Hidrolat Nepeta cataria L. - uticaj na gajene i korovske vrste

AB  - Kako bi se smanjili neželjeni efekti prekomerne upotrebe herbicida u poljoprivredi teži se primeni alternativnih, ekološki prihvatljivih načina suzbijanja korova. Hidrolati predstavljaju nusproizvode nastale tokom procesa ekstrakcije etarskih ulja. U radu je ispitan efekat hidrolata macine trave (Nepeta cataria L., fam. Lamiaceae) na klijanje semena i rast korenka i stabaoca kukuruza (Zea mays L.), soje (Glycine max (L.) Merr.), štira (Amarantus retroflexus L.) i pepeljuge (Chenopodium album L.). Po 25 semena svake ispitivane vrste je postavljeno u Petri posude na filter hartiju i potom tretirano 10, 20, 50 i 100% hidrolatom N. cataria, odnosno destilovanom vodom u slučaju kontrole. Eksperiment je izvršen u 4 ponavljanja. Semena kukuruza i soje su stavljena u Petri posude prečnika 15 cm i tretirana sa 15 ml hidrolata, dok su semena štira i pepeljuge stavljena u Petri posude prečnika 8 cm i tretirana sa 8 ml hidrolata. Petri posude su potom stavljene u klima komoru gde su 10 dana bile u uslovima 22°C/20°C tokom 8/16-časovnog fotoperioda i pri vlazi od 60±1%. Svakog dana je ocenjivana klijavost tretiranih semena, a po završetku ogleda je merena dužina stabaoca i korenka svih test biljaka.

AB  - The aim of this study was to determine the effect of the hydrolates obtained as the by-products of the Thymus vulgaris essential oil steam distillation process. The bioassays, which were undertaken in order to determine the effect on germination and initial growth of seedlings of some cultivated and weed species, were performed under controlled conditions with different concentrations of the hydrolates. Seeds of Glycine max, Helianthus annuus, Zea mays, Triticum aestivum, Daucus carota subsp. sativus, Allium cepa, Medicago sativa, and Trifolium repens, and six weed species—Amaranthus retroflexus, Chenopodium album, Portulaca oleracea, Echinochloa crus-galli, Sorghum halepense, and Solanum nigrum—were treated with 10, 20, 50, and 100% T. vulgaris hydrolate solution. The obtained results showed that the T. vulgaris hydrolate had the least negative effect on the germination of cultivated species, such as soybean, sunflower and maize, whereas clover and alfalfa were the most sensitive. By comparison, all the tested weed species expressed high susceptibility. It can be concluded that the T. vulgaris hydrolate has an herbicidal effect, in addition to its potential as a biopesticide in terms of integrated weed management.

Determination of tropane alkaloids in corn puffs by the LC-MS/MS

AB  - The interest in tropane alkaloids (TA) as food contaminants is increasing. A sensitive and selective LC-MS/MS method was applied for the analysis of corn puff samples from the Serbian market. Only atropine was quantified in 22% of the samples. In case of scopolamine, although not quantified, it was detected in 22% of the samples. Whether the acute reference dose (ARfD) could be exceeded was checked on a case-by-case basis for the individual products under assessment. Due to their low body weight and relatively high snack consumption, preschool children were at the highest risk of TA exposure. Assuming that the average consumption is 50 g of corn puffs per day, the sample with the highest concentration of TAs (2.05 µg/kg, 1.58 μg/kg of atropine) could contribute with 32.0% to the ARfD, of which 24.7% owing to atropine. If the same amount of corn puffs is consumed by older age classes, corresponding exposure contributions to the ARfD would progressively decline, down to 8.4% for adult population. The study revealed no health risk from TAs exposure through the consumption of the corn puffs in Serbian population.
AB  - Interesovanje za tropanske alkaloide kao kontaminante hrane je u porastu. Za analizu kukuruznog flipsa sa tržišta Srbije primenjena je osetljiva i selektivna LC-MS/MS metoda. Atropin je kvantifikovan u 22% uzoraka. U slučaju skopolamina, koji nije kvantifikovan, detektovano je u 22% uzoraka. Da li može doći do prekoračenja akutne referentne doze provereno je za svaki pojedinačni proizvod koji je uvršten u procenu. Mala telesna masa i relativno velike konzumirane količine snek-proizvoda ističu predškolsku decu kao grupu u najvećem riziku od izloženosti tropanskim alkaloidima. Pod pretpostavkom o konzumiranju 50 grama kukuruznog flipsa dnevno, uzorak s naj­ većom koncentracijom tropanskih alkaloida (2,05 µg/kg, 1,58 mg/kg atropina) može doprineti sa 32,0% od akutne referentne doze, od čega 24,7% odgovara atropinu. Ukoliko istu količinu kukuruznog flipsa konzumiraju starije uzrasne grupe, doprinos sledstvene izloženosti tropanskim alkaloidima bi progresivno opadao do 8,4% za odraslu populaciju. Studija nije ukazala na zdravstveni rizik usled izloženosti tropanskim alkaloidima putem konzumiranja kukuru­ znog flipsa za populaciju u Srbiji.

