Identification and genetic characterisation of xanthomonas campestris pv. Campestris as an oilseed rape pathogen in Serbia

AB  - During 2010, black rot symptoms were observed in oilseed rape plants grown in a commercial plot in Serbia. Ten bacterial isolates obtained from diseased plants, and identified as Xanthomonas campestris pv. campestris (Xcc) based on pathogenicity, physiological and biochemical tests, PTA-ELISA and 16S rDNA sequences analysis, were investigated in detail. Strains were characterized by comparing them by rep-PCR fingerprints using ERIC and (GTG)(5) primers. The 16S rDNA sequences of strains TUr1 and TUr6 were deposited in GenBank under accession Nos. KF057196 and KF057197, respectively. Phylogenetic analysis of the 16S regions showed high similarity level for oilseed rape representative strains and Xcc strains of different origin isolated from kale, cabbage and broccoli.

AB  - Komorač (Foeniculum vulgare Mill.) je aromatična biljka iz familije Apiaceae. Plodovi komorača (Foeniculi fructus) obično sadrže 2-6% etarskog ulja, čija je glavna komponenta trans-anetol koji čini 60-90%. Imaju široku upotrebu u medicini i ishrani, ali i u poljoprivrednoj proizvodnji. Na komoraču je u svetu i kod nas zabeležen veliki broj bolesti, među kojima su gljivične najbrojnije, ali se javljaju i bakterioze i fitoplazme. Utvrđena je i pojava insekata i parazitne cvetnice Cuscuta sp.
AB  - Fennel (Foeniculum vulgare Mill.) is aromatic plant from Apiaceae family. Fennel fruits (Foeniculi fructus) usually contain 2-6% of essential oil, with trans-anethole which is main component with 60-90%. The fruits are widely used in medicine and nutrition, but also in organic agricultural production. In the world and in our country it was found a large number of diseases, including fungal which are more abundant, than bacterial and phytoplasmas on the fennel. It was determined and the appearance of insects and parasitic flower dodder (Cuscuta sp.).

Application of semi-selective mediums in routine diagnostic testing of Pseudomonas savastanoi pv. phaseolicola on common bean seeds

AB  - Halo blight, caused by Pseudomonas savastanoi pv. phaseolicola (Psp), is considered to be an important bacterial disease on common bean (Phaseolus vulgaris L.) in Serbia. Use of pathogen-free seeds is one of the most effective control measures against this disease. The aim of this study was to evaluate a detection method for Psp on untreated common bean seeds (23 genotypes) from commercial crops grown within Serbia. Detection of this pathogen was made by plating onto the modified sucrose peptone (MSP) and Milk Tween (MT) semi-selective mediums from soaked whole common bean seed. Colonies growing on the MSP medium were light yellow, convex and shiny, whereas on the MT medium, they were creamy white, flat and circular. The pathogenicity of the obtained strains was confirmed by the inoculation of germinated bean seed. The isolates recovered from the seed assay were further confirmed to be Psp by using both Enzyme-linked immunosorbent assay (ELISA) and Polymerase Chain Reaction (Nested-PCR) detection methodologies. The International Seed Testing Association (ISTA) method selected for this work was found to be effective in detecting the presence of Psp in common bean seed. The bacterium Psp was detected in only two of the 23 seed samples analyzed by this method, which shows that the bacterium is not widespread in Serbia.

Detection and identification of Xanthomonas axonopodis pv. phaseoli on bean seed collected in Serbia

AB  - Laboratory assays were conducted to detect and identify Xanthomonas axonopodis pv. phaseoli on bean seeds collected from naturally contaminated commercial crops. Semi-selective media, viz. Xanthomonas campestris pv. phaseoli agar and milk tween agar were used to detect the bacterium from whole bean seed extract. Colonies of the bacterium were yellow, mucoid and convex on these media and zones of hydrolysis formed around them. The pathogenicity of the strains was confirmed by stem inoculation of young bean plants. Identification of the strains was proved by using rapid methods (ELISA and PCR). The results showed the presence of seed-borne X. axonopodis pv. phaseoli in 20 out of the 23 assayed bean cultivars that are commercially grown in Serbia. These results indicate that the seed of the bean cultivars grown in our country is highly infected by the bacterium.

