Mamlić i sar.

AB - In order to be competitive on the market, the production of biopreparations needs to be optimized, modelled, and assessed in the early stages of its development. The aim of this paper was to optimize medium for the production of Trichoderma harzianum K179 biocontrol agent, to analyze its kinetics at enlarged laboratory scale and finally economic analysis of the production of this high-value product through simulation modelling. The results showed that the bioprocess of T. harzianum K179 bioagent production in a laboratory bioreactor on the medium with optimal composition (dextrose 10 g l−1, soy flour 6.87 g l−1, K2HPO4 1.51 g l−1, KCl 0.5 g l−1, and MgSO4 × 7H2O 0.5 g l−1), at stirring speed of 1.75 × g and aeration intensity of 1.5 vvm, can be shortened from 96 to 36 h. The results of bioprocess economic analysis showed that with a 25-year project lifetime and an investment payback time of 7.58 years, this project represents an economically viable system.

AB - Maize is the most important field crop in Serbia. This agricultural crop, like others, is susceptible to infection by a number of pathogens. Among the most important pathogens, Helminthosporium carbonum and Penicillium spp. were mentioned. Protection of maize from fungal pathogens is mainly controlled by the application of fungicides which are known to have a negative impact on the environment. However, the discovery of new, biological ways of protection would enable a significant reduction of negative effects on the environment and human health. The usage of microorganisms in biological protection is becoming very important, especially in organic and integral production. Trichoderma is certainly a genus that has great potential in protecting plants from fungal pathogens, so the aim of this research was to optimize the medium composition for the production of Trichoderma harzianum biocontrol agent effective against the maize pathogens, H. carbonum and Penicillium sp.

AB - Crop breeding is a continuous process aimed at increasing yields and improving crop resistance to biotic and abiotic stresses. Recently, frequent and often unpredictable variations in climatic and market conditions have led to the fact that classical breeding methods cannot always provide solutions and a timely response to new challenges in agricultural production. With all this in mind, the Institute of Field and Vegetable Crops (IFVCNS) established the Centre of Excellence for Innovations in the Breeding of Climate-tolerant Plants - Climate Crops with the aim of introducing and applying new breeding techniques (NBT), along with efficient phenotyping platforms in crop breeding programs in IFVCNS. It is expected that the activities within Climate Crops will have a positive impact on the excellence and innovative capacities of IFVCNS in the field of breeding plants tolerant to the extreme weather conditions that occur as a result of climate change.

AB - Alfalfa seeds have been of great economic and economic importance since ancient times, because it is considered that alfalfa was the first cultivated fodder plant. Today it is very widespread and is cultivated in more than 80 countries on all continents from the temperate to the tropical zone (Katić et al., 2000).
Alfalfa (Medicago sativa L.) is the most represented forage crop both in the world and in our country, primarily due to the chemical composition of its plant mass. The protein content in the dry matter of alfalfa varies between 18 and 25%, depending on the stage of plant development, cutting, variety and method of utilization. The mineral content is 9.87%, while the calcium content ranges from 1.4 to 1.9%. The content of fatty substances is low, on average 1.78% (Katić et al., 2004).

Oplemenjivanje lucerke u Institutu traje neprekidno
Belorusija, Ukrajina, Turska, Maroko).
očekivati nove, visokoprinosne i visokokvalitetne sorte lucerke. Savremene metoda
populacija diploidne lucerke (M. lessingii Fisch. & C.A.Mey. ex Kar.), što predstavlja prvi
pokušaj povećanja tolerantnosti lucerke na kisela zemljišta. U oplemenjivanju lucerke
Ispitivanje kvaliteta, posebno hranljive vrednosti i
AB - Breeding of alfalfa at the Institute of Field and Vegetables in Novi Sad began after the World
War II, using autochthonous populations, and mass selection, as the oldest breeding method.
The basis of successful alfalfa breeding is a rich genetic collection, which was collected not
only from indigenous populations, but also by the exchange of genotypes with domestic and
foreign research institutions and gene banks, as well as through the participation of researchers in international projects. Alfalfa breeding at the Institute has been going on
continuously for more than six decades, through several cycles, using different breeding
methods, which resulted in the development of 22 alfalfa cultivars in scribed in Serbia and 9
cultivars released abroad (EU, Belarus, Ukraine, Turkey, Morocco). From a global
perspective, the end of the 20 th and the beginning of the 21 st century was marked by the
introduction of a new concept of alfalfa breeding - semi-hybrid breeding, as one of the ways
to increase the alfalfa biomass yield, and feed producers can expect the release of new, high-
yielding and high-quality cultivars in the coming years. Modern methods of biotechnology -
molecular markers, which are significant from the aspect of increasing the efficiency of
alfalfa breeding, are also intensively applied in breeding work. In recent years, the Institute
has been more intensively dealing with breeding strategies related to the introduction of the
genetic basis of acid soil resistance/Al tolerance using diploid alfalfa (M. caerulea)
populations, which represents the first attempt to increase alfalfa tolerance to acid soils. In
alfalfa breeding, research is directed towards the creation of cultivars with high production of
biomass, as well as the study of the durability of the varieties. Testing the quality, especially
the nutritional value and digestibility of forage, as well as increasing the level of resistance to
economically significant diseases and alfalfa lodging, are one of the directions of work on
alfalfa breeding at the Institute.

