First Report of Brown Spot on Stored Apple Fruits Caused by Stemphylium vesicarium in Serbia

AB  - Apple is one of the most economically important fruit crops worldwide, and postharvest fungal diseases can cause significant losses during storage (Petreˇs et al. 2020). Apple fruits (cv. Fuji) with necrosis symptoms were collected during the fall of 2022 from an ultra-low-oxygen cold storage facility in Titel, Serbia. The fruits originated from the apple orchards in Titel (45°12947.10N, 20°15923.60E). The pathogens were isolated from collected fruit samples using standard phytopathological techniques. Fruits were surface-sterilized, rinsed with sterile water, and aseptically cut in half, and small fragments collected from the border of healthy and diseased tissue were placed on potato dextrose agar (PDA) medium in Petri dishes and incubated at 25 ± 1°C in the dark for 7 days. The obtained 11 isolates were identified to the genus level as Alternaria (incidence, 46%), Penicillium (36%), Fusarium (9%), and Stemphylium (9%) based on their morphological characteristics. Pathogenicity of all the isolates was confirmed on apple fruits of cultivars Fuji and Golden Delicious. The fruits were surface-sterilized, sprayed with 5 ml of conidial suspension (1 × 105 conidia/ml), and incubated at room temperature for 21 days. Symptoms developed on inoculated fruits were the same as symptoms observed on apple fruit samples collected from cold storage.

AB  - Sugar beet is one of the most profitable crops in Serbia. The charcoal root rot is an emerging disease of sugar beet that significantly reduces yield and affects the economic viability of production. The objectives of this research were to determine the antagonist activity of selected Trichoderma harzianum isolates (T1 to T13) on two Macrophomina phaseolina isolates in vitro using a dual culture test and a volatile compounds test, as well as in vivo. The efficacy of these treatments was compared to two commercial biopesticides (Bacillomix® Original and Trifender Pro®) and one synthetic fungicide (Funomil 700 WG®). The strongest inhibition in the dual culture test was observed in the treatment with T. harzianum T2 against both tested M. phaseolina isolates. Also, the volatile compounds produced by isolates T2 and T12 exhibited the strongest inhibitory effect on M. phaseolina isolates (from 48.22 to 62.75%). Differences in efficacy were found not only between tested Trichoderma isolates, but also between M. phaseolina isolates used for inoculation which indicates different susceptibility of pathogen population to bioagent. The in vivo test confirmed the strong antagonistic effect of the T. harzianum T2 and T12 isolates. Bacillomix® Original, Trifender Pro® and Funomil® 700 WG did not express satisfactory effects in pathogen control.

AB  - Ciljevi ovog rada su bili da se utvrdi dominantni prouzrokovač truleži korena šećerne repe, da se detektovanjem razlika u patogenim, morfološkim i molekularnim karakteristikama proširi ukupno znanje o prouzrokovaču ugljenaste truleži, zatim da se izoluje biološki agens iz zemljišta i izvrši njegova molekularna identifikacija do nivoa vrste, kao i da se u in vitro i in vivo ispitivanjima utvrdi potencijal biološkog agensa za dalju primenu u suzbijanju ovog štetnog oboljenja korena šećerne repe.
AB  - The goals of this work were to determine the dominant causal agent of sugar beet root rot, to expand overall knowledge about the causal agent of charcoal rot by detecting differences in pathogenic, morphological and molecular characteristics, to isolate the biological agent from the soil and conduct molecular identification, as well as to test the potential in vitro and in vivo for further use in the control of this important sugar beet pathogen.

AB  - Cilj ovog rada je bio da se u datim agroekološkim uslovima ispitaju dinamika nakupljanja suve materije u zrnu i visina prinosa u zavisnosti od hibrida kukuruza različite dužine vegetacije. Ogled je postavljen po RCB dizajnu sa 12 hibrida, u četiri ponavljanja, na oglednom polju Rimski Šančevi, Instituta za ratarstvo i povrtarstvo. Praćenje dinamike nakupljanja suve materije u zrnu je vršeno u periodu od svilanja do fiziološke zrelosti, na svaka tri dana, za svaki hibrid. Prikupljeni podaci o dinamici suve materije u zrnu su analizirani u programu SegReg. Na osnovu vremenske analize, kod svih hibrida je nakupljanje suve materije pratilo linearni trend do maksimuma tj. prelomne tačke, od koje se sadržaj suve materije nije menjao. Kod najranijeg hibrida (NS-1) je maksimalni sadržaj suve materije (0.255g) postignut 39 dana nakon svilanja (DNS), dok je kod najkasnijeg hibrida (NS-12) maksimalni sadržaj suve materije iznosio 0.249g i postignut je 42 DNS.
AB  - The aim of this study was to examine the dynamics of dry matter accumulation in the grains of maize hybrids of different maturity groups and their yield in the provided agroecological conditions. The trial was organised as a RCB design, with 12 hybrids in four replications, and was conducted at the Rimski Šančevi experimental field of Institute of Field and Vegetable Crops in Novi Sad (IFVCNS). The progression of dry matter accumulation in the grain was monitored in the period from silking to physiological maturity, at three-day intervals, for each hybrid. Collected data were analyzed in the SegReg program. The dry matter accumulation followed a linear trend until the breaking point (linear plateu regression), from which its content remained stable. The earliest hybrid (NS-1) achieved the maximum dry matter content (0.255g) at 39 days after silking (DAS), while the latest hybrid (NS-12) reached its maximum dry matter content (0.249g) at 42 DAS.