Elaboration of methods for detection of Pseudomonas savastanoi pv. phaseolicola on bean seeds

AB  - Pseudomonas savastanoi pv. phaseolicola detection on artificially inoculated bean seeds was investigated. The method of the International Seed Federation - ISF (2006) was used. It includes bacteria extraction from seeds, isolation on semiselective media and checking the pathogenicity of investigated isolates. For verification of results, quick new methods of investigation were used (ELISA test and PCR). The results show that semiselective media MT (Milk Tween Agar) and MSP (Modified Sucrose Peptone Agar) can be appropriate for isolation of this bacterium. Pathogenicity of the investigated isolates was confirmed on cotyledon leaves of bean. ELISA test and PCR confirmed that all investigated isolates and reisolates belong to the bacterium P.s.pv. phaseolicola.
AB  - U radu je ispitivana mogućnost utvrđivanja prisustva bakterije Pseudomonas savastanoi pv. phaseolicola sa veštački inokulisanog semena pasulja. Korišćena je metoda International Seed Federation (ISF), koja uključuje ekstrakciju bakterija iz semena, izolaciju na poluselektivne podloge i proveru patogenosti dobijenih izolata. U cilju potvrde rezultata korišćene su brze savremene metode identifikacije (ELISA test i PCR). Rezultati dobijeni tokom rada pokazuju da su poluselektivne podloge MT (Milk Tween Agar) i MSP (Modified Sucrose Peptone Agar) pogodne za izolaciju ove bakterije. Patogenost odabranih izolata dokazana je na kotiledonim listovima pasulja. Primenom ELISA testova i PCR, potvrđeno je da svi ispitivani izolati i reizolati pripadaju bakteriji P.s.pv. phaseolicola.

ELISA test and PCR confirmed that all investigated isolates and reisolates belong to the bacterium P.s.pv. phaseolicola., U radu je ispitivana mogućnost utvrđivanja prisustva bakterije Pseudomonas savastanoi pv. phaseolicola sa veštački inokulisanog semena pasulja. Korišćena je metoda International Seed Federation (ISF), koja uključuje ekstrakciju bakterija iz semena, izolaciju na poluselektivne podloge i proveru patogenosti dobijenih izolata. U cilju potvrde rezultata korišćene su brze savremene metode identifikacije (ELISA test i PCR). Rezultati dobijeni tokom rada pokazuju da su poluselektivne podloge MT (Milk Tween Agar) i MSP (Modified Sucrose Peptone Agar) pogodne za izolaciju ove bakterije. Patogenost odabranih izolata dokazana je na kotiledonim listovima pasulja. Primenom ELISA testova i PCR, potvrđeno je da svi ispitivani izolati i reizolati pripadaju bakteriji P.s.pv. phaseolicola.",

AB  - Xanthomonas axonopodis pv. phaseoli detection on artificially inoculated bean seeds was investigated. The method of the International Seed Federation - ISF (2006) was used. It included bacteria extraction from seeds and isolation on semiselective media with pathogenicity test of the investigated isolates. ELISA and PCR were used for verification of results. The results showed that the semiselective media MT (Milk Tween Agar) and XCP1 (Xanthomonas campestris pv. phaseoli Agar) were very suitable for isolation of X. a. pv. phaseoli. Pathogenicity was confirmed on young bean plants. ELISA test and PCR confirmed that all investigated isolates and reisolates belong to the bacterium X.a.pv. phaseoli.
AB  - U radu je ispitivana mogućnost utvrđivanja prisustva bakterije Xanthomonas axonopodis pv. phaseoli na veštački inokulisanom semenu pasulja. Ispitivanja su vršena po metodi International Seed Federation (ISF), ekstrakcijom bakterija iz semena i izolacijom na poluselektivne podloge uz proveru patogenosti dobijenih izolata. Race identification of Pseudomonas syringae pv. Glycinea on commercial soybean varieties in Serbia