populations, which represents the first attempt to increase alfalfa tolerance to acid soils. In
alfalfa breeding, research is directed towards the creation of cultivars with high production of
biomass, as well as the study of the durability of the varieties. Testing the quality, especially
the nutritional value and digestibility of forage, as well as increasing the level of resistance to
economically significant diseases and alfalfa lodging, are one of the directions of work on
alfalfa breeding at the Institute.",

AB - Considering the existence of genotype × environment interaction, grain yield performance testing of newly created maize genotypes is performed in several locations and/or years in trials with smaller experimental plots (MESPT). This should ensure the selection of the best genotypes, which is of critical importance for the breeding process. Certainly, the level of grain yield and yield stability are the most important features when it comes to breeding corn within the framework of the appropriate maturation groups. It is important to choose the locations where the experiments will be carried out, with the aim of covering the range of soil and climatic conditions of the target cultivation area. As it is about conducting experiments where neither soil nor climatic factors can be controlled, and due to a number of other circumstances, the data we get in these experiments are not always perfect.

AB - Seed as a highly-valuable resource is preserved in collections for many years. Although the seed is kept under optimal conditions, monitoring of germination and the presence of fungi during seed preservation is of great importance. Therefore, the aim of this paper was to examine the seed health status and germination of forty-three onion accessions kept in the timespan of 15 years in the Institute of Field and Vegetable Crops collection. Germination of seed samples varied from 7-93%. The presence of fungi in the collection was determined on thirty-three tested samples. Fungi from the genera Acremonium, Alternaria, Aspergillus, Cladosporium, Epicoccum, Fusarium and Penicillium were developed. The following Fusarium species identified on the seeds were F. proliferatum, F. graminearum, F. sporotrichoides, F. solani, F. pseudograminearum and F. equiseti. Based on factor analysis, Fusarium and Penicillium were the species which affected germination, while the occurrence of Alternaria species on onion seed is connected to the year of harvest.

Although the seed is kept under optimal conditions, monitoring of germination and the presence of fungi during seed preservation is of great importance. Therefore the aim of this paper is to examine the seed health status and germination of 43 onion accessions kept in the timespan for 15 years in the Institute of Field and Vegetable Crops collection. Germination of seed samples varied from 7-93%. The presence of fungi in the collection was determined on 33 tested samples. Fungi from the genera Acremonium, Alternaria, Aspergillus, Cladosporium, Epicoccum, Fusarium and Penicillium were developed. The following Fusarium species identified on the seeds were F. proliferatum, F. graminearum, F. sporotrichioides, F. solani, F. pseudograminearum and F. equiseti. Based on factor analysis, Fusarium and Penicillium affected germination, while the occurrence of Alternaria species on onion seed is connected to the year of harvest.

AB - Chickpea (Cicer arietinum L.) is one of the most commonly consumed legume crops worldwide, cultivated in more than 55 countries (FAO 2017). However, in Serbia it is a novel crop grown on approximately 120 acres, but the area under this crop slightly increases each year. Fusarium wilt caused by F. oxysporum f. sp. ciceris is one of the most economically important diseases in most chickpea-growing areas, but there has been no formal report of the Fusarium wilt of chickpea in Serbia. In June 2018, the first symptoms of Fusarium wilt were registered at Rimski Sancevi (Vojvodina Province), Serbia (N 45'19.3119, E 19'49.9339), as wilted chickpea plants grouped in patches on approximately 5% of plants in a 3-acre area. Symptoms of yellowing and necrosis of foliage appeared as late wilt in the podding stage. Roots of affected plants showed no external discoloration, but a cross-section showed dark-brown discoloration of xylem tissue. To isolate the causal agent, symptomatic plants were collected and 10 cuttings of root symptomatic tissue were surface disinfected with 2% sodium hypochlorite solution for 5 min, rinsed three times in sterile distilled water, air dried on sterilized filter paper, and plated on potato dextrose agar (PDA) and water agar (WA) amended with streptomycin sulfate. After 7 days incubation at 25C in the dark, isolates were preliminarily identified according to their morphological characters described by Leslie and Summerell (2006).