AB  - Primena fungicida u suzbijanju zemljišnih patogena ne predstavlja efikasnu i ekonomski opravdanu meru. Takođe, oni mogu da naruše balans korisnih mikroorganizama u zemljištu, dok njihove rezidue utiču na zagađenje životne sredine. U nastojanju da se smanji upotreba sintetičkih fungicida, primena bioloških agenasa može imati važnu ulogu u „eco-frendly“ poljoprivrednim sistemima. Biološki agensi kao što su vrste iz rodova Bacillus i Trichoderma se najčešće upotrebljavaju u biološkoj borbi protiv zemljišnih patogena. Cilj ovog istraživanja je bio da se ispita antagonistički uticaj Bacillus sp. i Trichoderma spp. na izolate Macrophomina phaseolina (ŠR55(3)/09 i ŠR23/11) u in vitro uslovima. Za ispitivanje je korišten Bacillus sp., 74 koji je komercijalno dostupan kao mikrobiološko đubrivo pod nazivom Bacillomix® Original (Bacillomix Co., Srbija). Metodom inkorporacije komercijalne suspenzije mikrobiološkog đubriva u KDA hranljivu podlogu ispitane su koncentracije preparata od 1, 2, 5 i 10%. U cilju ispitivanja osetljivosti M. phaseolina prema Trichoderma spp. korišteno je 13 izolata koji su predhodno identifikovani kao Trichoderma harzianum. Test dvojne kulture (TDK) je korišten kako bi utvrdili antagonistički uticaj ispitivanih izolata.

Za ispitivanje je korišten Bacillus sp., 74 koji je komercijalno dostupan kao mikrobiološko đubrivo pod nazivom Bacillomix® Original (Bacillomix Co., Srbija). Metodom inkorporacije komercijalne suspenzije mikrobiološkog đubriva u KDA hranljivu podlogu ispitane su koncentracije preparata od 1, 2, 5 i 10%. U cilju ispitivanja osetljivosti M. phaseolina prema Trichoderma spp. korišteno je 13 izolata koji su predhodno identifikovani kao Trichoderma harzianum. Test dvojne kulture (TDK) je korišten kako bi utvrdili antagonistički uticaj ispitivanih izolata.",

AB  - Sugar beet is susceptible to a number of root rot diseases, and they are regularly occurring in Serbia. Economically most important pathogens are Macrophomina phaseolina and Fusarium spp.. Macrophomina phaseolina is predominantly found in isolates from sugar beet, especially during dry and hot summers. For more than thirty years already, our team of researchers follows the occurrence and etiology of sugar beet root rot in Serbia. The aim of this experiment was to evaluate the effect of mineral nutrition on root rot in commercial sugar beet cultivars.

AB  - Charcoal rot of sugar beet, caused by Macrophomina phaseolina in recent years has become a main concern for farmers in Serbia. Since control measures are mostly preventive with only partial effect, introduction of alternative measures, such as biological control based on augmentation of mycoparasites, represents the most promising approach. ITS region and tef gene sequence analysis of ten Trichoderma spp. isolates from sugar beet and soybean rhizosphere was done to confirm species identity. All isolates were confirmed to be Trichoderma harzianum.

However, other factors influence its incidence as well, some of which are the choice of cultivar and plant nutrition. These factors have been put together in field trial, which tested reaction of eight sugar beet genotypes (G1-G8) and 20 variants of NPK nutrition.

AB  - Cercospora leaf spot caused by the fungus Cercospora beticola is the most significant foliar disease of sugar beet in Serbia. Measures to reduce damage of this disease are based on the use of tolerant varieties, crop rotation and fungicide application. Climate changes and the resistance of Cercospora beticolastrains to fungicides from the strobilurin group further complicate the fight against this disease. In extremely favourable years for the development of disease with high temperatures and precipitation during the summer months, growers in Serbia perform 4-8 fungicide treatments.

AB  - There are three main yellowing viruses that damage the beet crop in Europe: Beet yellows virus (BYV), Beet mild yellowing virus (BMYV) and Beet chlorosis virus (BChV). Although not a true yellowing virus, Beet mosaic virus (BtMV) is found in sugar beet in Europe and can cause damage. The yellowing viruses are transmitted by aphids. The recent ban of neonicotinoids in the EU endangered sugar beet production in terms of crop protection against aphids. Since, Republic of Serbia agenda is to become member state of the EU, in order to do that it is necessary to completely synchronise legislation with the EU in all domains, including agriculture and crop protection.

