AB  - In the present study the effects of individual and combined essential oils (EOs) extracted from onion (Allium cepa L.) bulb and garlic (Allium sativum L.) clove on the growth of Aspergillus versicolor and sterigmatocystin (STC) production were investigated. The EOs obtained by hydrodistillation were analyzed by GC/MS. Twenty one compounds were identified in onion EO. The major components were: dimethyl-trisulfide (16.64%), methyl-propyl-trisulfide (14.21%), dietil-1,2,4-tritiolan (3R, 5S-, 3S, 5S- and 3R, 5R-isomers) (13.71%), methyl-(1-propenyl)-disulfide (13.14%), and methyl-(1-propenyl)-trisulfide (13.02%). The major components of garlic EO were diallyl-trisulfide (33.55%), and diallyl-disulfide (28.05%). The mycelial growth and the STC production were recorded after 7, 14, and 21 d of the A. versicolor growth in Yeast extract sucrose (YES) broth containing different EOs concentrations. Compared to the garlic EO, the onion EO showed a stronger inhibitory effect on the A. versicolor mycelial growth and STC production. After a 21-d incubation of fungi 0.05 and 0.11 mu g/mL of onion EO and 0.11 mu g/mL of garlic EO completely inhibited the A. versicolor mycelial growth and mycotoxins biosynthesis. The combination of EOs of onion (75%) and garlic (25%) had a synergistic effect on growth inhibition of A. versicolor and STC production.

Incidence of Tilletia species in non-processed seed of Triticum aestivum in Vojvodina, Serbia

AB  - With the growing importance of organic production and resurgence of bunt fungi in organic farming conditions, the present study was undertaken to determine incidence and contamination level of Tilletia species in conventionally produced non-processed seed of wheat in Vojvodina. Analysis of 151 samples of basic, certified and commercial seed revealed that 129 samples were contaminated with Tilletia species. The predominant species was T. caries, which was found in 127 samples. However, teliospores of Tilletia species, which have not been registered in Serbia before, were found in five samples. These teliospores had a prominent gelatinous sheath together with conspicuous, deep reticulation and it was assumed that some of them might be of the quarantined T. contraversa, whilst others were identified as T. bromi. Contamination level of basic and certified seed was about one teliospore per 10 seeds, while four out of 16 commercial seed samples were contaminated with more than 900 teliospores per seed which exceeds the allowed contamination level.

Identification of Tilletia species using rep-PCR fingerprinting technique

AB  - Analyzing 167 non-processed seed samples of wheat, it was found that 145 samples (86.8 %) were contaminated with Tilletia species, while 22 (13.2 %) samples were not contaminated. By using rep-PCR fingerprinting technique, it was found that DNA isolates of T. tritici originated from Serbian wheat samples had 80 % similarity with positive control for T. tritici. One isolate shared similarity of 60% with T. tritici, T. controversa and T. laevis. It was supposed that this isolate belongs to T. bromi. Isolate of T. laevis shared a similarity of 70 % with isolates of T. tritici and T. controversa, while T. walkeri was more than 10 % similar with T. tritici, T. controversa and T. laevis. Although T. controversa and T. tritici had high percent of genetic similarity, they were clustered separately. Our results suggest that rep-PCR fingerprinting could be a useful tool for monitoring presence of morphologically similar Tilletia species in wheat production areas.
AB  - Istraživanjem kontaminiranosti uzoraka semena pšenice teleutosporama Tilletia vrsta, utvrđeno je da je od 167 uzoraka nedorađenog semena pšenice, bilo kontaminirano 145 (86,8%), dok su 22 uzorka (13,2%) smatrana nekontaminiranim. Identifikacija Tilletia vrsta izvršena je rep-PCR fingerprinting tehnikom. Izolati prikupljeni na teritoriji Republike Srbije identifikovani su kao T. tritici, s obzirom da su sa pozitivnom kontrolom imali genetičku sličnost veću od 80%. Jedan od izolata koji je vodio poreklo iz opštine Apatin bio je oko 60% genetički sličan sa izolatima T. tritici, T. controversa i T. laevis. Pretpostavljeno je da pripada vrsti T. bromi. Genetička sličnost izolata T. walkeri i vrsta: T. tritici, T. laevis i T. controversa, iznosila je nešto više od 10%. Genetička sličnost T. tritici, T. controversa i T. laevis bila je oko 70 %. I pored visokog procenta genetičke sličnosti između T. controversa i T. tritici, napravljena je razlika među njima, što rep-PCR fingerprinting tehniku čini veoma podesnom za praćenje prisustva morfološki sličnih Tilleria vrsta prilikom kontrole kvaliteta semena pšenice.

