presence of fungi during seed preservation is of great importance. Therefore, the aim of this paper was to examine the seed health status and germination of forty-three onion accessions kept in the timespan of 15 years in the Institute of Field and Vegetable Crops collection. Germination of seed samples varied from 7-93%. The presence of fungi in the collection was determined on thirty-three tested samples. Fungi from the genera Acremonium, Alternaria, Aspergillus, Cladosporium, Epicoccum, Fusarium and Penicillium were developed. The following Fusarium species identified on the seeds were F. proliferatum, F. graminearum, F. sporotrichoides, F. solani, F. pseudograminearum and F. equiseti. Based on factor analysis, Fusarium and Penicillium were the species which affected germination, while the occurrence of Alternaria species on onion seed is connected to the year of harvest.

AB  - Xanthomonas campestris pv. campestris (Xcc) is a harmful pathogen of the Brassicaceae agricultural plants in Serbian agro-ecological conditions. The bacterium survives in a soil, plant residues, seeds, wild, and weedy plants. The aim of this study was to characterize Xcc isolates that persist in field soil and seeds where they represent a risk to cause disease of Brassicaceae plants. Collection of soil samples was carried out in the period 2015 - 2017 in four locations regions Vojvodina with cabbage cultivation in monoculture or with crop rotations. The cabbage seed samples were collected in the same period in Futog and Despotovo locations (agricultural farms) and in local farmer markets. All isolations were done on SX and YDC agar plates with the addition of cycloheximide with 250 mg/l. Based on the pathogenicity tests, morphological and biochemical characteristics, 28 identified Xcc isolates were selected, 12 from soil and 16 from seeds. Molecular confirmation of the Xcc isolates was done using multiplex PCR (m-PCR) with Xcc-specific primer sets DLH 120/125 and Zup 2309/2310. In all selected isolates and in the reference Xcc strain NCPPB1144 the expected 370 and 619 bp PCR fragments were amplified. Characterization of isolates by rep-PCR fingerprinting using BOX A1R and (GTG)5 primers resulted in informative patterns. Cluster analysis grouped the strain Xcc NCPPB 1144 together with all Xcc isolates, while other tested Xanthomonads were clustered separately. Based on the similarity level of 90%, we obtained three out of 12 as representative isolates from soil and four out of 16 representative seeds isolates. The characterization of Xcc isolates from soil and seeds will enable their comparison with those pathogenic isolated from leaves of Brassicaceae plants.

AB  - Seme je veoma važan resurs koji se čuva u kolekciji u višegodišnjem periodu. Iako se seme skladišti u optimalnim uslovima, veoma je važno pratiti klijavost i njegovo zdravstveno stanje. Cilj ovog rada je da se ispita zdravstveno stanje i klijavost četrdeset tri uzorka semena crnog luka, koji su čuvani u kolekciji Instituta za ratarstvo i povrtarstvo 1–15 godina. Klijavost crnog luka varirala je u opsegu 7–93%. Na trideset tri uzorka je identifikovano prisustvo gljiva iz sedam rodova: Acremonium, Alternaria, Aspergillus, Cladosporium, Epicoccum, Fusarium i Penicillium. U okviru roda Fusarium utvrđeno je prisustvo vrsta F. proliferatum, F. graminearum, F. sporotrichioides, F. solani, F. pseudograminearum i F. equiseti. Na osnovu faktorske analize vrste koje su uticale na klijavost su Fusarium i Penicillium, dok je pojava vrsta iz roda Alternaria povezana sa godinom ubiranja semena.

The following Fusarium species identified on the seeds were F. proliferatum, F. graminearum, F. sporotrichioides, F. solani, F. pseudograminearum and F. equiseti. Based on factor analysis, Fusarium and Penicillium affected germination, while the occurrence of Alternaria species on onion seed is connected to the year of harvest., Seme je veoma važan resurs koji se čuva u kolekciji u višegodišnjem periodu. Iako se seme skladišti u optimalnim uslovima, veoma je važno pratiti klijavost i njegovo zdravstveno stanje. Cilj ovog rada je da se ispita zdravstveno stanje i klijavost četrdeset tri uzorka semena crnog luka, koji su čuvani u kolekciji Instituta za ratarstvo i povrtarstvo 1–15 godina. Klijavost crnog luka varirala je u opsegu 7–93%. Na trideset tri uzorka je identifikovano prisustvo gljiva iz sedam rodova: Acremonium, Alternaria, Aspergillus, Cladosporium, Epicoccum, Fusarium i Penicillium. U okviru roda Fusarium utvrđeno je prisustvo vrsta F. proliferatum, F. graminearum, F. sporotrichioides, F. solani, F. pseudograminearum i F. equiseti. Na osnovu faktorske analize vrste koje su uticale na klijavost su Fusarium i Penicillium, dok je pojava vrsta iz roda Alternaria povezana sa godinom ubiranja semena.",