AB  - Aspergillus flavus is the main producer of aflatoxin B1, one of the most toxic contaminants
of food and feed. With global warming, climate conditions have become favourable for aflatoxin
contamination of agricultural products in several European countries, including Serbia. The
infection of maize with A. flavus, and aflatoxin synthesis can be controlled and reduced by
application of a biocontrol product based on non‐toxigenic strains of A. flavus. Biological control
relies on competition between atoxigenic and toxigenic strains. This is the most commonly used
biological control mechanism of aflatoxin contamination in maize in countries where aflatoxins
pose a significant threat. Mytoolbox Af01, a native atoxigenic A. flavus strain, was obtained from
maize grown in Serbia and used to produce a biocontrol product that was applied in irrigated and
non‐irrigated Serbian fields during 2016 and 2017. The application of this biocontrol product
reduced aflatoxin levels in maize kernels (51–83%). The biocontrol treatment had a highly
significant effect of reducing total aflatoxin contamination by 73%. This study showed that
aflatoxin contamination control in Serbian maize can be achieved through biological control
methods using atoxigenic A. flavus strains.

AB  - Hot water treatments of apple fruits prior to long-term storage have a high potential as a physical measure against apple fruit
storage rot caused by plant pathogenic fungi. Fusarium species are increasingly found as causal agents of postharvest apple
fruit rot. The effect of high temperatures on casual agents of Fusarium fruit rot, F. avenaceum and F. graminearum, was tested
in this article. In in vitro assay, spore suspensions of F. avenaceum and F. graminearum were exposed to high temperatures
(ranging from 45 to 90 °C) at different duration (ranging from 30 s to 20 min) and the treatments that significantly inhibited
mycelial growth of the isolates (53 and 57 °C for 3 and 5 min) were tested in vivo by hot water treatment of inoculated apple
fruits of Fuji cultivar. The obtained results showed that the most promising hot water treatment should be between 53 and
57 °C and between 3 and 5 min of the exposure period.

AB  - Emerging mycotoxins such as moniliformin (MON), enniatins (ENs), beauvericin (BEA), and fusaproliferin (FUS) may contaminate maize and negatively influence the yield and quality of grain. The aim of this study was to determine the content of emerging Fusarium mycotoxins in Serbian maize from the 2016, 2017, and 2018 harvests. A total of 190 samples from commercial maize production operations in Serbia were analyzed for the presence of MON, ENs, BEA, and FUS using liquid chromatography-tandem mass spectrometry (LC-MS/MS). The obtained results were interpreted together with weather data from each year. MON, BEA, and FUS were major contaminants, while other emerging mycotoxins were not detected or were found in fewer samples (<20%). Overall contamination was highest in 2016 when MON and BEA were found in 50–80% of samples. In 2017 and 2018, high levels of MON, FUS, and BEA were detected in regions with high precipitation and warm weather during the silking phase of maize (July and the beginning of August), when the plants are most susceptible to Fusarium infections. Since environmental conditions in Serbia are favorable for the occurrence of mycotoxigenic fungi, monitoring Fusarium toxins is essential for the production of safe food and feed.

AB  - Recently, garlic rust may have developed into a significant problem for our agro-ecological climate. Evaluation trials of winter garlic rust occurrence and the disease severity have been conducted in five locations. Intensity of infection was evaluated 3 times. Simultaneously, leaf samples were collected for microscopic examination. In all locations during third decade of April individual uredosori sporadically occurred in garlic leaves, relatively early compared to its previous years occurrence. The highest rust rate was in third evaluation, when majority of plants were completely covered with uredosoris. Microscopic examination of uredosori, in all samples revealed physiologically mature urediniospores indicating a number of secondary infections.
AB  - Rđa na belom luku u našim agroekološkim uslovima postaje sve značajnije oboljenje, koje se pojavljuje intenzivnije u poslednjih nekoliko godina. Ispitivanja pojave i intenziteta napada rđe na jesenjem belom luku, sprovedena su na pet lokaliteta u 2014. godini. Ocena intenziteta infekcije vršena je tri puta, a istovremeno su prikupljani uzorci listova za mikroskopska ispitivanja. Pojava pojedinačnih uredosorusa na listovima luka primećena je relativno rano, u odnosu na prethodne godine, u trećoj dekadi aprila. Pojava patogena utvrđena je na svim ispitivanim lokalitetima. Najveći intenzitet zaraze zabeležen je prilikom trećeg pregleda, kada su ustanovljene biljke potpuno prekrivene uredosorusima parazita. Mikroskopskim pregledom uredosorusa, kod svih prikupljenih uzoraka u različitim terminima, utvrđene su fiziološki zrele uredospore, što je ukazalo na više sekundarnih infekcija.