AB - Monitoring the health status of narrow-leaved ash tree seedlings (forest office Morović, locality Vinična) in the early spring of 2015, after the catastrophic floods in May 2014, revealed presence of large dark necrotic areas on 1–2 year old sprouts. The isolation of the fungal pathogen was done by standard phytopathological protocols. Three representative isolates (K41, K42 and K78) were preliminary detected and purified by a single-spore technique for further morphological, molecular analyses and pathogenicity testing. Morphological characteristics classified the isolates as Fusarium tricinctum. Tested isolates on narrow leaved ash sprouts caused reddish brown elongated necrotic lesions averaged 20.1 mm. Two marker genes, translation elongation factor 1-alpha (TEF1-α) and internal transcribed spacer (ITS1), were used in this study. Using the Basic Local Alignment Search Tool (BLAST) searching engine, nucleotide sequences were compared to all related sequences. Alignement score resulted in 98.9% identities with F. tricinctum for isolate K78, while isolates K41 and K42 showed 94.1% and 94.3% identities with F. tricinctum complex respectively. To the best of our knowledge, this is the first report of F. tricinctum pathogen infection on flood stressed narrow-leaved ash trees in Serbia.

AB - Agriculture is one of the “victims” of climate change and one of the most severely affected sectors. It has become evident that as the climate changes, crop production strategies must change as well, including primarily adaptations through breeding and crop management. In the future, it is expected that integrative approaches that combine -omics technologies by using bioinformatic tools will facilitate the identification of target genes and markers for complex traits and facilitate crop adaptation to the changing environment. Within its activities, Twinning project CROPINNO will implement at and validate different phenotyping and multi-omics tools in breeding for improved stress resilience. Sunflower is chosen as a model crop for validation of different tools and approaches since it is considered as potential model crop for adaptation to a changing environment. Activities within CROPINNO are aimed at: i) pre-screening sunflower genotypes from IFVCNS collection using for biotic and abiotic stress resilience using different phenotyping methods; ii) study of effects of drought on sunflower plants at chromatin and transcriptional level; iii) performing whole genome SNP analysis in order to develop SNP-based markers for drought stress resilience; iv) performing integrated data analysis and comparative bioinformatics for drought responses in order to unveil possible direct correlations between stress-induced genes transcriptional variation and histone modification levels and design of networks of candidate genes for sunflower drought tolerance. Models, tools and know-how developed on sunflower will be transferred and implemented in the breeding programs of other main field crops at IFVCNS and Western Balkans region.

AB - Sunflower breeding and selection are continual processes designed to increase its yield and improve the resilience to biotic and abiotic stresses. Breeders have been successful in producing a large number of varieties using conventional breeding. However, classical breeding methods cannot always provide solutions for tackling frequent and often unpredictable variations in climatic and market conditions. As response to shortcomings of classical breeding, Institute of Field and Vegetable Crops (IFVCNS) established Center of Excellence for Innovations in Breeding of Climate-Resilient Crops – Climate Crops aimed at introduction and application of new techniques such as (epi)genomic selection and genome editing, along with e fficient phenotyping platforms, for more effi cient trait introduction. Climate Crops aims to step up and stimulate excellence and innovation capacity of IFVCNS in sunflower improvement and enhance its ability to respond and create innovative solutions for the challenges that sunflower production, and agriculture as a whole face - climate changes and need to feed the increasing population.