AB  - Hyssop (Hyssopus officinalis L.) is a perennial polymorphous plant species with essential oil mainly accumulated in the flowers and leaves. It is grown in Serbia for the needs of pharmaceutical companies and tea production, because of its quality and chemical composition. During a routine quality control of hyssop seeds collected from Rumenka (Vojvodina Province), in 2018, fungal infection followed by seed rot was noticed on an average of 22%. Infected seeds were covered with white mycelium followed with violet pigmentation occurring under the seeds. The presence of Fusarium spp. was confirmed with microscopic observation. Isolation was done aseptically by arranging infected seeds onto surface of potato dextrose agar (PDA), and incubated at 25 °C with a 12-h photoperiod (Mathur and Kongsdall, 2003). After seven days, 12 Fusarium spp. isolates were designated as JBL 4003/1 - 4003/12. Pathogenicity test was performed in vitro using a modified agar slant method in the test tube with PDA amended. After 10 days, fungal mycelia of tested isolates caused seed rot and seedling decay, like naturally infected hyssop seeds. All isolates were re-isolated and sub-cultured on Potato Dextrose Agar (PDA) and Carnation Leaf Agar (CLA) using a hyphal tip transfer technique, fulfilling Koch's postulates. Isolate JBL 4003/1 was distinguished based on pathogenicity and cultural characteristics. It caused seed rot after four days, on PDA colony was fast growing reaching 6-8 cm in diam. in five days, forming abundant, whitish to peach aerial mycelium followed with beige to light brown pigmentation in agar.

AB  - The competitiveness of Bradyrhizobium japonicum inoculation strain against indigenous rhizobia was examined in a soil pot experiment. The effect of inoculation strain was evaluated under different soil conditions: with or without previously grown soybean and applied commercial inoculant. Molecular identification of inoculation strain and investigated rhizobial isolates, obtained from nodules representing inoculated treatments, was performed based on 16S rDNA and enterobacterial repetitive intergenic consensus (ERIC) sequencing. Inoculation strain showed a significant effect on the investigated parameters in both soils. Higher nodule occupancy (45% vs. 18%), nodule number (111% vs. 5%), nodule dry weight (49% vs. 9%), shoot length (15% vs. 7%), root length (31% vs. 13%), shoot dry weight (34% vs. 11%), shoot nitrogen content (27% vs. 2%), and nodule nitrogen content (9% vs. 5%) was detected in soil without previously grown soybean and applied commercial inoculant. Soil had a significant effect on the shoot, root and nodule nitrogen content, while interaction of experimental factors significantly altered dry weight and nitrogen content of shoots, roots and nodules, as well as number of nodules. Nodulation parameters were significantly related with shoot dry weight, shoot and nodule nitrogen content. Symbiotic performance of inoculation strains in the field could be improved through co-selection for their competitiveness and effectiveness.

AB  - Background: Bacillus spp., known to promote growth and reduce disease of various field and vegetable crops, are frequently found in soils. The objective of the study was to select effective Bacillus spp. isolates with multiple plant growth properties and antifungal activities and to examine their effect on germination of soybean. Bacterial isolates were screened for production of indole-3-acetic acid (IAA) and siderophores and solubilization of phosphate. The ability of bacterial isolates to inhibit the growth of seven phytopathogenic fungi affecting soybean was determined using a dual plate assay. Bacillus spp. were further selected and examined in a seed germination test. All Bacillus spp. isolates were positive for IAA production, while siderophore production and P-solubilization were observed in 80% and 20% bacterial isolates, respectively. Bacillus spp. exhibited the highest antifungal activity against Diaporthe caulivora, followed by Diaporthe sojae, Diaporthe eres, Diaporthe longicolla and Macrophomina phaseolina and the least antagonistic effect toward Fusarium graminearum and Fusarium subglutinans. Selected isolates of B. subtilis significantly affected final germination, shoot length, root length, shoot dry weight and root dry weight of two soybean cultivars. The most effective Bacillus spp. isolates could be used as potential inoculants for improving soybean productivity.

AB  - Bio-priming is a new technique of seed treatment that improves seed germination, vigor, crop growth and yield. The objective of this study was to evaluate the effectiveness of Bradyrhizobium japonicum (commercial strains) and Bacillus megaterium (newly isolated strains) as a single inoculant and co-inoculant during seed bio-priming to improve seed germination and initial seedling growth of two soybean cultivars. The treated seeds were subjected to germination test (GT), cold test (CT) and accelerated aging test (AAT). B. megaterium significantly improved all parameters in GT and CT; final germination, shoot length, root length, root dry weight, and seedling vigor index in AAT, as compared to control. In addition, co-inoculation significantly increased all parameters except shoot dry weight in GT; all parameters in CT; germination energy, shoot length, root length, and seedling vigor index in AAT, in comparison to the control. Moreover, Br. japonicum significantly improved the germination energy, shoot length, shoot dry weight, root dry weight, and seedling vigor index in GT; all parameters in CT; shoot length, root length, and seedling vigor index in AAT, compared with non-primed seeds. Thus, B. megaterium strains could be used in soybean bio-priming as a potential single inoculant and co-inoculant, following proper field evaluation.

The control of sunflower pathogens is conditioned by the limited management strategies, and it is focused on genetic resistance and chemical treatments. However, the excessive usage of agrochemicals leads to the loss of soil fertility and gradual decrease of microbial diversity, which is subsequently reflected in reduced growth and yield. For as much, the use of Bacillus spp. is becoming more frequent in modern agriculture as an environmentally sustainable and safe alternative to synthetic pesticides and fertilizers. Spore-forming Bacillus spp. are well known to protect plants from seed or soil-borne pathogens by the synthesis of various metabolites with antifungal activity, such as hydrolytic enzymes and antibiotics. These bacteria are also reported to enhance plant growth through the phytohormone production and nutrient acquisition. This study aimed to select the most effective Bacillus spp. from a group of ten antagonistic strains by antifungal activity assay.