AB  - This study was undertaken in order to investigate the biological activity of chemically characterized Carum carvi essential oil (EO) and its effect on oxidative stress parameters in stored product pests. Gas chromatography-mass spectrometry depicted carvone (68.22 +/- 0.62%) and limonene (21.80 +/- 0.54%) as the major compounds of said EO. Different concentrations of EO were tested for fumigant activity and showed prominent results following a time-response relationship in both tested species. The changes in the antioxidative defense system, catalase (CAT), superoxide dismutase (SOD), and glutathione-S-transferase (GST) activities, as well as the level of lipid peroxidation (MDA) and the content of reduced glutathione (GSH) were estimated in time-dependent response. The results demonstrated that C. carvi could have a significant role in the future, as an environmentally friendly alternative for the management of stored product pests.

LC-MS/MS Determination of Tropane Alkaloids in Maize Crop

AB  - The undemanding LC-MS/MS method was developed for the synchronized analysis of atropine and scopolamine in maize crop. The dSPE was carried out with 1% acetic acid in acetonitrile/water and a mixture of magnesium sulphate, sodium chloride and sodium citrate. The analytes were separated on a Zorbax XDB C18 column using methanol/water as the mobile phase in gradient mode. The detection was done using a tandem masss spectrometry (MS/MS) in the positive ESI. The tropane alkaloids exhibited excellent linearity in the range of 2-20 µg/kg with the LOQ of 5 µg/kg for maize. The extraction recoveries of atropine and scopolamine were 65.7 and 85.5% with the intraday RSDr 10.25 and 4.29%, respectively. The validated method was appled to real maze samples. One sample contained 18.8 µg/kg of atropine and 6.3 µg/kg of scopolamine.

abstract = "The undemanding LC-MS/MS method was developed for the synchronized analysis of atropine and scopolamine in maize crop. The dSPE was carried out with 1% acetic acid in acetonitrile/water and a mixture of magnesium sulphate, sodium chloride and sodium citrate. The analytes were separated on a Zorbax XDB C18 column using methanol/water as the mobile phase in gradient mode. The detection was done using a tandem masss spectrometry (MS/MS) in the positive ESI. The tropane alkaloids exhibited excellent linearity in the range of 2-20 µg/kg with the LOQ of 5 µg/kg for maize. The extraction recoveries of atropine and scopolamine were 65.7 and 85.5% with the intraday RSDr 10.25 and 4.29%, respectively. The validated method was appled to real maze samples. One sample contained 18.8 µg/kg of atropine and 6.3 µg/kg of scopolamine.",

AB  - The programmes for pesticide residues monitoring in food which are continually carried out in the world ensure that the consumers are not exposed to pesticide residues as well as that only the registered compounds are used in the production. All the member countries of the EU are obliged to observe the official control of pesticide residues in food, in accordance with the Regulations EC 396/2005. At present, in the Republic of Serbia, the programme for monitoring the pesticide residues in food is being prepared as the results of numerous analyses of random samples of fruit, in the market over the past years justify the introduction of such a programme.
AB  - Programi praćenja ostataka pesticida u hrani koji se kontinuirano sprovode u svetu osiguravaju potrošače da ne budu izloženi nepoželjnom nivou ostataka pesticida, kao i da se u proizvodnji koriste samo registrovana jedinjenja. Sve zemlje članice EU su dužne da sprovode zvaničnu kontrolu prisustva ostataka pesticida u hrani, u saglasnosti sa Regulativom EC 396/2005. U Republici Srbiji je u pripremi monitoring program za ostatke pesticida u hrani, a rezultati analiza većeg broja slučajno izabranih uzoraka voća, na našem tržištu, tokom proteklih godina ukazuju na opravdanost uvođenja ovakvog programa.