AB  - Bacterial blight is important and frequent disease of soybean in Vojvodina province. Isolation of the pathogen was carried out from infected soybean leaves of several cultivar grown in Vojvodina province using beef extract and the medium enriched with 5% sucrose. Only the representative isolates were chosen for further investigation. Pathogen city of the obtained isolates including referent strain (National Collection of Plant Pathogenic Bacteria, United Kingdom-NCPPB 3318) was proved by inoculating soybean plants in cotyledon stage (Balkan variety), by spraying with bacterial suspension (conc.108CFU/ml). Pathogen city of studied isolates was also confirmed by infiltration on tobacco leaves. Cultural and biochemical-physiological characteristics were tested. Differential soybean cultivars: Acme, Chippewa, Flambeau, Harosoy, Lindarin, Merit and Norchief were used to determine physiological race of the bacterium. Young plants were inoculated in two ways: by leaves rubbing with sterile cotton swab dipped into the bacterial suspension, and by spraying leaves under the pressure. Based on the results obtained it was shown that strains which cause bacterial blight of soybeans in Vojvodina, belong to Pseudomonas syringe pave. lysine race 4.
AB  - Bakteriozna pegavost soje čiji je prouzrokovač Pseudomonas syringae pv. glycinea (Coerper) Young, u Srbiji se javlja redovno i to često u jakom intenzitetu. Izolacija patogena vršena je sa obolelih delova lista soje različitih sorti. Izolacije su vršene na podlogu od mesnog ekstakta (MPA) i podlogu obogaćenu saharozom (NSA). Patogenost izolata je dokazana pozitivnom hipersenzitivnom reakcijom (HR) na duvanu. Provera patogenosti dobijenih izolata, kao i referentnog izolata ove bakterije (NCPPB 3318), dokazana je i inokulacijom mladih biljaka soje u fazi kotiledona (sorta Balkan), prskanjem i potapanjem u suspenziju bakterija. Odgajivačke i biohemijsko-fiziološke odlike ispitane su standardnim bakteriološkim metodama. Pripadnost dobijenih izolata fiziološkim rasama određena je korišćenjem diferencijalnog sortimenta soje: Acme, Chippewa, Flambeau, Harosoy, Lindarin, Merit i Norchief. Mlade biljke su inokulisane na dva načina: prskanjem pod pritiskom i povredom listova sunđerom natopljenim suspenzijom bakterija. Zaključeno je da izolati bakterije P. s. pv. glycinea, sa soje u Vojvodini, pripadaju rasi 4.

Pseudomonas syringae pv. glycinea economically harmful pathogen of soybean in Vojvodina

AB  - Pseudomonas syringae pv. glycinea (Coerper) Young causal agent of bacterial blight is important and frequent disease of soybean in Vojvodina province. Isolation of the pathogen was carried out during 2005. from infected soybean leaves of several cultivar. Pathogenicity of the obtained isolates including referent strain (National Collection of Plant Pathogenic Bacteria, United Kingdom-NCPPB 3318) was proved by inoculation soybean plants at cotyledon stage (Balkan variety) and by hypersensitive reaction (HR) on tobacco leaves. Cultural and biochemical-physiological characteristics were tested according to known methods. Bacterium identification was also confirmed by serological method (agglutination tests, IF, ELISA,) with appropriate antibodies of P. s. pv. glycinea NEOGEN Europe Ltd., Scotland, UK. All investigated isolates showed positive reaction with mentioned antibodies. Molecular technique (PCR) based on chlorosis inducing phytotoxin coronatine confirm that studied isolates belong to P. s. pv. glycinea. Differential soybean cultivars: Acme, Chippewa, Flambeau, Harosoy, Lindarin, Merit and Norchief were used to determine physiological race of the bacterium. Young plants were inoculated in two ways: by leaves rubbing with sterile cotton swab dipped into the bacterial suspension and by spraying leaves under the pressure. Based on the obtained results it was shown that strains which cause bacterial blight of soybeans in Vojvodina, belonged to P. s. pv. glycinea race 4. Reaction of 12 soybean varieties and two lines (L-220 230, P-9241) from different maturity groups were investigated under natural field infection and under glass house conditions. Statistically significant differences of sensitivity between individual genotypes and lines were determined. Genotypes 000 and 00 maturity group were the most sensitive.