Fusarium rot of onion and possible use of bioproduct

AB  - Several species of Fusarium are causal agents of onion rot in field and storage. Most prevalent are F. oxysporum f. sp. cepae and F. solani, and recently F. proliferatum, a toxigenic species. Most frequently isolated fungi in our field experiments were F. solani and F. proliferatum with different pathogenicity. Certain differences in antagonistic activity of Trichoderma asperellum on different isolates of F. proliferatum and F. solani have been found in in vitro study in dual culture, expressed as a slower inhibition of growth of the former, and faster of the latter pathogen. Antagonistic abilities of species from genus Trichoderma (T. asperellum) are important, and have already been exploited in formulated biocontrol products in organic and conventional production, in order to prevent soil borne pathogens inducing fusarium wilt and rot. The importance of preventing onion infection by Fusarium spp., possible mycotoxin producers, has been underlined.
AB  - Fuzariozna trulež luka najčešće je prouzrokovana vrstama Fusarium oxysporum f. sp. cepae i F. solani, a i odskora i toksikogeni F. proliferatum u našim uslovima gajenja i čuvanja luka u skladištima. Utvrđena je najveća učestalost F. proliferatum i F. solani na lukovicama iz polja. Razlike izolata u patogenosti ispoljile su se u različitom uticaju na klijanje lučica, izduživanje klice i intenzitet propadanja. Utvrđena je različita antagonistička aktivnost Trichodrema asperellum na izolate F. proliferatum i F. solani, sporija inhibicija u slučaju prvog i izražena u slučaju drugog patogena u in vitro ogledima. Antagonistička svojstva vrsta iz roda Trichoderma se iskorišćavaju za formulaciju bioloških preparata primenjivih u organskoj i konvencionalnoj proizvodnji, u prevenciji oboljenja luka koja prouzrokuju zemljišni paraziti, pre svega fuzarioznog uvenuća i truleži. Istaknut je značaj bioloških preparata u zaštiti zdravstvene bezbednosti proizvođača i potrošača, s obzirom da su ovi patogeni potencijalni producenti mikotoksina.

AB  - U svetu postoji 514 nacionalnih/međunarodnih kolekcija mikroorganizama u 66 zemalja, koje uključuju i dve kolekcije bakterija azotofiksatora u Srbiji: Collection of Bacteria (853 soja bakterija i jedna vrsta gljive)- Institute of Soil Science Department of soil Microbiology; Belgrade; i Novi Sad Collection of Nitrogen Fixing Bacteria (44 soja) - Institute of Field Vegetable Crops, Novi Sad (World Data Center on Microorganisms). Neki mikroorganizmi sa našeg podneblja deponovani su u međunarodnim kolekcijama, ali o tome naučna javnost nije upoznata. Pored navedenih, u Srbiji postoje veoma bogate, posebno sojevima gljiva, nezvanične kolekcije koje se čuvaju u pojedinim institucijama (npr. preko 3600 izolata od 84 determinisane vrste gljiva u Institutu za kukuruz "Zemun Polje"; 65 sojeva gljiva poreklom iz pšenice i brojne radne kolekcije s drugih biljnih vrsta u Naučnom institutu za ratarstvo i povrtarstvo, Novi Sad; 262 soja gljiva u Institutu za stočarstvo, Zemun; 242 determinisana izolata fitopatogenih bakterija na Poljoprivrednom fakultetu u Beogradu). Zbog nedostatka potrebne opreme i primene nestandardizovanih postupaka konzerviranja, originalnost i vitalnost ovih mikroorganizama nije moguće očuvati u dužem vremenskom periodu.

Besides stated collections in Serbia, there are very rich unofficial collections, especially in fungal strains, stored in certain institutions ( e.g. over 3,600 isolates out of 84 determined fungal species are stored at the Maize Research Institute, Zemun Polje; 65 fungal strains originating from wheat and numerous working collections from other plant species arc stored at the Institute of Field and Vegetable Crops, Novi Sad; 262 fungal strains are stored at the Institute for Animal Husbandry, Zemun; there are 242 determined isolates of phytopathogenic bacteria at the Faculty of Agriculture, Belgrade). Due to a lack of the necessary equipment and the application of non-standardised procedures in conservation, it is not possible to maintain the originality and vitality of these microorganisms in a longer period of time.

AB  - Aktuelna istraživanja u zaštiti bilja, najlakše je sagledati kroz tekuće projekte, koje finansira Ministarstvo za nauku i životnu sredinu i Ministarstvo poljoprivrede, šumarstva i vodoprivrede-Uprava za zastitu bilja i Pokrajinski sekretarijat za nauku i tehnološki razvoj. Međutim, u finansiranju nekih projekata učestvuju gradske i opštinske uprave. Svega dva projekta možemo okarakterisati kao međunarodna, što govori o našoj zatvorenosti i izolovanosti unutar institucija u kojima rade specijalisti zaštite bilja bez obzira na titule i zvanja. Osiromašena privreda i nezainteresovanost za ulaganje samo su jedna od posledica opšteg trenda.