AB - Chickpea (Cicer arietinum L.) is one of the most commonly consumed legume crops worldwide However, in Serbia, it is a novel crop grown on approximately 120 acres. Fusarium wilt caused by F. oxysporum f. sp. ciceris is one of the most economically important disease in the most chickpea-growing areas, but yet there has been no formal report of the Fusarium wilt of chickpea in Serbia. In June 2018, the first symptoms of Fusarium wilt were registered as wilted chickpea plants grouped in patches on approximately 5% plants in the area of 3 acres. Symptoms as yellowing and necrosis of foliage appeared as late wilt in the podding stage. Roots of affected plants showed no external discoloration, but cross-sections showed darkbrown discoloration of xylem tissue. To isolate the causal agent, symptomatic plants were collected, and ten cuttings of symptomatic root tissue were surface disinfected with 2% sodium hypochlorite solution for 5 min, rinsed three times in sterile distilled water, air-dried on sterilized filter paper and plated on potato dextrose agar (PDA) and water agar (WA) amended with streptomycin sulfate. After seven days of incubation, isolates were preliminarily identified according to their morphological characters and Fusarium Laboratory Manual. For each isolate 100, micro/macroconidia and chlamydospores were measured. Conidia were hyaline; macroconidia sickle-shaped, with blunt ends, 2-4 septa (10.1-17.7 x 3.1-5.8 μm); microconidia ellipsoidal, 0-1 septa (4.9-8.6-2.7-3.5 μm). Chlamydospores were globose (4.3-8.8 μm). Representative isolates (K343, K375 and K378) were purified by a single-spore technique for further analyses (Leslie and Summerell, 2006). Molecular identification of three representative isolates (K343, K375 and K378) was made by sequencing the rRNA internal transcribed spacer (ITS) region and translation elongation factor 1α (TEF1) gen. For all isolates, the ITS and TEF1 genes were amplified and sequenced with primers ITS1/4 EF1-728 and EF1-986. Based on a BLAST search of the NCBI nucleotide database, the ITS sequences (GenBank MK920204.1, MK928423.1 and MK928424.1) had 99.8% identity with F. oxysporum f. sp. ciceris isolate (MK074845.1). The TEF1 (GenBank MN788462.1, MN788463.1 and MN788464.1) had 96.3 -100% identities with F. oxysporum f. sp. ciceris isolate (FJ538245.1). The pathogenicity test was conducted on 7-day old plants according to the drench method. The concentration of inoculation suspensions was adjusted on 1x106, and ten plants per isolate were tested and inoculated with 10 ml of suspension. Control plants were drenched with 10 ml of sterilized distilled water. Nine days after inoculation, first symptoms as leaves wilting and white mycelia presence around stem base occurred on plants inoculated with isolate K378. On day 11, the first symptoms occurred on plants inoculated with isolates K343 and K375 as well. Up to the 13th day after inoculation, all plants were wilted, and the pathogen was successfully reisolated and confirmed as F. oxysporum f.sp. ciceris. To the best of our knowledge, this is the first report of F. oxysporum f.sp. ciceris causing Fusarium wilt on chickpea in Serbia.

AB - The most limiting factors in sunflowers production are weather conditions and plant diseases. Charcoal rot of sunflower is disease caused by soil-borne fungus Macrophomina phaseolina (Tassi) Goid. that gains importance in temperate regions. Control of charcoal rot is complicated due to pathogens long persistence in form of microsclerotia in the soil, often-delayed appearance of visible symptoms in plants and large number of host-plants. Due to that, development and use of resistant or tolerant sunflower genotypes is one of the most effective methods for charcoal rot management. The aim of this study was to determine variability of NS-sunflower inbred lines and hybrids in response to M. phaseolina infection and to detect tolerance within selected genotypes. Totally, 24 NS-sunflower commercial hybrids and 70 inbred lines were tested for tolerance to pathogen M. phaseolina at Rimski Šančevi, Novi Sad, during 2010 and 2011. For artificial inoculation two methods were used - Unwounded Stem Based Inoculation (USBI) which is close to natural infestation process and Toothpick Method which considers infestation by microslerotia through wound in plants.

AB - Plants are constantly exposed to biotic and abiotic stress and are forced to e fficiently balance growth, reproduction and tolerance to different pathogenic organisms. Trichoderma species belong to beneficial fungi that are capable for root colonization which frequently enhances root growth and development, crop productivity, resistance to abiotic stress and nutrients uptake and utilization. Therefore, seed priming with Trichoderma spp. has successfully demonstrated to improve seed germination and seedling establishment for many field crops, such as wheat, sugar beet, maize, soybean, and sunflower. Due to that, objectives of this study was to test growth promoting ability of Trichoderma spp. isolates, which previously indicated good antagonistic activity, on sunflower seedlings. Trichoderma spp. isolates were obtained from soil samples originating from various soil types and localities in Serbia, but mainly from the Vojvodina Province, in 2012.