Uzorci su prikupljeni tokom 2005. godine, iz više lokaliteta u Vojvodini. Patogenost dobijenih izolata je dokazana pozitivnom hipersenzitivnom reakcijom (HR) na duvanu, kao i inokulacijom biljaka soje u fazi kotiledona (sorta Balkan). Odgajivačke i biohemijsko-fiziološke odlike ispitane su standardnim metodama. Na osnovu dobijenih rezultata, ispitivani izolati poseduju karakteristike Ia fluorescentne grupe fitopatogenih bakterija roda Pseudomonas. Za razlikovanje P. s. pv. syringae od P. s. pv. glycinea korišćeni su specijalni diferencijalni testovi. Rezultati ukazuju da prvi patogen koristi eritritol, betain, sorbitol i L-laktat, izazivajući nekrozu zelenih plodova višnje i trešnje, dok P. s. pv. glycinea pokazuje negativnu reakciju u pomenutim testovima. Identifikacija bakterija potvrđena je i serološkim testovima (metod aglutinacije, IF, ELISA), korišćenjem odgovarajućih antitela bakterije P. s. pv. glycinea, (NEOGEN Europe Ltd., Scotland, UK). Molekularnom metodom identifikacije (PCR), korišćenjem odgovarajućih prajmera, potvrđeno je da proučeni izolati sa soje pripadaju vrsti P. s. pv. glycinea. Pripadnost dobijenih izolata fiziološkim rasama određena je korišćenjem diferencijalnog sortimenta soje: Acme, Chippewa, Flambeau, Harosoy, Lindarin, Merit i Norchief. Zaključeno je da izolati bakterije P. s. pv. glycinea, sa soje u Vojvodini, pripadaju rasi 4. Reakcija 12 sorti i dve linije soje (L-220 230, P-9241), različitih grupa zrenja, prema P. s. pv. glycinea, ispitana je u uslovima spontane infekcije u polju i staklari. Genotipovi soje, obuhvaćeni ovim proučavanjima, ispoljili su različit nivo osetljivosti prema P. s. pv. glycinea. Postoje statistički značajne razlike osetljivosti između pojedinih genotipova i linija. Genotipovi sa najkraćom vegetacijom (000 i 00 grupa zrenja) pokazali su se kao najosetljiviji.

Zaključeno je da izolati bakterije P. s. pv. glycinea, sa soje u Vojvodini, pripadaju rasi 4. Reakcija 12 sorti i dve linije soje (L-220 230, P-9241), različitih grupa zrenja, prema P. s. pv. glycinea, ispitana je u uslovima spontane infekcije u polju i staklari. Genotipovi soje, obuhvaćeni ovim proučavanjima, ispoljili su različit nivo osetljivosti prema P. s. pv. glycinea. Postoje statistički značajne razlike osetljivosti između pojedinih genotipova i linija. Genotipovi sa najkraćom vegetacijom (000 i 00 grupa zrenja) pokazali su se kao najosetljiviji.",

Rapid identification of phytopathogenic bacteria originating from vegetables by the BIOLOG System

AB  - The purpose of our study was to investigate the possibility of using the BIOLOG System for a rapid identification of various phytopathogenic bacteria (all in all 19 strains) from different vegetable crops. The results of the study have shown that the BIOLOG System indeed enables a rapid identification of numerous strains of phytopathogenic bacteria belonging to the species Pseudomonas syringae, Xanthomonas campestris and Erwinia carotovora. After four, six or twenty - four hours of incubation, identification down to the level of pathovar (Ps.pv. phaseolicola, P.s.pv. syringae, P.s.pv. tomato, X.c.pv. phaseoli and Xc.pv. vesicatoria) or subspecies (E.c.ssp. carotovora and E.c.ssp. atroseptica) was possible for 14 out of 19 investigated strains. For the others, the identification proved correct at the level of species or genus. Some pathovars were wrongly identified.
